Objective: To evaluate the hepatic and renal protective potential of coriander seeds and leaves using animal feed model. Methods: Coriander seeds-and leaves-based sauces were administrated to normal(Study Ⅰ) and carbon tetrachloride(2 mL/kg B.W.)-induced hepatotoxic rabbits(Study Ⅱ). Hepatic and renal biomarkers like aspartate aminotransferase,alanine aminotransferase. alkaline phosphatase, superoxide dismutase and catalase were measured. Results: Coriander leaves-based sauce exerted more decline(P<0.05) in serum aspartate aminotransferase, alanine aminotransferase and alkaline phosphatase levels in the study Ⅱ as 18.32%. 19.91% and 21.24%, respectively. While, hepatic superoxide dismutase and catalase levels were raised significantly(P<0.05) in both studies. Renal parameters also depicted positive impact by the provision of developed sauces. Conclusions: Coriander seeds and leaves based sauces are effective in alleviating the hepato/renal toxicity. The hepatoprotective effect of coriander leaves is more pronounced as compared to coriander seeds.

Objective: To investigate hypocholesterolemic and hypoglycemic potential of citrus peel extract and powder using rodent experimental ***: Considering the fact, rat feeding trial was carried out for a period of 56 d to access the prophylaxis of citrus peel flavonoids by employing normal(study Ⅰ),hyperglycemic(study Ⅱ) and hypercholesterolemic(study Ⅲ) rats. Each study was further divided into three groups to ensure the provision of selected diets, i.e.,control, functional and nutraceutical diets. Likewise, levels of low density lipoproteins and triglycerides reduced 11.39% and 7.89% respectively in hypercholesterolemic rats. Moreover, nutraceutical diet alleviated the sera glucose level by 8.96% in study Ⅱ.Conclusions: Conclusively, inclusion of citrus peel bioflavonoids in dietary therapies is a promising strategy to modulate lipidemic and glycemic attributes without imparting any deleterious effect on hematological parameters.

To evaluate the potential of both jamun (Syzygium cumini) seed and fruit extracts against *** Sprague Dawley rats were used to evaluate hypoglycemic potential of jamun extracts. Purposely, jamun fruit and seed’s ethanolic extracts based diets were provided to normal and high sucrose diet induced hyperglycemic/diabetic rats for sixty days. The serum glucose and insulin levels were monitored at monthly intervals to evaluate hypoglycemic effect of jamun *** results of instant research depicted that both seed and fruit extracts reduce the blood glucose level significantly and also regulate the insulin levels in hyperglycemic rats. It was noted that jamun fruit extract attenuated serum glucose levels to 5.35% and 12.29% in normal and hyperglycemic rats, respectively; while insulin levels were improved by 2.82% and 6.19%, correspondingly. Whereas, jamun seed extract reduced glucose to 7.04% & 14.36% and showed 3.56% & 7.24% higher insulin levels in normal & hyperglycemic rats, *** present research revealed that both jamun fruit and seeds have potent prophylactic role against hyperglycemia. In this respect, diet based regimen may be tailored using jamun fruit/seed and their extracts to alleviate hyperglycemia.

Objective: To explore the hypoglycaemic and hypocholesterolemic potential of conventional and supercritical extracts of black ***: Purposely, rat modelling was carried out for 2 months by designing three studies i.e. study Ⅰ(normal rats), study Ⅱ(hyperglycaemic rats) and study Ⅲ(hypercholesterolemic rats). Each study was further divided into three groups based on diet ***, functional diet(contained extract of black cumin prepared by using conventional solvent) and nutraceutical diet(contained extract of black cumin prepared by supercritical fluid extraction system).Results: During whole trial, an abating trend was observed in the level of serum cholesterol with maximum reduction(12.8%) in nutraceutical group of study Ⅲ. Low density lipoprotein and triglyceride level was also lowered maximum in study Ⅲ as17.1% and 11.6%, respectively. Whereas, highest decline in glucose level was in nutraceutical group of study Ⅱ as 11.2%.Conclusions: Inclusion of black cumin extracts in diet significantly lowers the occurrence of hyperglycaemia and hypercholesterolaemia. Furthermore, hypoglycaemic and hypocholesterolemic potential of nutraceutical diet is more prominent as compared to functional diet.

The diagnosis of non-alcoholic fatty liver disease(NAFLD)and metabolic dysfunction-associated fatty liver disease only on the basis of laboratory parameter score such as Hepatic Steatosis Index which includes liver enzymes,gender,basal metabolic index,and presence of diabetic mellitus is not sufficient to exclude other causes of deranged liver enzymes especially medications and autoimmune related liver *** the guideline suggests ultrasound is the preferred first-line diagnostic procedure for imaging of NAFLD,as it provides additional diagnostic information and the combination of biomarkers/scores and transient elastography might confer additional diagnostic accuracy and evident from previous similar studies too.

Parkinson’s disease(PD),classified under the category of a neurological syndrome,affects the brain of a person which leads to the motor and non-motor *** motor symptoms,one of the major disabling symptom is Freezing of Gait(FoG)that affects the daily standard of living of PD *** treatments target to improve the symptoms of *** of PD at the early stages is an arduous task due to being indistinguishable from a healthy *** work proposed a novel attention-basedmodel for the detection of FoG events and PD,andmeasuring the intensity of PD on the United Parkinson’s Disease Rating *** separate datasets,that is,UCF Daphnet dataset for detection of Freezing of Gait Events and PhysioNet Gait in PD Dataset were used for training and validating on their respective *** results show a definite rise in the various performance metrics when compared to landmark models on these problems using these *** results strongly suggest that the proposed state of the art attention-based deep learning model provide a consistent as well as an efficient solution to the selected *** valueswere obtained for various performance metrics like accuracy of 98.74%for detection FoG,98.72%for detection of PD and 98.05%for measuring the intensity of PD on *** model was also analyzed for robustness against noisy samples,where also model exhibited consistent *** results strongly suggest that the proposed model provides a better classification method for selected problem.

Reasons of Vaccine Hesitancy among Pakistanis with Available COVID-19 Vaccine: The Next Challenge in the Fight against COVID-19

The government of Pakistan started vaccination campaign for the frontline healthcare pro-viders, as they are more likely to be exposed to COVID-19 patients. In the next step, it ex-tended the free vaccination campaign to include people above 60 and 50 years of age re-spectively. The drive is now open to all citizens >18 years of age. There is a significant re-luctance to get vaccinated even though the government has provided it free of cost. The project uses self-reporting method to assess reasons of vaccine hesitancy among Pakistanis and possible solutions to overcome this problem. Concern for serious side effects was the most common issue 154 (48.42%) reported among all groups of people, including the healthcare workers. The major sources of COVID-19 related information were television 117 (36.8%) and healthcare professionals 105 (33%). The participants reported that they can be convinced to get vaccinated if more published data related to vaccine efficacy and safety is available 118 (37.1%), they observe no side effect among vaccine recipients 90 (28.3%) or higher government officials get the same vaccine 39 (12.3%). Vigorous analysis and publication of data generated is paramount to increase the percentage of people being vaccinated. The healthcare professionals should share evidence-based knowledge related to vaccine efficacy and safety through television and social media to increase acceptance rates and hence, decrease mortality and morbidity from COVID-19 in Pakistan.

This study focused on the encapsulation of vancomycin(VAN) into liposomes coated with a red blood cell membrane with a targeting ligand, daptomycin–polyethylene glycol–1,2-distearoyl-sn-glycero-3-phosphoethanolamine, formed by conjugation of DAPT and Nhydroxysuccinimidyl-polyethylene glycol-1,*** formulation is capable of providing controlled and targeted drug delivery to the bacterial cytoplasm. We performed MALDI-TOF, NMR and FTIR analyses to confirm the conjugation of the targeting ligand via the formation of amide bonds. Approximately 45% of VAN could be loaded into the aqueous cores, whereas 90% DAPT was detected using UV–vis spectrophotometry. In comparison to free drugs, the formulations controlled the release of drugs for > 72 h. Additionally, as demonstrated using CLSM and flow cytometry, the resulting formulation was capable of evading detection by macrophage cells. In comparison to free drugs, red blood cell membrane–DAPT–VAN liposomes, DAPT liposomes, and VAN liposomes reduced the MIC and significantly increased bacterial permeability, resulting in > 80% bacterial death within 4 h. Cytotoxicity tests were performed in vitro and in vivo on mammalian cells,in addition to hemolytic activity tests in human erythrocytes, wherein drugs loaded into the liposomes and RBCDVL exhibited low toxicity. Thus, the findings of this study provide insight about a dual antibiotic targeting strategy that utilizes liposomes and red blood cell membranes to deliver targeted drugs against MRSA.
