Three-dimensional visualization of planta clathrin-coated vesicles at ultrastructural resolution

biological systems are the sum of their dynamic three-dimensional(3D)***,it is critical to study biological structures in 3D and at high resolution to gain insights into their physiological *** microscopy of metal replicas of unroofed cells and isolated organelles has been a key technique to visualize intracellular structures at nanometer ***,many of these methods require specialized equipment and personnel to complete ***,we present novel accessible methods to analyze biological structures in unroofed cells and biochemically isolated organelles in 3D and at nanometer resolution,focusing on Arabidopsis clathrin-coated vesicles(CCVs).While CCVs are essential trafficking organelles,their detailed structural information is lacking due to their poor preservation when observed via classical electron microscopy protocols ***,we establish a method to visualize CCVs in unroofed cells using scanning transmission electron microscopy tomography,providing sufficient resolution to define the clathrin coat ***,the samples are prepared directly on electron microscopy grids,removing the requirement to use extremely corrosive acids,thereby enabling the use of this method in any electron microscopy ***,we demonstrate that this standardized sample preparation allows the direct comparison of isolated CCV samples with those visualized in ***,to facilitate the high-throughput and robust screening of metal replicated samples,we provide a deep learning analysis method to screen the“pseudo 3D”morphologies of CCVs imaged with 2D ***,our work establishes accessible ways to examine the 3D structure of biological samples and provide novel insights into the structure of plant CCVs.

Degradation of organic pollutants by a Co_3O_4 -graphite composite electrode in an electro-Fenton-like system

A novel composite electrode was constructed by pressing together Co3O4 and graphite and it was used as the cathode in an electro-Fenton-like (EFL) system. The poor electron transport characteristic of Co3O4 was overcome by incorporating graphite. In situ electro-catalytic generation of hydroxyl radicals (·OH) occurred at high current efficiencies from pH 2-10, extending the traditional Fenton reaction pH range. Cyclic voltammetry and AC impedance spectrometry were used to characterize the composite electrode. The ability of the EFL system to degrade organic compounds was investigated using sulforhodamine b (SRb) and 2,4-dichlorophenol (2,4-DCP) as probes. Decoloration of SRb (1.0×10-5 mol/L) was complete (100%) in 150 min and SRb was effectively degraded from pH 2-10. The decomposition of SRb was studied using Fourier transform infrared spectroscopy (FT-IR) and total organic carbon (TOC) analysis and results indicated that the final degradation products were carbon dioxide, carboxylic acids and amines. The EFL system also decomposed 2,4-DCP and the degradation was 98.6% in 240 min. Electro-catalytic degradation of SRb occurs by a ·OH mechanism. After 5 times reused, the degradation rate of SRb did not significantly slow down. The electrode shows excellent potential for use in advanced oxidation processes (AOPs) used to treat persistent organic pollutants (POPs) in wastewater.

SEA FLUX OF TRACE METALS IN SAANICH INLET

The sea fluxes of trace metals, POC, and settled material were studied in anoxic sea water, Saanich Inlet, b.C., Canada with sediment traps. This paper discusses the change of mass fluxes of sediment, trace metals and POC for various seasons and depths. The annual mean of settled material is 1.56 g.m-2 .a-1, 84.6 mg. m-2. a-1 for POC, 60.0 mg.m-2. a-1 for Cu, 16.5 mg.m-2.a-1 for Pb, 189 mg.m-2. a-1 for Zn, 2.20mg.m-2.a-1 for Cd,699 mg.m-2.a-1 for Fe,38.8 mg.m-2.-1 for Co, and 84.6 mg.m-2.a-1 for *** relations between the average fluxes of trace metals and POC, the fluxes of trace metals and settled matter, and the sea fluxes of trace metals and Fe are in linear progression. The resident times of elements as calculated from sea flux, are 1.1 a. for Cu, 0.014 a. for Pb, 0.50 a. for Zn, 3.8 a. for Cd, 0.16 a. for Fe, 0.39 a. for Co,and 1.14 a. for *** order of resident times is as follows: Pb-Fe-Co-Zn-Cu-Ni-Cd. Marked elevation of b-type natriuretic peptide in patients with heart failure and preserved ejection fraction

Marked elevations of b-type natriuretic peptide （bNP） are not generally seen in patients with heart failure and preserved ejection fraction （HFpEF）. The objective of this study was to examine the clinical and laboratory characteristics of a large cohort of patients with HFpEF and markedly elevated bNP. A retrospective examination of 421 inpatients at a university hospital admitted with a diagnosis of HFpEF was performed. Clinical and echocardiographic data in 4 groups of patients with levels of bNP ≤ 100 pg/mL, 100-400 pg/mL, 400-1,000 pg/mL and 〉 1,000 pg/mL were compared. Patients with HFpEF and bNP 〉 1,000 pg/mL （28% of the population） were characterized by impaired renal function and greater use of anti-hypertensive medications. A subset of these patients with bNP 〉 1,000 pg/mL had normal renal function （21%） and were significantly older, more frequently female, and tended to have lower ejection fractions. Conversely, patients with HFpEF and bNP ≤100 pg/mL were younger and had preserved renal function. bNP was inversely related to the likelihood of subsequent admission for heart failure, but not to myocardial infarction or death. In conclusion： bNP 〉 1,000 pg/mL is seen in almost 1/3 of patients hospitalized with HFpEF. This elevation of bNP often reflects impaired renal function, but can also be seen in patients with preserved renal function but relatively impaired systolic function.

Oleic acid Stimulates the Formation of Adipocyte-Like Cells from bovine Satellite Cells via <i>G-Protein Coupled Receptor 43</i>and <i>CCAAT/Enhancer-binding Protein beta</i>

Numerous physiological and pathological processes are controlled by free fatty acids, which act as signaling molecules in mammals. We hypothesized that oleic acid (Ole) might stimulate the formation of satellite-cell-derived intramuscular adipose tissue. The role of Ole as a ligand of G-protein-coupled receptor 43 (GPR43) was previously identified. Thus, the objective of the current study was to determine the effect of Ole on GPR43 and factors related to the adipogenic differentiation of bovine satellite cells (bSC). Treatments of 100 μM and 500 μM Ole tended to induce greater (P CCAAT/enhancer-binding protein β (C/EbPβ) compared to all other doses. The mRNA abundance of peroxisome proliferator-activated receptor γ (PPARγ) was not altered (P > 0.10) by treatment. The addition of 100 μM and 500 μM of Ole upregulated (P GPR43 mRNA expression. Protein level of GPR43 was increased (P by 100 μM of Ole treatments. Addition of Ole to bSC cultures induced transformation of myogenic cells into adipocyte-like cells that formed cytoplasmic lipid droplets. Increased expression of C/EbPβ in response to Ole might suppress myogenic differentiation. After the treatment of cells with Ole, increased expression of GPR43 could lead to phosphorylation of 5’ AMP-activated protein kinase α (AMPKα). Altogether, the results indicated that increased Ole might stimulate adipose tissue accumulation within the skeletal muscle of cattle by promoting adipogenic differentiation and activation of GPR43 in satellite cells.

Interaction of ergotamine with liver Cytochrome P450 3A in rats

This study was conducted to investigate the effect of the ergot alkaloid, ergotamine (ET), on the induction of CYP3A and the interaction in vivo and in vitro with ET. Sprague-Dawley rats were treated intraperitoneally for 4 days as follows: control (injecting with 0.5 ml of only corn oil);dexamethasone treatment (injecting with 100mg/kg of dexamethasone in corn oil);and ergotamine treatment (injecting with 100mg/kg of ergotamine in corn oil). Liver tissues were collected from each group (n = 5, total of 30 rats) and liver microsomes were prepared. Cytochrome CYP3A activity was evaluated using ET and its isomer as substrates in medium containing liver microsomes and NADPH at 37℃ for 30 min. HPLC was used to measure the disappearance of the substrate and the appearance of the metabolites. Liver microsomes from rats pretreated with dexamethasone were five times more (P 0.05) in activity of CYP3A when compared to the control group (5.2 vs. 7.0 nM ET/min/mg protein;SE = 4.83) or ET isomer (1.5 vs. 4.7 nM ET isomer/ min/mg protein;SE = 1.70). When ketoconazole was used as specific inhibitor of CYP3A, ergotamine metabolisms were inhibited in a dose dependent fashion reaching a maximum at an inhibitor to substrate ratio of greater than one and LD50 at 0.5 nM of ketoconazole/mg protein. The data presented in this study suggest that although the ergot alkaloids ergotamine and its isomer are ideal substrates for the isozyme CYP3A, these compounds have no effect on the induction of CYP3A after 4 days of treatment.

Liver cytochrome P450 system as affected by endophyte-infected tall fescue seed extracts and ergot alkaloids

Endophyte infected tall fescue (E+) is the base diet for nearly all beef cattle in the southern USA. It has been linked to a variety of toxicological conditions due to the presence of large numbers of ergot alkaloids. This study was designed to investigate the effects of E+ seed extract and selected ergot alkaloids on the detoxification pathway by cytochrome P450 (CYP3A4) enzyme system. Tests were performed using the P450- Glo CYP3A4 enzyme activity kit (Promega, WI), according to the manufacturer’s manual. Luminescence was measured using a single tube TD20/20 luminometer. Endophyte infected tall fescue seed was extracted with 50/50 methanol/25 mM ammonium carbonate, cleaned and concentrated on Strata-X reversed phase column (Phenomenex). The extracts were evaluated on an HPLC, and then tested using a serial dilution method. Commercially available ergonovine (EN), ergocorine (ER), bromocryptine (bC) and ergocryptine (EC) were tested individually using 0 to 44 nM concentrations. Seed extract of E+ produced a significant (P P being most potent, followed by ER, bC, and then EN (70%, 40%, 30% and 10% at 44 nM concentration). The similarity of the inhibition curves of seed extract to that of the commercially available ergot alkaloids suggests a related mode of action and that the use of such ergot alkaloids and CYP3A4 assay is a good model to study the toxicity of tall fescue. Furthermore, it provides the foundation to identify the individual toxic components of purified endophyte infected tall fescue extract.

Rare presentation of post-transplant lymphoproliferative disorder isolated to gastroesophageal junction

Post transplant lymphoproliferative disorder (PTLD) represents a life threatening disorder occurring after transplantation, ranging from a polyclonal mononucleo- sis like illness to a monomorphic high grade neoplasm with cytologic and histopathologic evidence indica- tive of transformation to lymphoma. PTLD of diffuse large b-cell lymphoma (DLbCL) subtype, isolated to the esophagus is a rare diagnosis. We describe the first case of an immunocompromised adult patient diagnosed with DLbCL-PTLD limited to his esophagus without an associated mass or locoregional lymphade-nopathy on imaging since the institution of the revised Cheson criteria, which includes positron emission tomography-computed tomography as the standard staging modality. Even more unique to our case was the suggestion of underlying cytomegalovirus (CMV) gastritis leading to a hypothesis about a less well understood relationship between CMV and Epstein barr virus (EbV). In the post transplant setting, im- munocompromised state, or EbV positive state, upper gastrointestinal symptoms should prompt investigation with an upper endoscopy (EGD). Additionally, specific to our case, the fact that the patients' presentation was suspicious for CMV gastritis raises the possibility that the CMV infection predated his PTLD increasing his risk of acquiring PTLD. This reemphasizes the im- portance and diagnostic utility of early screening with EGD in patients after transplantation.
