A Mixture of Glycosylated proBNP and BNP-32 Is a Suitable Calibrator for BNP Immunoassays

Aim: Measurement of B-type natriuretic peptide (BNP) is widely used as a diagnostic and risk assessment tool for cardiovascular disease. Recent studies have demonstrated that BNP-32 and its precursor proBNP circulate in the blood stream, and that most commercial BNP immunoassays measure both forms. However, recombinant or synthetic BNP-32 is used as the standard for those BNP immunoassays. This gap between clinical samples and the standard might be a potential source of variation in BNP measurements among assays. The purpose of this study is to validate a more suitable calibrator for BNP immunoassays. Methods: External BNP calibrators containing both BNP-32 and glycosylated proBNP were prepared at two concentration levels. Target BNP concentrations of the low and high levels were 40 and 160 pg/mL, respectively. And to reflect clinical samples, the molar ratios of BNP-32 to glycosylated proBNP in these concentration levels were adjusted to 50:50 and 25:75, respectively. BNP concentrations of plasma samples along with the external BNP calibrators were measured at two commercial labs and using an automated analyzer MI02 Shionogi®BNP (MI02). These samples and the calibrators were also measured using an immunoradiometric assay (IRMA) as a standard assay procedure. Concentrations of the plasma samples measured at the labs or using the MI02 were adjusted according to a comparison of the measured concentrations of the external BNP calibrators with the IRMA. Results: After measured concentrations of the plasma samples were adjusted using the external BNP calibrators, the correlation between each measurement and the IRMA was improved. The range of the slopes according to Passing-Bablok regression analysis narrowed from 0.628 - 0.955 to 0.911 - 1.005. Conclusions: Our data suggests that a mixture of BNP-32 and glycosylated proBNP at different ratios by concentration level is suitable for calibration to minimize variations in BNP measurements among immunoassays.

A 61-kb biosynthetic gene cluster(BGC),which is accountable for the biosynthesis of hibarimicin(HBM)B from Microbispora rosea *** TP-A0121,was heterologously expressed in Streptomyces coelicolor M1154,which generated a trace of the target products but accumulated a large amount of shunt *** on rational analysis of the relevant secondary metabolism,directed engineering of the biosynthetic pathways resulted in the high production of HBM B,as well as new HBM derivates with improved antitumor *** results not only establish a biosynthetic system to effectively synthesize HBMs-a class of the largest and most complex Type-Ⅱpolyketides,with a unique pseudo-dimeric structure-but also set the stage for further engineering and deep investigation of this complex biosynthetic pathway toward potent anticancer drugs.

Objective:To evaluate the combination therapy of pyronaridine tetraphosphate and diminazene aceturate against Babesia in vitro and in ***:Bioinformatic analysis was performed using atom pair *** in vitro combination test was performed against Babesia bovis and Theileria ***,the in vivo chemotherapeutic efficacy of pyronaridine tetraphosphate in combination with diminazene aceturate was investigated against the growth of Babesia microti in mice using a fluorescence inhibitory ***:Pyronaridine tetraphosphate and diminazene aceturate exhibited nearly similar molecular *** in vitro combination of pyronaridine tetraphosphate and diminazene aceturate was synergistic on Babesia bovis and additive on Theileria *** addition,5 mg/kg pyronaridine tetraphosphate combined with 10 mg/kg diminazene aceturate inhibited Babesia microti growth significantly compared with those observed after treatment with 25 mg/kg diminazene aceturate alone from day 6 post treatment to day 12 post *** combination therapy also normalized the hematological parameters of infected ***:An oral dose of pyronaridine tetraphosphate combined with a subcutaneous dose of diminazene aceturate inhibits Babesia in vitro and in mice,suggesting it might be a new paradigm for the treatment of babesiosis.

A two-step surface-based 3D deep learning pipeline for segmentation of intracranial aneurysms

The exact shape of intracranial aneurysms is critical in medical diagnosis and surgical *** voxel-based deep learning frameworks have been proposed for this segmentation task,their performance remains *** this study,we offer a two-step surface-based deep learning pipeline that achieves significantly better *** proposed model takes a surface model of an entire set of principal brain arteries containing aneurysms as input and returns aneurysm surfaces as output.A user first generates a surface model by manually specifying multiple thresholds for time-of-flight magnetic resonance angiography *** system then samples small surface fragments from the entire set of brain arteries and classifies the surface fragments according to whether aneurysms are present using a point-based deep learning network(PointNet++).Finally,the system applies surface segmentation(SO-Net)to surface fragments containing *** conduct a direct comparison of the segmentation performance of our proposed surface-based framework and an existing voxel-based method by counting voxels:our framework achieves a much higher Dice similarity(72%)than the prior approach(46%).

作为隔离型单向直流变换器常用的整流器件之一,二极管结电容易与变压器漏感发生谐振,导致严重的电压尖峰问题。为此,提出一种基于二次侧移相的单向隔离型谐振开关电容变换器(UI-RSCC)拓扑,其二次侧整流单元由一个开关半桥和二极管半桥组成,避免了二次侧二极管振荡的产生,可实现所有开关器件的零电压软开关(ZVS)。该文在分析UI-RSCC拓扑的基础上,研究了其输入直流电容的电压均衡机制,提出了相应的输出电压控制策略及其软开关条件。结果表明,UI-RSCC可实现输入直流电容电压的自动均衡,降低了开关器件的电压应力;通过复用部分开关,减少了所需的开关器件数量;使用电容滤波即可实现所有开关器件的软开关,且无需额外的钳位电路。最后,通过仿真及实验结果验证了理论分析的正确性。

Niche Differentiation of Phenol-Degrading Microorganisms in UASB Granular Sludge as Revealed by Fluorescence in situ Hybridization
