Notch signaling controls chondrocyte hypertrophy via indirect regulation of Sox9

RBPjk-dependent Notch signaling regulates both the onset of chondrocyte hypertrophy and the progression to terminal chondrocyte maturation during endochondral ossification. It has been suggested that Notch signaling can regulate Sox9 transcription, although how this occurs at the molecular level in chondrocytes and whether this transcriptional regulation mediates Notch control of chondrocyte hypertrophy and cartilage development is unknown or controversial. Here we have provided conclusive genetic evidence linking RBPjk-dependent Notch signaling to the regulation of Sox9 expression and chondrocyte hypertrophy by examining tissuespecific Rbpjk mutant（Prx1Cre;Rbpjkf/f）, Rbpjk mutant/Sox9 haploinsufficient（Prx1Cre;Rbpjkf/f;Sox9f/1）,and control embryos for alterations in SOX9 expression and chondrocyte hypertrophy during cartilage development. These studies demonstrate that Notch signaling regulates the onset of chondrocyte maturation in a SOX9-dependent manner, while Notch-mediated regulation of terminal chondrocyte maturation likely functions independently of SOX9. Furthermore, our in vitro molecular analyses of the Sox9 promoter and Notch-mediated regulation of Sox9 gene expression in chondrogenic cells identified the ability of Notch to induce Sox9 expression directly in the acute setting, but suppresses Sox9 transcription with prolonged Notch signaling that requires protein synthesis of secondary effectors.

Miombo woodlands near human settlements are under significant pressures from human activities,with negative consequences on their structure and *** studies are limited,we assessed the structure and species composition of a portion of miombo woodland along an increasing disturbance gradient from a national park,through a buffer zone to communal lands in northeast *** concentric plots of 2 and 11 m radii were established in each area to record woody species composition,diameter,height,basal area,density and volume as well as evidence of *** of site,growth stage and their interaction on vegetation diversity and structural parameters were tested using a general linear model(GLM).Principal component analysis(PCA) tested the association between species and site and ANOVA the differences in the level of disturbance across *** diversity did not differ between sites except for evenness,which increased with *** and richness were greater in seedlings and saplings than mature trees across sites,*** and mature tree diameters differed significantly between *** and density of mature trees declined with increasing disturbance while seedling densities peaked at intermediate disturbance levels(buffer zone).Tree harvesting,was more evident in the buffer zone and in the communal area relative to the national *** contrast,fire frequency was greater in the national park and in the buffer zone relative to the communal *** results of this study identify a true miombo woodland dominated by Brachystegia boehmii with a stable population,as illustrated by an inverse-j shape in diameter class distribution on all sites,and that these woodlands are generally resilient to disturbances,maintaining similar species composition and structure at various levels of ***,continued monitoring of disturbance levels and miombo woodland response is recommended to ensure sustainable utilisation of

Performance on the Functional Movement Screen in older active adults

Background:The Functional Movement Screen(FMS^(TM)) has become increasingly popular for identifying functional limitations in basic functional *** exploratory and descriptive study was undertaken to confirm feasibility of performing the FMS^(TM) in older active adults,assess prevalence of asymmetries and to evaluate the relationship between functional movement ability,age,physical activity levels and body mass index(BMI).Methods:This is an observational study;97 men(n = 53) and women(n = 44) between the ages of 52 and 83 *** was computed and self-reported physical activity levels were *** were grouped by age(5-year intervals),BMI(normal,over-weight,and obese)and *** participant's performance on the FMS^(TM) was digitally recorded for later ***:The youngest age group(50–54 years) scored highest in all seven tests and the oldest age group(75+) scored lowest in most of the tests compared to all other age *** subjects in the 'normal weight' group performed no different than those who were in the 'overweight' group;both groups performed better than the 'obese' *** the 97 participants 54 had at least one *** pairwise correlations between the total FMS^(TM) score and age(r =-0.531),BMI(r =-0.270),and the measure of activity level(r = 0.287) were significant(p juries in the older population.

Understanding how individuals manage costs during the migration period is challenging because individuals are difficult to follow between sites; the advent of hybrid Global Positioning Systemacceleration （ACC） tracking devices enables researchers to link spatial and temporal attributes of avian migration with behavior for the first time ever. We fitted these devices on male Greenland white-fronted geese Anser albifrons flavirostris wintering at 2 sites （Loch Ken, Scotland and Wexford, Ireland） to understand whether birds migrating further during spring fed more on winter- ing and staging areas in advance of migration episodes. Although Irish birds flew significantly further （ca. 300 kin） than Scottish birds during spring, their cumulative hours of migratory flight, flight speed during migration, and overall dynamic body ACC （i.e., a proxy for energy expenditure） were not significantly different. Further, Irish birds did not feed significantly more or expend significantly more energy in advance of migration episodes. These results suggest broad individual plasticity in this species, although Scottish birds arriving on breeding areas in Greenland with greater energy stores （because they migrated less） may be better prepared for food scarcity, which might increase their reproductive success.
