TRPA1 are cation channels expressed in sensory neurons and in several other cell types. This channel is specifically activated by ally isothiocyanate (AITC), the pungent component of mustard oil, as well as by other electrophilic compounds. Although TRPA1 expression in central glia has been reported, its subcellular localization and its expression in peripheral glia have not been investigated before. In this paper we report the molecular and functional expression of TRPA1 in rat cortical astrocytes. Real-time RT-PCR identified low but significant amounts of TRPA1 mRNA in cortical astrocytes while no signal was seen in peripheral glia isolated from dorsal root ganglia (DRG) or in a glial cell line (DITNC-1). Calcium imaging showed AITC-induced signals in astro-cytes while no response in peripheral glia. AITC induced calcium signals in astrocytes in the presence and in the absence of extracellular calcium, suggesting an intracellular localization of TRPA1 channels. Whole cell electrophysiological recordings were performed in astrocytes, in peripheral glia and in DITNC-1 cells transfected with TRPA1 during AITC application. In TRPA1-transfected DITNC-1 cells typical TRPA1 currents were recorded with a reversal potential near 0 mV, consistent with the opening of a non-selective cation channel. No such currents were recorded in untransfected DITNC-1 cells, in astrocytes and in peripheral glial cells, where even high concentrations of AITC (up to 10 mM) induced no significant outward current. In astrocytes AITC transiently induced an outward rectifying current with the reversal potential near ?90 mV, consistent with K channel activation, likely activated by intracellular release of calcium. Our results suggest that TRPA1 channels are molecularly and functionally expressed in calcium-containing organelles of rat cortical astrocytes, with no expression in the plasma membrane.

Body Balance and Core Training in Parkinson’s Disease: A Longitudinal Crossover Study

Objective: The objective of this study was to verify the effects of two different training protocols based on Core Stability and Body Balance exercises in subjects with Parkinsons’s Disease. Methods: Eight elderly men with PD (age of 73 ± 9 years, weight 69 ± 16 kg, height 164 ± 13 cm, 2 ± 1 of Disability Score according to Hoehn and Yahr scale). The disability score was evaluated using Hoehn and Yahr scale. This study was 12-month-long organized in 3 blocks of 4 months each. The sample was divided in two groups formed by 4 subjects each group. During the first four months, one group performed core stability exercises (CSG) while the other group exercised itself through a Balance Exercises Program (BG). After this first part of the study both protocols were stopped for the following 4 months. Finally, the group were reversed (counter balance design) for the last 4 months of physical activity. The two groups were measured twice, before (T0) and after the treatment (T1). Both treatments were designed in order to investigate the improvement in core muscles performance in order to grant a better balance control and to reduce the risk of falling. Results: statistically significant variations were found in the flight time of Sit-to-Stand (p < 0.05, +27%) and in the step length (p < 0.05, ?3%). In the CSG group, statistically significant variations were detected in Speed of Steps (p < 0.05, +5%), Step Cadence (p < 0.05, +3%), and Left Stride Duration (p < 0.05, ?3%) (Table 1). Conclusion: Both Body Balance and Core training can be considered two good physical exercise methods for people with PD. This study highlights the positive effects of this training protocol on legs strength improvements and on the balance control while walking.

Esophageal achalasia is a chronic and progressive motility disorder characterized by absence of esophageal body peristalsis associated with an impaired relaxation of lower esophageal sphincter(LES) and usually with an elevated LES pressure, leading to an altered passage of bolus through the esophago-gastric junction. A definitive cure for achalasia is currently unavailable. Palliative treatment options provide only food and liquid bolus intake and relief of symptoms. Endoscopic therapy for achalasia aims to disrupt or weaken the lower esophageal sphincter. Intra-sphincteric injection of botulinum toxin is reserved for elderly or severely ill patients. Pneumatic dilation provides superior results than botulinum toxin injection and a similar mediumterm efficacy almost comparable to that attained after surgery. Per oral endoscopic myotomy is a promising option for treating achalasia, but it requires increased experience and further objective and long-term follow up. This article will review different endoscopic treatments in achalasia, and summarize the short-term and long-term outcomes.

Gastrointestinal complications are a frequent cause of morbidity after transplantation and may affect up to 40% of kidney transplant recipients. Here we report a rare case of idiopathic giant esophageal ulcer in a kidney transplant recipient. A 37-year-old female presented with a one-week history of odynophagia and weight loss. Upon admission, the patient presented cold sores, and a quantitative cytomegalovirus polymerase chain reaction was positive(105 copies/ml). An upper endoscopy demonstrated the presence of a giant ulcer. Serological test and tissue biopsies were unable to demonstrate an infectious origin of the ulcer. Immunosuppression was reduced and everolimus was introduced. An empirical i.v. therapy with acyclovir was started, resulting in a dramatic improvement in symptoms and complete healing of the ulcer. Only two cases of idiopathic giant esophageal ulcer in kidney transplant recipients have been reported in the literature; in both cases, steroid therapy was successful without recurrence of symptoms or endoscopic findings. However, this report suggests that correction of immune imbalance is mandatory to treat such a rare complication.

Effectiveness of Grassed Buffer in Reducing Spinosad Runoff: Real Data and FOCUS Output

In Italy high-quality vines are grown on sloping fields where pesticide runoff to surface water is possible and vegetated buffers are suggested as mitigation measure. Spinosad is an insecticide used to control pests in vineyards. For regulatory purposes, FOCUS modelling is used to calculate pesticides runoff reduction provided by a vegetated buffer, but there is an urgent need for field-based results. A field trial with artificial runoff was performed in September 2017 in Northern Italy to evaluate the efficiency of a grassed buffer to reduce spinosad runoff. Trial conditions were based on FOCUS scenarios but made worse to draw prudent conclusions. For the first time in Italy, the trial was conducted according to Good Laboratory Practices to increase reliability of results and reproducibility of the study for regulatory purposes. Five plots were tested, each simulating a grassed buffer of 12 m length on a slope of 10% - 13%. The artificial runoff was 200 mm in 3 hours and rainfall pre- and during runoff was 45 mm. Results show that the 12 m buffer completely retained runoff and effectively mitigated runoff concentration, and mean reduction of spinosad concentration was 59%. A width effect exists, and every 6.5 m of buffer reduces concentration by 50%. Comparison of field results with FOCUS modelling shows that both VFSMod and LM models provide acceptable estimation of runoff reduction for a 5 m buffer, while LM seems more precise. For a 10 m buffer the VFSMod is very accurate, while LM underestimates reduction observed in the field.
