Effects of endothelin ETA receptor blocker LU 135252 on cardiac remod-eling and survival in a hypertensive rat model of chronic heart failure

Aim:To investigate whether the endothelin ETAreceptor blocker provides similarbenefit on cardiac remodeling and survival in a hypertensive rat model of chronicheart failure（CHF）.Methods:Male stroke-prone spontaneously hypertensive（SHR-SP）rats were subjected to permanent ligation of the left coronary artery andwere treated for 6 weeks with the endothelin ETAreceptor blocker LU 135252(***-1.d-1)starting 24 h after ligation or ***-operated rats servedas normal *** mean arterial blood pressure（MAP）,heart rate（HR）,leftventricular end-diastolic pressure（LVEdP）,left ventricular contractility(LVdp/dtmax),left ventricular inner diameter（LVd）and circumference（LVC）,septalthickness,left ventricular interstitial collagen content（ICC）and heart weight（HW）were measured at the end of the ***:Compared with the untreatedgroup,LU 135252 tended to increase HW（1.43±0.03 vs 1.38±0.04 g;P>0.05）,increased LVd（7.65±0.24 mm vs 6.58±0.14 mm;Pdly increased LVC（30.11±0.83 mm vs 24.82±0.85 mm;Pd reduced left ventricular ICC（3.79%±0.09% vs 6.71%±0.11%;Pd MAP（132±6mmHg vs142±4 mmHg;P>0.05）,reduced LVEdP（144mmHg vs 27±4 mmHg;Pdimproved LV dp/dtmax（4230±450 mmHg/s vs 1950±400 mmHg/s;Pd significantly（13% vs 11%;P=NS）.Conclusion:In this hyper-tensive rat model of CHF,chronic endothelin ETA receptor blockade with LU 135252improves cardiac hemodynamics,however,it does not affect long-term survivaland worsens cardiac ***,endothelin ETAreceptor antagonists areunlikely to have an important role in the management of patients with CHF.

AIM: To elucidate the cardioprotective effects of T-type calcium channel blocker mibefradil and compare with that of the angiotensin-converting enzyme inhibitor ramipril in a stroke-prone spontaneously hypertensive rats (SHR- SP) model of congestive heart failure (CHF) after myocardial infarction (MI). METHOdS: SHR-SP rats were subjected to permanent ligation of the left anterior decending coronary artery. Treatment with mibefradil (10 mg·kg-1·d-1), ramipril (10 mg·kg-1·d-1), or placebo was initiated 4 weeks before surgery and continued up to 6 weeks after induction of MI. Sham-operated rats served as controls. RESULTS: In placebo-treated MI rats, six weeks after MI, left ventricular circumference, inner diameter, and left ventricular end-diastolic pressure (LVEdP) were increased, whereas mean arterial blood pressure (MAP) and maximum rate of rise of left ventricular pressure (dp/ dtmax) were decreased compared with sham-operated controls (Pd MI rats, heart weight, heart weight to body weight ratio and interstitial collagen content were reduced (PdP was slightly decreased (P>05), and dp/dtmax was improved (Pd with placebo-treated MI rats. In contrast, in mibefradil-treated MI rats, heart weight, heart weight to body weight ratio were slightly but not signifi- cantly reduced, LVEdP was slightly elevated compared with placebo-treated MI rats, and was elevated (Pd with ramipril-treated MI rats, although interstitial collagen content were reduced (Pd with placebo-treated MI ***: Chronic treatment with ramipril diminishes cardiac remodeling of heart failure after MI to a greater extent than mibefradil. Moreover, 6 weeks after MI, mibefradil treatment results in a slight rise in LVEdP compared with placebo-treated rats. Salacca zalacca(Gaertn.) Voss(family Arecaceae) is the snake fruit commonly known in Malay language as salak in Malaysia. This exotic fruit has diverse and potential pharmacological properties due to its high antioxidant content. It is often consumed due to its sweet taste. The abundant natural sugar and fibre along with minerals and vitamin makes it a nutritious fruit. Phytochemical investigation on this fruit has revealed the presence of flavonoids, phenolics, glycosides as well as some volatile and aromatic compounds, including gallic acid, quercetin, chlorogenic acid, epicatechin, proanthocyanidins, lycopene and β-carotene. Pharmacological studies on the fruit flesh and peel have shown some tremendous antioxidant, anti-inflammatory, anticancer and antidiabetic potential. This review provides the botanical information of Salacca zalacca as well as its scientific investigations involving the distinct pharmacological and phytochemical benefits. This could help in highlighting the lacking data and research gaps on this plant.

Mitochondrial protection by low doses of insulin-like growth factor-Ⅰin experimental cirrhosis

AIM： To characterize the mitochondrial dysfunction in experimental cirrhosis and to study whether insulin-like growth factor-Ⅰ （IGF-Ⅰ ） therapy （4 wk） is able to induce beneficial effects on damaged mitochondria leading to cellular protection. METHOdS： Wistar rats were divided into three groups： Control group, untreated cirrhotic rats and cirrhotic rats treated with IGF-Ⅰ treatment （2 μg/1O0 g bw/d）. Mitochondrial function was analyzed by flow cytometry in isolated hepatic mitochondria, caspase 3 activation was assessed by Western blot and apoptosis by TUNEL in the three expedmental groups. RESULTS： Untreated cirrhotic rats showed a mitochondrial dysfunction characterized by a significant reduction of mitochondrial membrane potential （in status 4 and 3）; an increase of intramitochondrial reactive oxigen species （ROS） generation and a significant reduction of ATPase activity. IGF-Ⅰ therapy normalized mitochondrial function by increasing the membrane potential and ATPase activity and reducing the intramitochondrial free radical production. Activity of the electron transport complexes Ⅰ and Ⅲ was increased in both cirrhotic groups. In addition, untreated cirrhotic rats showed an increase of caspase 3 activation and apoptosis. IGF- Ⅰ therapy reduced the expression of the active peptide of caspase 3 and resulted in reduced apoptosis. CONCLUSION： These results show that IGF- Ⅰ exerts a mitochondrial protection in experimental cirrhosis leading to reduced apoptosis and increased ATP production.

didymosphenia geminata (Lyngbye) Schmidt is a type of diatom that exists in Chile as an introduced species, invading the country and its rivers. We collected samples of d. geminata from two sampling points in Chile, assessing their viability and response to control agents. Fresh d. geminata showed more than 90% of viable granular forms (containing granules in their cytoplasm);however, the dry form presents near 50% viability. By creating dry d. geminata through exposure to 38°C temperatures for 7 days, viability values of the granular form decreased to 20%. d. geminata kept at room temperature for more than 4 weeks reported values of granular forms at 50%, while samples that were refrigerated at 4°C maintained values of granular forms at 90% for 4 weeks. Previous studies suggest that high salt concentration affects the viability of d. geminata. When taking wet d. geminata samples and exposing them to a solution of 10% NaCl for 10 minutes, we observed no differences compared to the control samples, finding granular forms at 90%. When the d. geminata was exposed to a 5% soap solution, reductions of over 90% of the granular forms were observed. Our results suggest that the viability of d. geminata is associated with the granular content within their cytoplasm, and that it is possible to alter laboratory conditions for their study. These early studies are important in order to better manipulate the model in the laboratory, allowing us to obtain new evidence regarding the microalgae’s biology through in vitro studies.

Variogram Analyses of Soil Characters in Glacial Moraine Landscapes

The objective of this research is to analyze variogram analyses of soil characters in Glacial Moraine Landscapes. The research site is located in sloping landscapes, Kuehren, North Germany. The survey method was detailed using maps with scales of 1：5,000. Soil sampling was performed by soil pits and borings and completely analyzed in laboratory. Collected data were evaluated by geostatistics program for spatial soil variability analyses. The variogram models show that spatial soil variability ranges between 70-120 m （mean： 85 m）. Effective distances of sampling are calculated at around 50 m. The range values of soil characters are proportional with the range of elevation （range： 70 m, effective distance： 40 m）. The relief determines mainly the spatial variability of soil characters.

The objective of this research is to analyze optimal interpolation and Kriging mapping of soil characters in Glacial Moraine Landscapes. The research site is located in sloping landscapes, Kuehren, North Germany. The survey method was detailed using maps with scales of 1：5,000. Soil sampling was performed by soil pits and borings and completely analyzed in laboratory. Collected data were evaluated by Geostatistics program for spatial soil variability analyses. All maps （produced by Kriging interpolation） picture redistribution of soil nutrients and soil fractions and all map isolines run in similar directions according to landscape nets. The position in the landscape is responsible for increased soil variability. Soil variability becomes higher with decreasing elevation; this means it increases from hilltops to lower slopes. All observed soil characters show relationships to the soil variability. This variability system is caused by convex depressions and hedgerows （Knicks） function as barriers for the redistribution of transported material and offsite sedimentation. Therefore fluxes can be assessed by soil gain and loss balances.

Premenstrual dysphoric disorder(PMdd) affects nearly 5% of women of reproductive age. Symptomatic heterogeneity, together with largely unknown genetics, has greatly hindered its effective treatment. In the present study, analysis of genomic sequencing-based copy number variations(CNVs) called from 100 kb white blood cell dNA sequence windows by means of semisupervized clustering led to the segregation of patient genomes into the d and V groups, which correlated with the depression and invasion clinical types,respectively, with 89.0% consistency. Application of diagnostic CNV features selected using the correlation-based machine learning method enabled the classification of the CNVs obtained into the d group, V group, total patient group, and control group with an average accuracy of 83.0%. The power of the diagnostic CNV features was 0.98 on average, suggesting that these CNV features could be used for the molecular diagnosis of the major clinical types of PMdd. This demonstrated concordance between the CNV profiles and clinical types of PMdd supported the validity of symptom-based diagnosis of PMdd for differentiating between its two major clinical types, as well as the predominantly genetic nature of PMdd with a host of overlaps between multiple susceptibility genes/pathways and the diagnostic CNV features as indicators of involvement in PMdd etiology.

Treatment with Enterococcus faecalis CECT7121 Is Not Effective as Therapy in Mice with an Established Allergy Status

In allergies, an unbalanced immune response towards a T helper (Th) 2 profile with high levels of Immunoglobulin (Ig) E is produced. We have demonstrated that the pre-administration of Enterococcus faecalis CECT7121 prevents the development of allergy in ovalbumin-immunized mice. In this work, we evaluated whether this bacterium can also revert an established allergic status. Mice were immunized with ovalbumin (OVA) and after that, were inoculated with an E. faecalis CECT7121 suspension. In immunized animals, serum specific immune response, proliferative activity of memory splenocytes, and levels of Th2 cytokines were assessed. The in vivo active cutaneous anaphylaxis test was also performed. The treatment with E. faecalis CECT7121 only increased anti-OVA IgG2a levels. No differences were observed in other specific immunological parameters. Probiotic-treatment did not prove to have any desensitizing effect on mice. These results, together with those recently published, can be concluded that this bacterium would not be appropriate for the treatment of allergic symptoms.
