Effects of adacolumn selective leukocytapheresis on plasma cytokines during active disease in patients with active ulcerative colitis

AIM： To investigate the relationship between ulcerative colitis （UC） clinical activity index （CAI） and circulating levels of IL-1ra, IL-10, IL-6 and IL-18. METHODS： Blood levels of IL-1ra, IL-10, IL-6 and IL-18 were measured in 31 patients with active UC, the mean CAI was 11.1, ranging from 5-25; and 12 healthy individuals as controls. Patients were given granulocyte and monocyte adsorptive apheresis （GMA） with Adacolumn. Leucocytes which bear the FcyR and complement receptors were adsorbed to the column leucocytapheresis carriers. Each patient could receive up to 11 GMA sessions over 8 wk. RESULTS： We found strong correlations between CAI and IL-10 （r = 0.827, P 〈 0.001）, IL-6 （r = 0.785, P 〈 0.001） and IL-18 （r = 0.791, P 〈 0.001）. IL-1ra was not correlated with CAI. Following GMA therapy, 24 of the 31 patients achieved remission and the levels of all 4 cytokines fell to the levels in healthy controls. Further, blood levels of IL-1ra and IL-10 increased at the column outflow and inflow at 60 min suggesting release from leucocytes that adhered to the carriers. CONCLUSION： Elevated blood levels of IL-6 and IL-18 together with peripheral blood granulocytes and monocytes/macrophages in patients with active UC show activative behaviour and increased survival time can be proinflammatory and the targets of GMA therapy.

利用淋巴肉瘤细胞上肿瘤相关抗原(TAA)的单克隆抗体,来评价末梢血液淋巴细胞的补体依赖抗体细胞毒性试验在诊断地方性淋巴肉瘤(EBL)上的应用。28头EBL 牛中有27头表现出细胞毒素指数(CI)超过31.7,CI 值31.7作为 EBL 阳性的最低值;其余1头牛的末稍血液淋巴细胞(PBL)和肿瘤细胞其 CI 值分别为28.0和61.0。另外,有一例被牛白血病病毒(BLV)感染而未出现淋巴肉瘤症状的牛和9头未感染 BLV的牛,其 CI 值均小于31.7。根据血相检查,TAA 阳性细胞往往是呈不典型的形态。因为 TAA 阳性细胞在 EBL 牛的外周血液里循环,使用单克隆抗体进行 PBL 的 CDAC 试验,是诊断 EBL 适用的诊断方法。另外,也可用于无白血病或临床症状不明显的 EBL 病例诊断。

Effects of K. Lysolecithin on Blood Levels of Monoamines in Mice

In this research, Lysolecithin-a substance made with 100% natural ingredients - was given to ICR mice as medication to measure its periodic effect on the noradrenalin (NA), dopamine (DA), and serotonin (5-HT) levels of the brain. Both ICR and SAM mice were separated into two groups - control group and Lysolecithin (K. Lysolecithin: hydrolytic lysolecithin) medicated group, and given 1-week preparation period. The K. Lysolecithin group was given 500mg/kg of K. Lysolecithin at 0.2mL per dosage for 4 weeks, and the control group was given the same amount of dosage of water during the same period. NA, DA and 5-HT concentrations were measured from the blood before medication and 8 weeks / 12 weeks / 16 weeks after the first medication. For the SAM mice, 8 weeks after they were medicated with K .Lysolecithin, Morris Water Maze Test was conducted for 7 consecutive days and then the concentrations were measured by drawing blood from the heart. The K. Lysolecithin medicated group showed a tendency to have a statistically significant higher concentrations of 5-HT and NA in the blood. Also, periodic examination showed that the monoamine levels were highest in the 12th week and declined thereafter.

Clinically diagnosed late-onset fulminant Wilson's disease without cirrhosis: A case report

A 64-year-old woman was referred to our hospital with jaundice of the bulbar conjunctiva and general fatigue. After admission, she developed hepatic encephalopathy and was diagnosed with fulminant hepatitis based on the American Association for the Study of Liver Disease(AASLD) position paper. Afterwards, additional laboratory findings revealed that serum ceruloplasmin levels were reduced, urinary copper levels were greatly elevated and Wilson's disease(WD)-specific routine tests were positive, but the Kayser-Fleischer ring was not clear. Based on the AASLD practice guidelines for the diagnosis and treatment of WD, the patient was ultimately diagnosed with fulminant WD. Then, administration of penicillamine and zinc acetate was initiated; however, the patient unfortunately died from acute pneumonia on the 28 th day of hospitalization. At autopsy, the liver did not show a bridging pattern of fibrosis suggestive of chronic liver injury. Here, we present the case of a patient with clinically diagnosed late-onset fulminant WD without cirrhosis, who had positive disease-specific routine tests.

Correlation of N-myc downstream-regulated gene 1 expression with clinical outcomes of colorectal cancer patients of different race/ethnicity

AIM： To evaluate the role of N-myc downstream- regulated gene 1 （NDRG1） expression in prognosis and survival of colorectal cancer patients with different ethnic backgrounds. METHODS： Because NDRG1 is a downstream target of p53 and hypoxia inducible factor-1α （HIF-1α）, we examined NDRG1 expression together with p53 and HIF-1α by irnmunohistochernistry. A total of 157 colorectal cancer specimens including 80 from Japanese patients and 77 from US patients were examined. The correlation between protein expression with clinicopathological features and survival after surgery was analyzed. RESULTS： NDRG1 protein was significantly increased in colorectal tumor compared with normal epithelium in both Japanese and US patient groups. Expression of NDRG1 protein was significantly correlated with lymphatic invasion, venous invasion, depth of invasion, histopathological type, and Dukes＇ stage in Japanese colorectal cancer patients. NDRG1 expression was correlated to histopathological type, Dukes＇ stage and HIF-1α expression in US-Caucasian patients but not in US-African American patients. Interestingly, Kaplan-Meier survival analysis demonstrated that NDRG1 expression correlated significantly with poorer survival in US-African American patients but not in other patient groups. However, in p53-positive US cases, NDRG1 positivity correlated significantly with better survival. In addition, NDRG1 expression also correlated significantly with improved survival in US patients with stages Ⅲ and IV tumors without chemotherapy. In Japanese patients with stages Ⅱ and Ⅲ tumors, strong NDRG1 staining in p53- positive tumors correlated significantly with improved survival but negatively in patients without chemotherapy. CONCLUSION： NDRG1 expression was correlated with various clinicopathological features and clinical outcomes in colorectal cancer depending on the race/ethnicity of the patients. The object of this study is to obtain experimental data for the boiling heat transfer coefficients and the pressure drops for flow of CO in a horizontal small tube, of which inner diameter is 1.0mm. The experimental conditions considered in this study are following range of variables: evaporation temperature is 0℃, mass flux is from 100 to 800 kg/(ms), heat flux is from 5 to 40kW/m, quality is from 0.1 to 0.8. As a result, the experimental values of the frictional pressure drop are smaller than that given by Chisholm-Laird’s correlation. In the boiling heat transfer coefficient of CO, the nucleate boiling is predominant when the mass flux is low and the forced convection evaporation is predominant when the mass flux is high. Thus it can be assumed that the mechanism of boiling heat transfer of CO in a small tube is similar to that in a tube of relatively large diameter.

Bioethanol Production from Molasses by Yeasts with Stress-Tolerance Isolated from Aquatic Environments in Japan

Bioethanol is a safe and renewable source of energy that continues to be a research focus, since fossil fuels have been linked to global warming and nuclear energy sources are affected by the increased safety concerns following the 2011 nuclear power plant accident in Japan. In general, bioethanol is converted from a biomass by yeast fermentation. The production efficiency of this bioethanol is not sufficiently high, and its practical use as a substitute for fossil fuels and nuclear energy is thus limited. For the industrial production of bioethanol, the yeast fermentation of biomass cultures containing high concentration sugar, NaCl, and ethanol is necessary, but this might induce phenomena in which the stresses arising in the yeasts weaken their cells during fermentation. As described herein, we isolated 1028 strains of yeasts from natural aquatic environments: Japan’s Tama River and Lake Kasumigaura. Among them, 412 strains were fermentative yeasts and 31 strains showed high fermentation ability under a 30% sorbitol + 10% ethanol condition. These strains were identified as Torulaspola delbrueckii, Wickerhamomyces anomalus, Candida glabrata, Pichia kudriavzevii, Saccharomyces cf. cerevisiae/paradoxus, and Lachancea kluyveri. The strains T. delbrueckii, W. anomalus, and C. glabrata also showed tolerance against 15% NaCl. Most importantly, S. cf. cerevisiae/paradoxus H28 and L. kluyveri F2-67 produced 57.4 g/L and 53.9 g/L ethanol from molasses (sucrose 104.0 g/L, fructose 33.4 g/L, and glucose 24.8 g/L) within 48 hrs at 25°C, respectively.
