Screening of agonistic activities against four nuclear receptors in wastewater treatment plants in Japan using a yeast two-hybrid assay

To assess the potential endocrine disruptive effects through multiple nuclear receptors （NRs）, especially non-steroidal NRs, in municipal wastewater, we examined the agonistic activities on four NRs （estrogen receptor α, thyroid hormone receptor α, retinoic acid receptor ct and retinoid X receptor α） of untreated and treated wastewater from municipal wastewater treatment plants （WWTPs） in Japan using a yeast two-hybrid assay. Investigation of the influent and effluent of seven WWTPs revealed that agonistic activities against steroidal and non-steroidal NRs were always detected in the influents and partially remained in the effluents. Further investigation of four WWTPs employing conventional activated sludge, pseudo-anoxic-oxic, anoxic-oxic and anaerobic-anoxic-oxic processes revealed that the ability to reduce the agonistic activity against each of the four NRs varies depending on the treatment process. These results indicated that municipal wastewater in Japan commonly contains endocrine disrupting chemicals that exert agonistic activities on steroidal and non-steroidal NRs, and that some of these chemicals are released into the natural aquatic environment. Although the results obtained in yeast assays suggested that measured levels of non-steroidal NR agonists in the effluent of WWTPs were not likely to cause any biological effect, further study is required to assess their possible risks in detail.

Overexpression of INCREASED CAMBIAL ACTIVITY, a putative methyltransferase, increases cambial activity and plant growth

Cambial activity is a prerequisite for secondary growth in plants; however, regulatory factors controlling the activity of the secondary meristem in radial growth remain elusive. Here, we identified INCREASED CAMBIAL ACTIVITY （ICA）, a gene encoding a putative pectin methyltransferase, which could function as a modulator for the meristematic activity of fascicular and interfascicular cambium in Arabidopsis. An overexpressing transgenic line, 35S：＇1CA, showed accelerated stern elongation and radial thickening, resulting in increased accumulation of biomass, and increased levels of cytokinins （CKs） and gibberellins （GAs）. Expression of genes encoding pectin methylesterases involved in pectin modification together with pectin methyltransferases was highly induced in 355：：ICA, which might contribute to an increase of methanol emission as a byproduct in 35S：ICA. Methanol treatment induced the expression of GA- or CK-responsive genes and stimulated plant growth. Overall, we propose that ectopic expression of ICA increases cambial activity by regulating CK and GA homeostasis, and methanol emission, eventually leading to stem elongation and radial growth in the inflorescence stem.

Radiofrequency ablation as treatment for pulmonary metastasis of colorectal cancer

Radiofrequency ablation (RFA) causes focal coagulation necrosis in tissue. Its first clinical application was reported in 2000, and RFA has since been commonly used in both primary and metastatic lung cancer. The procedure is typically performed using computed tomography guidance, and the techniques for introducing the electrode to the tumor are simple and resemble those used in percutaneous lung biopsy. The most common complication is pneumothorax, which occurs in up to 50% of procedures; chest tube placement for pneumothorax is required in up to 25% of procedures. Other severe complications, such as pleural effusion requiring chest tube placement, infection, and nerve injury, are rare. The local efficacy depends on tumor size, and local progression after RFA is not rare, occurring in 10% or more of patients. The local progression rate is particularly high for tumors > 3 cm. Repeat RFA may be used to treat local progression. Short- to mid-term survival after RFA appears promising and is approximately 85%-95% at 1 year and 45%-55% at 3 years. Long-term survival data are sparse. Better survival may be expected for patients with small metastasis, low carcinoembryonic antigen levels, and/or no extrapulmonary metastasis. The notable advantages of RFA are that it is simple and minimally invasive; preserves pulmonary function; can be repeated; and is applicable regardless of previous treatments. Its most substantial limitation is limited local efficacy. Although surgery is still the method of choice for treatment with curative intent, the ultimate application of RFA may be to replace metastasectomy for small metastases. Randomized trials comparing RFA with surgery are needed.

Objective Shock heart syndrome(SHS)is associated with lethal arrhythmias(ventricular tachycardia/ventricular fibrillation,VT/VF).We investigated whether liposome-encapsulated human hemoglobin vesicles(HbVs)has comparable persistent efficacy to washed red blood cells(wRBCs)for improving arrhythmogenesis in the subacute to chronic phase of *** Optical mapping analysis(OMP),electrophysiological study(EPS),and pathological examinations were performed on blood samples from Sprague-Dawley rats following induction of hemorrhagic *** hemorrhagic shock,the rats were immediately resuscitated by transfusing 5%albumin(ALB),HbV,or *** rats survived for 1 *** and EPS were performed on Langendorff-perfused *** arrhythmias and heart rate variability(HRV)were evaluated using awake 24-h telemetry,cardiac function by echocardiography,and pathological examination of *** OMP showed significantly impaired action potential duration dispersion(APDd)in the left ventricle(LV)in the ALB group whereas APDd was substantially preserved in the HbV and wRBCs *** VT/VF was easily provoked by EPS in the ALB *** VT/VF was induced in the HbV and wRBCs ***,spontaneous arrhythmias,and cardiac function were preserved in the HbV and wRBCs *** showed myocardial cell damage and Connexin43 degradation in the ALB group,all of which were attenuated in the HbV and wRBCs *** LV remodeling after hemorrhagic shock caused VT/VF in the presence of impaired *** to wRBCs,HbV persistently prevented VT/VF by inhibiting persistent electrical remodeling,preserving myocardial structures,and ameliorating arrhythmogenic modifying factors in the subacute to chronic phase of hemorrhagic shock-induced SHS.

Reliability of Relative Paleointensity Recorded in Chinese Loess-Paleosol Sediments

A detailed paleomagnetic and rock-magnetic investigation spanning loess L7 to paleosol S8 has been carried out at the Baoji and Xifeng sections. Results of anisotropy of magnetic susceptibility confirm that the studied loess-paleosol sediments retain primary sedimentary fabrics. Stepwise thermal demagnetization shows that two well-defined magnetization components can be isolated from both loess and paleosol specimens. A low-temperature component, isolated between 100°C and 200°C, is consistent with the present geomagnetic field direction, and a high-temperature component, isolated between 200-300°C and 620-680°C, includes clearly normal and reversed polarities. Isothermal remanent magnetization and thermomagnetic analyses indicate that characteristic remanent magnetization is mainly carried by magnetite and hematite. The Day plot, together with the stratigraphic variations of rock-magnetic parameters, shows that the uniformity of magnetic mineralogy and grain size fulfills the criteria for relative paleointensity （RPI） studies. RPI records have been constructed using natural remanent magnetization （NRM） intensity after thermal demagnetization at 300°C normalized by low-frequency magnetic susceptibility （NRM 300 /χ）. The results show that the RPI record from the Baoji section, where pedogenesis is quite weak, is compatible with the stacked PISO-1500 paleointensity record, suggesting that it might reflect the paleointensity variation of the geomagnetic field. The RPI record from the Xifeng section, where pedogenesis is rather strong, indicates a clear dissimilarity with the stacked PISO-1500 paleointensity record, implying that it does not reflect the paleointensity variation of the geomagnetic field. Our new results show that the NRM 300 /χ from the strongly pedogenetic paleosols does not completely eliminate the pedogenetic （climatic） influence, so it might be unsuitable for a reliable paleointensity study.

Ordovician diorite-quartz diorite mylonite(Saganoseki quartz diorite) was discovered in Sambagawa metamorphic terrane at the northern margin of Saganoseki Peninsula,Kyushu Island,*** LA-ICP-MS zircon U-Pb geochronology revealed that the intrusion age of Saganoseki quartz diorite was 473.3±3.6 *** rocks show the volcanic arc affinity based on the trace element *** the basis of geochronological and geochemical results,Saganoseki quartz diorite is considered to be a member of the Early Paleozoic igneous rocks of the Kurosegawa tectonic *** quartz diorite is located just south of the Median Tectonic Line(MTL)and is in close contact with the pelitic and psammitic schist without any brittle shear zone.U-Pb ages of detrital zircon grains from two psammitic schists show the estimated sedimentation age of early Late Cretaceous,indicate that these psammitic schists are the member of Sambagawa metamorphic *** with these results and the mode of occurrence in the field,we argue that the Early Paleozoic igneous rocks of the Kurosegawa tectonic zone existed as an upper structural unit of the Sambagawa *** relationship is the key to reconstruct the Mesozoic tectonics of Japan as a part of East Asia,and its evolution through time.

Metabolomic Screening Applied to Rice FOX Arabidopsis Lines Leads to the Identification of a Gene-Changing Nitrogen Metabolism

Plant metabolomics developed as a powerful tool to examine gene functions and to gain deeper insight into the physiology of the plant cell. In this study, we screened Arabidopsis lines overexpressing rice full-length （FL） cDNAs （rice FOX Arabidopsis lines） using a gas chromatography-time-of-flight mass spectrometry （GC-TOF/MS）-based technique to identify rice genes that caused metabolic changes. This screening system allows fast and reliable identification of candi- date lines showing altered metabolite profiles. We performed metabolomic and transcriptomic analysis of a rice FOX Ara- bidopsis line that harbored the FL cDNA of the rice ortholog of the Lateral Organ Boundaries （LOB） Domain （LBD）/ Asymmetric Leaves2-1ike （ASL） gene of Arabidopsis, At-LBD37/ASL39. The investigated rice FOX Arabidopsis line showed prominent changes in the levels of metabolites related to nitrogen metabolism. The transcriptomic data as well as the results from the metabolite analysis of the Arabidopsis At-LBD37/ASL39-overexpressor plants were consistent with these findings. Furthermore, the metabolomic and transcriptomic analysis of the Os-LBD37/ASL39-overexpressing rice plants indicated that Os-LBD37/ASL39 is associated with processes related to nitrogen metabolism in rice. Thus, the combination of a metabolomics-based screening method and a gain-of-function approach is useful for rapid characterization of novel genes in both Arabidopsis and rice.

Design and Implementation of NAREGI SuperScheduler Based on the OGSA Architecture

NAREGI is a 5-year Japanese National Grid Project during 2003--2007, whose chief aim is to develop a set of grid middleware to serve as a basis for future e-Science. NAREGI also aims to lead the way in standardization of grid middleware, based on the OGSA architecture. Its super-scheduler is based on the proposed OGSA-EMS Architecture, in that it becomes the first working implementation that implements the documented component relationships within the OGSA-EMS architecture document v.l.0. Through the efforts and experience in the design and implementation, it has been confirmed that the documented OGSA-EMS architecture is quite feasible, but will require significant amount of refinement and speed improvements to finalize its detailed specifications. The super-scheduler also supports co-allocation across multiple sites to support automated execution of grid-based MPIs that execute across machines. Such a resource allocation requires sophisticated interactions between the OGSA-EMS components not covered in the current OGSA-EMS architecture, some of which are non-trivial. Overall, job scheduling with OGSA-EMS has proven to not only work, but also that its job allocation and execution time is within reasonable bounds.

Neoadjuvant treatment for esophageal squamous cell carcinoma

Squamous cell carcinoma and adenocarcinoma are types of esophageal cancer, one of the most aggressive malignant diseases. Since both histological types present entirely different diseases with different epidemiology, pathogenesis and tumor biology, separate therapeutic strategies should be developed against each type. While surgical resection remains the dominant therapeutic intervention for patients with operable esophageal squamous cell carcinoma(ESCC), alternative strategies are actively sought to reduce the frequency of post-operative local or distant disease recurrence. Such strategies are particularly sought in the preoperative setting. Currently, the optimal management of resectable ESCC differs widely between Western and Asian countries(such as Japan). While Western countries focus on neoadjuvant or definitive chemoradiotherapy, neoadjuvant chemotherapy followed by surgery is the standard treatment in Japan. Importantly, each country and region has established its own therapeutic strategy from the results of local randomized control trials. This review discusses the current knowledge, available data and information regarding neoadjuvant treatment for operable ESCC.
