An efficient implicit dynamic finite element method(FEM) for elastic 3D objects with uniform cross-sections was developed. In this method, the finite element mesh is generated in such a way that the object to be analyzed is at first sliced into layers with the same thickness along the generatrix and then each layer is discretized into the same pattern. This way of discretization makes the mass, viscosity, and stiffness matrices into the repetitive block tridiagonal matrices. The repetitive block tridiagonal matrix has the characteristic, that the sequence of matrices which appears in the Gaussian elimination for the repetitive block tridiagonal matrix is a rapid convergent sequence. The process of the Gaussian elimination can be terminated when the sequence converges. The rest of the sequence is not necessary to be stored. The present method can save the computational time and memory by utilizing this characteristic of the repetitive block tridiagonal matrix. A few examples of analyses including whole Hopkinson-bar analysis were performed to demonstrate the effectiveness of the present method. The present method is applicable not only to the elasto-dynamics but also to many other problems, such as thermal and electrical problems.

The purpose of this study is to improve the efficiency of the power generation system of a solar tower using fluid dynamics. The power generation system of a solar tower can be designed and constructed at relatively low cost. However, the energy output tends to be low for its physical size compared with other renewable energy production systems. The technical and scientific improvement of these types of generation systems has lost its momentum since the shutdown of the wellknown Spanish pilot plant “Manzanares Solar Chimney” in 1989, although it still has the potential to play a role in renewable energy in the future. We have focused on the tower component of the system to seek possible enhancements of the power output of the internal turbine. As a result of our fluid dynamic shape optimization, a diffuser-shaped tower was employed to increase the internal flow speed of a scaled model. The results show a remarkable improvement in the power output of the internal wind turbine.

Effect of L-lysine in culture medium on nodule formation by bone marrow cells

The aim of this study was to estimate the effect of L-lysine on nodule formation by rat bone marrow cells in vitro. In this study, L-lysine was added to medium for mesenchymal stem cell culture to promote proliferation and differentiation of the cells, and then nodule formation was estimated in an in vitro rat bone marrow cell culture. Bone marrow cells from the bone shafts of the femora of Fischer 344 rats were cultured in minimum essential medium with 20 μl of L-lysine solution at 10﹣4, 10﹣5, 10﹣6, 10﹣7 or 10﹣8 M. Dexamethasone was also added to the medium at 10 nM for differentiation of stem cells from bone marrow into osteoblast progenitor cells. The subculture was performed for 2 weeks. The quantity of osteocalcin in rat bone marrow cell culture with dexamethasone was 392 ng/ml. In the medium with dexamethasone and 10﹣8 M L-lysine, the quantity of osteocalcin was 437 ng/ml. Nodules only formed upon addition of 20 μl of L-lysine at 10﹣8 M. It was indicated that 10﹣8 M L-lysine should be the optimal concentration for calcification. For nodule formation by rat bone marrow cells in vitro, the optimum concentration of L-lysine in culture medium might be 20 μl of 10﹣8 M. L-lysine could play an important role in matrix production for bone formation in vitro.

In rabbit platelet rich plasma （PRP）, 5-hydroxytraptamine （5-HT,0.03～3μmol/L） induced decrease in light transmission （DLT） dose-de pendently with centralization, as revealed by electron microscopy. However, 5-HT did not induced platelet aggregation and release reaction. 5-HT-induced DLT was inhibited by methysergide （0.3～30μmol/L）, indomethacin （0.3～10μmol/L） and PGE1（0.01～0.3μmol/L） in a dose-dependent manner, but not EGTA（0.3～3mmol/L）. Collagen（Coll）, arachidonic acid （AA）, adenosine diphosphate （ADP） and a stable thromboxane A2 analoge（STA2） also induced DLT before aggregation by themselves, which was also inhibited by PGE1, but not inhibited by EGTA except for Coll However,Coll-, AA-, STA2-and ADP-induced DLT were reduced by pretreatment of PRP with 5-HT dose-depen-dently. The duration of DLT by Coll and AA were decreased from 151.4±5.93 sec and 15（?）38±0.60sec to 45.44±4.04 sec and 9.00±1.25 sec respectively (（?）±s（?） P<0.01), but not by ADP and STA2, 3μmol/L of ADP-and 0.3μmol/L of STA2-induced aggregation which was not accompanied with release reaction were enhanced by 5-HT pretreatment, but in those （Coil 5μg/ml, AA 100～200μmol/L and STA21～3 μmol/L） with release reaction, the amount of adenosine triphosphate（ATP）were suppressed significantly （P<0.001） by 5-HT pretreatment without the effect on the magnitude of aggregation, The mobilization of cytosolic free calcium concentration ([Ca2+]i) was observed after 5-HT treatment in single washed platelet, the results indicated that the basic level of [Ca2+] i was 64.78±3.24nmol/L and this level was increased dose-dependently and significantly at 30 sec after administration of 5-HT and the time of peak value of [Ca2+] i was at 90～100 *** similar time courses of suppression of ATP released during aggregation, in cases of Coll（5μg/ml）, AA （200μmol/L） and STA2（3μmol/L）, by 5-HT were also found in the present experiment.

Dextran coating on and among fibers of polymer sponge scaffold for osteogenesis by bone marrow cells in vivo

Although hydroxyapatite is commonly used as a scaffold for bone regeneration, sponges may be suitable because of the adaptability to the defect. To use as a scaffold, the fiber of sponge would be coated with any adhesive to storage stem cells in the sponges. Fiber in the structure of commercially available sponges was coated by immersion in dextran solution and air dried. After seeding of rat bone marrow cells (rBMCs), the sponges were implanted subcutis of rats for estimate osteogenesis in vivo. The level of osteocalcin was 25.28 ±5.71 ng/scaffold and that of Ca was 129.20 ±19.69 μg/scaffold. These values were significantly high- er than those in sponges without dextran coating (p <0.01). It was thought that rBMCs could be stored on the shelf by dextran deposition in the fiber of the sponge. In vivo examination, dextran induced osteogenesis by rBMCs in many spaces in the inner structure of the sponge.

The Effects of an Unloading Knee Brace and Insole with Subtalar Strapping for Medial Osteoarthritis of the Knee

Objectives: We evaluated whether a valgus knee brace and an insole with subtalar strapping could reduce pain and improve functional scores over 12 months in patients with medial knee osteoarthritis (OA). Methods: OA was confirmed by radiography in all patients, who were divided into three groups: exercise (n = 44), insole (n = 55), and brace (n = 19). Clinical knee functions and gait analyses were evaluated. Results: After 12 months, the clinical results for all groups had improved compared to pretreatment findings. Although the knee varus moment decreased and the gait speed increased when the insole was worn initially, the effects of the insole decreased at 6 and 12 months. In contrast, the knee varus moment decreased and the gait speed increased when the brace was initially fitted. The effects of the brace were maintained during the 12 months. Conclusions: The insole was effective for patients with Kellgren-Lawrence Grades II and III, and the knee brace was effective for patients with Grades III and IV conditions. Furthermore, the knee brace was more effective when worn for more than 6 months, while the effects of the insoles were not continuous.

Bone Formation in a Scaffold Composed of Cylindrical Hydroxyapatite and Tryptophan- or Lysine-Coated Sponge <i>in Vivo</i>

Because of the three-dimensional structure of bone or hard tissue such as a tooth, a scaffold is necessary for its regeneration by cellular engineering. Commonly, for in vivo examination, hydroxyapatite (HA) has been used as such a scaffold. Cylindrical HA with a hollow center, which included a columnar formalin-treated polyvinyl alcohol sponge, was used in this examination as a scaffold. The sponge had been coated with L-tryptophan or L-lysine before insertion into the hollow center of the HA. Rat bone marrow cells (rBMCs) derived from the femur were seeded in the sponge before insertion into the hollow center of HA. The number of rBMCs seeded in each sponge was 1.5 × 106. These scaffolds were implanted subcutaneously into the backs of Fischer 344 rats for 6 weeks. In the amino-acid-coated sponge in HA, osteogenesis was found histologically. An osteocalcin level of approximately 10 ug was measured in the scaffolds with L-tryptophan-coated formalized poly-vinyl alcohol sponge containing rBMCs, 4 ug on average in the scaffolds with L-lysine-coated sponge containing the cells and about 2 ug in each scaffold with uncoated sponge containing the cells. The structure of the scaffolds used in this study was thought to be suitable for osteogenesis by rBMCs. It was concluded that tryptophan, as a factor for bone formation by stem cells, functioned by promoting cell adhesion and the differentiation of stem cells into osteoblasts.
