Rebuilding insight into the pathophysiology of Alzheimer’s disease through new blood-brain barrier models

The blood-brain barrier is a unique function of the microvasculature in the brain parenchyma that maintains homeostasis in the central nervous ***-brain barrier breakdown is a common pathology in various neurological diseases,such as Alzheimer’s disease,stroke,multiple sclerosis,and Parkinson’s ***,it has been considered a consequence of neuroinflammation or neurodegeneration,but recent advanced imaging techniques and detailed studies in animal models show that blood-brain barrier breakdown occurs early in the disease process and may precede neuronal ***,the blood-brain barrier is attractive as a potential therapeutic target for neurological diseases that lack effective *** elucidate the molecular mechanism underlying blood-brain barrier breakdown and translate them into therapeutic strategies for neurological diseases,there is a growing demand for experimental models of human origin that allow for functional ***,several human induced pluripotent stem cell-derived blood-brain barrier models have been established and various in vitro blood-brain barrier models using microdevices have been *** in the Alzheimer’s disease field,the human evidence for blood-brain barrier dysfunction has been demonstrated and human induced pluripotent stem cell-derived blood-brain barrier models have suggested the putative molecular mechanisms of pathological blood-brain *** this review,we summarize recent evidence of blood-brain barrier dysfunction in Alzheimer’s disease from pathological analyses,imaging studies,animal models,and stem cell ***,we discuss the potential future directions for blood-brain barrier research.

As open substructures of fullerenes,aromaticπ-bowls are promising candidates as new organic semiconductors,as well as attractive hosts for *** demonstrate herein the synthesis and characterization of a novel C_(2v)symmetricπ-bowl,pyracyleno[6,5,4,3,2,1-pqrstuv]pentaphene(3).Bowl 3 was equipped with two distinctive reactive sites,allowing for bromination and cross-coupling reactions to readily yield functionalized bowls with two 2,4,6-trimethylphenyl(5)and triethylsilyl(TES)-ethynyl(6)substituents,***-temperature 1H NMR analysis and density functional theory(DFT)calculations indicated bowl-to-bowl inversions of 3,5,and 6 at room *** alternating the substituents,the crystal structures of the threeπ-bowls 3,5,and 6 could be controlled from 1D linear to 1D slipped to 2D herringbone packing motifs,providing insight into the packing behavior ofπ-bowls.1H NMR titration study indicated that the TES-ethynyl substituent enhanced the ability ofπ-bowl to bind C_(70)with an association constant of 2485 M−*** C_(70)molecules withπ-bowls 3 and 6 formed 1:1 complexes,in which C_(70)molecules aggregated into zig-zag and 1D linear arrays,*** hole mobility of 2.3 cm^(2)V^(−1)^s(−1)and electron mobility of 0.16 cm^(2)V^(−1)^s(−1)ofπ-bowl 3 and its complex with C_(70)were demonstrated,respectively,which proved a great value for the development of aromaticπ-bowl semiconductors with tunable properties for organic electronic devices.

Operationally Simple Enantioselective Silane Reduction of ketones by the [Ir(OMe)(cod)]₂/Azolium Catalytic System

An operationally simple protocol was designed for the enantioselective silane reduction (ESR) of ketones using air- and moisture-stable [Ir(OMe)(cod)]2 (cod = 1,5-cyclooctadiene) (3) as a metal catalyst precursor. This reaction was driven by chiral hydroxyamide-functionalized azolium salt 2. The catalytic ESR reaction could be performed under benchtop conditions at room temperature. Treatment of 2 with 3 in THF yielded the monodentate IrCl(NHC)(cod) (NHC = N-heterocyclic carbene) complex 4 in 93% yield, herein the anionic methoxy ligand of 3 serves as an internal base that deprotonates the azolium ring of 2. The well-defined Ir complex 4 catalyzed the ESR reaction of propiophenone (6) with (EtO)2MeSiH using the pre-mixing reaction procedure. Based on this success, the catalytic ESR reaction was designed and implemented using an in situ-generated NHC/Ir catalyst derived from 2 and 3. Thus, a wide variety of aryl ketones could be reduced to the corresponding optically active alcohols in moderate to excellent stereoselectivities at room temperature without temperature control. Since the high catalytic activity of 3 was observed, we next evaluated several other transition metal catalyst precursors for the catalytic ESR reaction under the influence of 2. This evaluation revealed that Ir(acac)(cod) (acac = acetylacetonate) (28) and [IrCl(cod)]2 (5) can be successfully used as metal catalyst precursors in the ESR reaction.

Electroencephalogram Signal Correlations between Default Mode Network and Attentional Functioning

Attentional issues may affect acquiring new information, task performance, and learning. Cortical network activities change during different functional brain states, including the default mode network (DMN) and attention network. We investigated the neural mechanisms underlying attentional functions and correlations between DMN connectivity and attentional function using the Trail-Making Test (TMT)-A and -B. Electroencephalography recordings were performed by placing 19 scalp electrodes per the 10 - 20 system. The mean power level was calculated for each rest and task condition. Non-parametric Spearman’s rank correlation was used to examine the correlation in power levels between the rest and TMT conditions. The most significant correlations during TMT-A were observed in the high gamma wave, followed by theta and beta waves, indicating that most correlations were in the parietal lobe, followed by the frontal, central, and temporal lobes. The most significant correlations during TMT-B were observed in the beta wave, followed by the high and low gamma waves, indicating that most correlations were in the temporal lobe, followed by the parietal, frontal, and central lobes. Frontoparietal beta and gamma waves in the DMN may represent attentional functions.

Enhancement of Visual Attention by Color Revealed Using Electroencephalography

Attention constitutes a fundamental psychological feature guiding our mental effort toward specific objects, concurrent with processes such as memory, reasoning, and imagination. Visual attention, crucial for selecting surrounding information, often decreases in older adults and patients with cerebrovascular disorders. Effective methods to enhance attention are scarce. Here, we investigated whether color information influences visual attention and brain activity during task performance, utilizing EEG. We examined 13 healthy young adults (seven women and six men;mean age: 21.2 ± 0.58 years) using 19-electrode electroencephalograms to assess the impact of color information on visual attention. The Clinical Assessment for Attention cancellation test was conducted under the black, red, and blue color conditions. Wilcoxon’s signed-rank test was used to assess differences in task performance (task time and error) between conditions. Spearman’s rank correlation was utilized to examine the correlation in power levels between task performance and color conditions. Significant variations in total task errors were observed among color conditions. The black condition exhibited the highest error frequency (0.7 ± 0.9 times), followed by the red condition (0.5 ± 0.8 times), with the lowest error frequency occurring in the blue (0.2 ± 0.4 times) condition (black vs. red: P = 0.03;black vs. blue: P = 0.00;red vs. blue: P = 0.032). No time difference was observed. The black condition showed negative delta and high-gamma correlations in the central electrodes. The red condition revealed positive alpha and low-gamma correlations in the frontal and occipital areas. Although no correlations were observed in the blue condition, it enhanced attentional performance. Positive alpha and low-gamma waves might be crucial for spotting attentional errors in key areas. The characteristics of supersonic jet noise from a Mach 2.0 nozzle with a thin-nozzle-lip thickness have been experimentally studied over a range of nozzle pressure ratios from 2.0 to 10.0. The acoustic measurements were conducted in an anechoic room providing a free-field environment. Shadowgraph technique was used as the tool to study qualitatively the shock wave patterns which are primarily responsible for shock related noise. The measured screech frequencies under overexpanded conditions are found to agree with Tam’s theoretical results.

本文主要是對日本華僑華人社會之中的祖先祭祀儀式“普度勝會”相關的歷史及現狀進行解析,并就其特質予以論述。長崎、神户及京都、橫濱及東京均爲當代華僑華人聚居的城市。在這些城市之中,華僑華人一直是通過由其祖籍地(僑鄉)——廣東、浙江、福建、台灣等各自同一祖籍地人員而組成的同鄉會、幫來經營社會活動。目前,日本華僑社會之中的普度勝會在所居華僑祖籍多爲福建地區的長崎、神户、京都三地的唐寺(黄檗宗寺院)、關帝廟每年都會舉行。旅日華僑們舉行的普度勝會是由清代傳入的具有强烈道教色彩的佛教儀式及禮儀,不過其特色還在於對因中日間戰争以及日本發生的大地震而遇難的中日兩國人士所進行的祭祀活動。旅居日本的華僑們的歷史交織在作爲祭祀祖先活動的普度會之中并傳承至今。筆者對這些方面予以關注,從而進一步論述旅日華僑舉行的普度會所具有的由近代以前發端的民俗文化傳承在當代的意義。

目的研究乙醇脱氢酶2（ADH2）和乙醛脱氢酶2（ALDH2）基因多态及饮酒习惯与男性结直肠癌易感性的关系。方法在江苏省进行了一项病例-对照研究（结直肠癌患者190例,人群对照222名）,调查研究对象的生活习惯,抽取静脉血,提取白细胞DNA,采用聚合酶链反应（PCR）和变性高效液相色谱法（DHPLC）检测研究对象的ADH2Arg47His（G-A）和ALDH2Glu487Lys（G-A）的基因型。结果①ADH2A/A和ALDH2G/G基因型显著增加结直肠癌的易感性。在调整了年龄和吸烟状况后,ADH2A/A基因型者与携带G等位基因者相比,发生结直肠癌的危险性上升到1.61（95%CI：1.09-2.38）;ALDH2G/G基因型者与携带A等位基因者相比,发生结直肠癌的危险性上升到1.79（95%CI：1.19-2.69）。②与ADH2G和ALDH2A等位基因携带者相比,同时携带ADH2A/A和ALDH2G/G基因型者发生结直肠癌的OR值上升到3.05（95%CI：1.67-5.57）;与不饮酒的ADH2G等位基因携带者相比,拥有ADH2A/A基因型的饮酒者的OR值上升到3.44（95%CI：1.84-6.42）;与不饮酒的ALDH2A等位基因携带者相比,拥有AL-DH2G/G基因型的饮酒者的OR值上升到2.70（95%CI：1.57-4.66）。结论ADH2和ALDH2基因多态性与男性结直肠癌的易感性相关。ADH2,ALDH2基因多态之间以及基因多态与饮酒习惯之间,在结直肠癌发生中存在显著的协同作用。

A Series of Soluble Planar Oligorylenes up to Hexarylene

Preparation of large rylenes often needs the use of solubilizing groups along the rylene backbone,which results in the distortion of the skeleton and thus broadening of the absorption *** this work,we successfully synthesized 2,5,8,16,19,22-hexakis[2,6-bis(tri-fluoromethyl)phenyl]hexarylene(H)by using an oxidative fusion reaction of perylene trimer in 5%*** product is soluble in common organic solvents and could be purified with column ***-crystal X-ray diffraction analysis revealed that the hexarylene core is not disturbed by the substituents very much and is almost *** longest absorption of H exhibits a sharp peak at 831 nm and an on-set at 875 nm in toluene with a small full-width-at-half-maximum(FWHM)of 660 cm^(-1).The molar extinction coefficient is as large as 131,000 M^(-1)cm^(-1).The substituents could not perturb the electronic properties of the hexarylene because aryl groups are perpendicular to the hexarylene core and the carbon atoms at 2,5,16,19-positions have negligible coeffi-cients on its HOMO and LUMO,estimated by theoretical *** a result,the hexarylene having less absorption at the visible light region was stable enough in solution under the LED room light for more than 1 *** strategy gives a useful guideline to establish the near-infrared materials that exhibit stability under ambient conditions.
