Intracellular Ca2+ is vital for cell *** of Ca2+ homeostasis contributes to human diseases such as heart failure,neuron-degeneration,and *** ensure an effective intracellular Ca2+ dynamics,various Ca2+ transport proteins localized in different cellular regions have to work in *** central role of mitochondrial Ca2+ transport mechanisms in responding to physiological Ca2+ pulses in cytosol is to take up Ca2+ for regulating energy production and shaping the amplitude and duration of Ca2+ transients in various *** the discovery that isolated mitochondria can take up large quantities of Ca2+ approximately 5 decades ago,extensive studies have been focused on the functional characterization and implication of ion channels that dictate Ca2+ transport across the inner mitochondrial *** mitochondrial Ca2+ uptake sensitive to non-specific inhibitors ruthenium red and Ru360 has long been considered as the activity of mitochondrial Ca2+ uniporter(MCU) .The general consensus is that MCU is dominantly or exclusively responsible for the mitochondrial Ca2+ *** multiple Ca2+ influx mechanisms(e.g.L-,T-,and N-type Ca2+ channel) have their unique functions in the plasma membrane,it is plausible that mitochondrial inner membrane has more than just MCU to decode complex intracellular Ca2+ signaling in various cell *** the last decade,four molecular identities related to mitochondrial Ca2+ influx mechanisms have been *** are mitochondrial ryanodine receptor,mitochondrial uncoupling proteins,LETM1(Ca2+ /H+ exchanger) ,and MCU and its Ca2+ sensing regulatory subunit ***,we briefly review recent progress in these and other reported mitochondrial Ca2+ influx pathways and their differences in kinetics,Ca2+ dependence,and pharmacological *** potential physiological and pathological implications are also discussed.

Covalent organic frameworks(COFs) have recently emerged as a new class of ***,integrated design is crucial to maximizing the performance of COF-incorporating photocatalytic ***,we compare two strategies of installing earth-abundant metal-based catalytic centers into the matrice of a 2 D COF named *** to NUS-55(Co)prepared from the post-synthetic metalation of coordination sites within the COF,the molecular co-catalyst impregnated NUS-55/[Co(bpy)3]Cl2 achieves a seven-fold improvement in visible light-driven H2 evolution rate to 2,480 μmol g^-1h^-1,with an apparent quantum efficiency(AQE) of 1.55% at 450 *** results show that the rational design of molecular anchoring sites in COFs for the introduction of catalytic metal sites can be a viable strategy for the development of highly efficient photocatalysts with enhanced stability and photocatalytic activities.

Sudden cardiac death （SCD） in children and adolescents is uncommon and yet it is devastating for both victim＇s family and the society. Recently, it was increasingly recognized that SCD in young patients with structurally normal heart may be caused by inheritable primary electrical diseases due to the malfunction of cardiac ion channels, a disease entity known as the ion channelopathies. Catecholaminergic polymorphic ventricular tachycardia （CPVT） is a specific form of ion channelopathy which can cause cardiac syncope or SCD in young patients by producing catecholamine-induced bi-directional ventricular tachycardia （BiVT）, polymorphic VT and ventricular fibrillation （VF） during physical exertion or emotion. We reported here an index case of CPVT caused by cardiac ryanodine receptor gene （hRyR2） mutation which presented as cardiac syncope and sudden cardiac arrest in a Chinese adolescent female.

Benign nontraumatic inflammatory stricture of the common bile duct (CBD) may result in obstructive jaundice, which can be misdiagnosed as a malignant tumor of the CBD preoperatively. Two cases with strictures of the mid portion of the common bile duct presenting with obstructive jaundice are reported herein. Preoperative radiological studies prompted us to confidently make the diagnosis of cholangiocarcinoma. However, the postoperative diagnosis on histological examination of the resected lesions was chronic inflammation and fibrosis. The complications of chronic duodenal ulcer are considered as the etiology of these two disorders.

在18世纪到19世纪的200年间,欧洲人才辈出,诞生的大数学家数不胜数。这期间,东方的数学却反常地沉寂。在明治维新以前,日本的数学成就远不如中国;但到了19世纪末,中国的数学反而不如日本。通过比较从明治维新到二战前后中日数学人才的培养,分析了中国这期间数学落后于日本和世界的原因。日本政府明治维新要求老百姓全面向西方学习乃原因之一,而中国人由于坚持"中学为体,西学为用"的信念,因而在追求真理的态度上始终不能全面以赴,也不失数学及其他科学落伍的重要原因。此外,中国早期学习西方以应用科技为主,缺乏对数学的热情,一直到20世纪20年代还没有认识到当代最先进的数学,始终未接触到学问的前沿。另外,日本数学的迅速兴起与其学习方法有密切的关系。没有做学问的热情,没有崇高的志愿,不可能产生杰出的数学家。今日中国数学的前途,端赖于年轻一代数学家的培养,研究生的培养则溯源于中学生的教育。期望重新燃烧起中国人对数学的热情。

Characterization and validation of somatic mutation spectrum to reveal heterogeneity in gastric cancer by single cell sequencing

Aging is a complex biological process that is believed to be caused by a gradual increase of oxidative damage to biological molecules,including mitochondrial DNA(mtDNA)mutations in tissue *** of these aging phenomena gradually occur in advanced age,which are associated with decline in the bioenergetic function of mitochondria and increase of reactive oxygen species(ROS)in somatic *** recent studies,we have demonstrated that ROS play an important role in the accumulation of large-scale deletions and point mutations of mtDNA as well as in the alteration of gene expression profiles elicited by oxidative stress in somatic tissues of aged *** found that the intracellular levels of hydrogen peroxide(HO)and 8-OHdG of aging human fibroblasts were higher than those of the young ***,an increase in the protein and enzyme activity levels of Mn-SOD and a concurrent decrease in the protein and enzyme activity of catalase were noted in HO-induced senescent human ***,the mRNA and protein levels of pyruvate dehydrogenase(PDH)were decreased but those of pyruvate dehydrogenase kinase(PDK)were increased in aging human *** changes in the two regulatory enzymes and other glycolytic enzymes resulted in enhanced reliance on glycolysis and a metabolic switch away from mitochondrial respiration for ATP supply in aging human *** findings from this and other laboratories together suggest that the respiratory function decline,increase in mitochondrial production of ROS,subsequent accumulation of mtDNA mutations,and the alteration in the expression of a few clusters of genes may contribute,in a synergistic manner,to the human aging.

Gastric cancer(GC) is a highly heterogeneous disease with multiple cellular types and poor ***, the cellular evolution and molecular basis of GC at the individual intra-tumor level has not been well demonstrated. We performed single-cell whole exome sequencing to detect somatic singlenucleotide variants(SNVs) and significantly mutated genes(SMGs) among 34 tumor cells and 9 normal cells from a patient with GC. The Complete Prediction for Protein Conformation(CPPC) approach directly predicting the folding conformation of the protein 3D structure with Protein Folding Shape Code, combined with functional experiments were used to confirm the characterization of mutated SMGs in GC cells. We identified 201 somatic SNVs, including 117 non-synonymous mutations in GC cells. Further analysis identified 24 significant mutated genes(SMGs) in single cells, for which a single amino acid change might affect protein conformation. Among them, two genes(CDC27 and FLG) that were mutated only in single cells but not in the corresponding tumor tissue, were recurrently present in another GC tissue cohort, and may play a potential role to promote carcinogenesis, as confirmed by functional characterization. Our findings showed a mutational landscape of GC at intra-tumor level for the first time and provided opportunities for understanding the heterogeneity and individualized target therapy for this disease.
