Head and neck squamous cell carcinoma(HNSCC)is one of the most frequent cancers *** main risk factors are consumption of tobacco products and alcohol,as well as infection with human papilloma *** therapeutic options comprise surgery,radiation,chemotherapy,targeted therapy through epidermal growth factor receptor inhibition,and immunotherapy,but outcome has remained unsatisfactory due to recurrence rates of~50%and the frequent occurrence of second *** availability of the human genome sequence at the beginning of the millennium heralded the omics era,in which rapid technological progress has advanced our knowledge of the molecular biology of malignant diseases,including HNSCC,at an unprecedented ***,microarray-based methods,followed by approaches based on next-generation sequencing,were applied to study the genetics,epigenetics,and gene expression patterns of bulk *** recently,the advent of single-cell RNA sequencing(scRNAseq)and spatial transcriptomics methods has facilitated the investigation of the heterogeneity between and within different cell populations in the tumor microenvironment(e.g.,cancer cells,fibroblasts,immune cells,endothelial cells),led to the discovery of novel cell types,and advanced the discovery of cell-cell communication within *** review provides an overview of scRNAseq,spatial transcriptomics,and the associated bioinformatics methods,and summarizes how their application has promoted our understanding of the emergence,composition,progression,and therapy responsiveness of,and intercellular signaling within,HNSCC.

Paleomagnetic age and palaeobiological significance of hominoid fossil strata of Yuanmou Basin in Yunnan

The two sections containing hominoid fossils in the Yuanmou Basin, Yunnan Prov-ince have been studied by means of magnetostratigraphy. The results are as follows. The Xiaohe section records over 8 polarity zones corresponding to Cande-Kent95 time scale, including C3Br.1n, C3Br.1r, C3Br.2n, C3Br.2r, C4n.1n, C4n.1r, C4n.2n and C4n.2r, attaching to Late Mio-cene. This section comprises two layers of hominoid fossils: the lower layer (the 1st layer in fig. 4) which records the lower portion of C4n.2r with paleomagnetic age of 8.20—8.10 Ma and the upper layer (the 16th layer in fig. 4) which records C3Br.1r with paleomagnetic age of 7.20—7.15 Ma. So the age of hominoid fossils in the Xiaohe section is about 8.20—7.15 Ma. The Leilao section records over 11 polarity zones corresponding to Cande-Kent95 time scale, including C3Br, C3Br.1n, C3Br.1r, C3Br.2n, C3Br.2r, C4n.1n, C4n.1r, C4n.2n, C4n.2r, C4n.3n and C4n.3r, attaching to Late Miocene. This section also consists of two layers of hominoid fossils: the lower layer (the 3rd layer in fig. 3) which records C4n.2r with the age of 8.20—8.10 Ma and the upper layer(the 19th layer in fig. 3) which records the middle portion of C3Br with the age of 7.15—7.1 Ma. So the age of hominoid fossils in the Leilao section is about 8.20—7.10 Ma. Sedimentary environments reflected in both sections are extremely similar, and the ages of hominoid fossils are of correspondency. The age of the lower and upper fossil layers is 8.20—8.10 Ma and 7.20—7.10 Ma respectively, which probably imply that hominoids living in Xiaohe and Leilao basins have uniform activity areas. There were hominoids activities all along during 8.00—7.00 MaBP in this field.

The genetic information coded in DNA leads to trait innovation via a gene regulatory network(GRN)in ***,we developed a conserved non-coding element interpretation method to integrate multi-omics data into gene regulatory network(CNEReg)to investigate the ruminant multi-chambered stomach *** generated paired expression and chromatin accessibility data during rumen and esophagus development in sheep,and revealed 1601 active ruminantspecific conserved non-coding elements(active-RSCNEs).To interpret the function of these activeRSCNEs,we defined toolkit transcription factors(TTFs)and modeled their regulation on rumenspecific genes via batteries of active-RSCNEs during *** developmental GRN revealed 18 TTFs and 313 active-RSCNEs regulating 7 rumen functional ***,6 TTFs(OTX1,SOX21,HOXC8,SOX2,TP63,and PPARG),as well as 16 active-RSCNEs,functionally distinguished the rumen from the *** study provides a systematic approach to understanding how gene regulation evolves and shapes complex traits by putting evo-devo concepts into practice with developmental multi-omics data.

BACKGROUND Transjugular intrahepatic portosystemic shunt(TIPS) placement is an effective intervention for recurrent tense ascites. Some studies show an increased risk of acute on chronic liver failure(ACLF) associated with TIPS placement. It is not clear whether ACLF in this context is a consequence of TIPS or of the pre-existing liver *** To better understand the risks of TIPS in this challenging setting and to compare them with those of conservative *** Two hundred and fourteen patients undergoing their first TIPS placement for recurrent tense ascites at our tertiary-care center between 2007 and 2017 were identified(TIPS group). Three hundred and ninety-eight patients of the same time interval with liver cirrhosis and recurrent tense ascites not undergoing TIPS placement(No TIPS group) were analyzed as a control group. TIPS indication,diagnosis of recurrent ascites, further diagnoses and clinical findings were obtained from a database search and patient records. The in-hospital mortality and ACLF incidence of both groups were compared using 1:1 propensity score matching and multivariate logistic *** After propensity score matching, the TIPS and No TIPS groups were comparable in terms of laboratory values and ACLF incidence at hospital admission. There was no detectable difference in mortality(TIPS: 11/214, No TIPS 13/214). During the hospital stay, ACLF occurred more frequently in the TIPS group than in the No TIPS group(TIPS: 70/214, No TIPS: 57/214, P = 0.04). This effect was confined to patients with severely impaired liver function at hospital admission as indicated by a significant interaction term of Child score and TIPS placement in multivariate logistic regression. The TIPS group had a lower ACLF incidence at Child scores < 8 points and a higher ACLF incidence at ≥ 11 points. No significant difference was found between groups in patients with Child scores of 8 to 10 *** TIPS placement for recurrent tense as
