INTRODUCTION The Mongolian gerbil(Meriones unguiculatus)has been used as an animal model for pharmacology(Wu et al.2008),parasitology(Lim et al.2004),and studies of reproduction(Kai et al.1995).The molecular mechanisms of several coat-color phenotypes are known in gerbils(Petrij et al.2001,2007).On the other hand,although albinism in gerbils was first reported by Matsuzaki et al.(1989),its molecular mechanism is still unclear.In a previous study,we reported the establishment of the albino gerbil strain(strain code:MON/Num/c)as a closed colony and the agouti-colored strain(strain code:MON/Num/a)as inbred.Our albino strain had 4 nucleotide substitutions and 2 missense mutations(R77H in exon 1 and Q473R in exon 5)in the tyrosinase(Tyr)gene(Ukaji et al.2016).

BACKGROUND Sorafenib is an oral drug that prolongs overall survival(OS)in patients with hepatocellular carcinoma.Adverse events,including hand-foot skin reaction(HFSR),lead to permanent sorafenib discontinuation.AIM To clarify the association between interventions for adverse events and patient prognosis.METHODS We performed a retrospective,multicenter study of patients treated with sorafenib monotherapy between May 2009 and March 2018.We developed a mutual cooperation system that was initiated at the start of sorafenib treatment to effectively manage adverse events.The mutual cooperation system entailed patients receiving consultations during which pharmacists provided accurate information about sorafenib to alleviate the fear and anxiety related to adverse events.We stratified the patients into three groups:Group A,patients without HFSR but with pharmacist intervention;Group B,patients with HFSR and pharmacist interventions unreported to oncologists(nonmutual cooperation system);and Group C,patients with HFSR and pharmacist interventions known to oncologists(mutual cooperation system).OS and time to treatment failure(TTF)were evaluated using the Kaplan-Meier method.RESULTS We enrolled 134 patients(Group A,n=41;Group B,n=30;Group C,n=63).The median OS was significantly different between Groups A and C(6.2 vs 13.9 mo,p<0.01)but not between Groups A and B(6.2 vs 7.7 mo,P=0.62).Group A vs Group C was an independent OS predictor(HR,0.41;95%CI:0.25-0.66;P<0.01).In Group B alone,TTF was significantly lower and the nonadherence rate was higher(P<0.01).In addition,the Spearman’s rank correlation coefficients between OS and TTF in each group were 0.41(Group A;P<0.01),0.13(Group B;P=0.51),and 0.58(Group C;P<0.01).There was a highly significant correlation between OS and TTF in Group C.However,there was no correlation between OS and TTF in Group B.CONCLUSION The mutual cooperation system increased treatment duration and improved prognosis in patients with HFSR.Future prospective studies(e.g.,randomized controlled trials)an

This paper treats the flow instabilities in a mixed flow pump with a vaned diffuser. Test pump has a positive slope of a head-flow performance curve at 65% flow rate of BEP (Best Efficiency Point) because of a rotating stall. Dynamic Particle Image Velocimetry (PIV) and pressure fluctuation measurements are used for investigating the propagation mechanism of a rotating stall. It was found that unstable performance was caused by periodical large scale abrupt backflow generated from the vaned diffuser to the outlet of impeller. Further, the relation between the static pressure at the inlet of diffuser vane and the internal flow condition was clarified. From these experimental results, in order to improve the positive slope of a head-flow performance curve, to suppress the growth of strong vortex toward the inlet of diffuser vane was proved to be a key point.

AIM: To identify the precise location of putative tumor suppressor genes (TSGs) on the short arm of chromo- some 8 in patients with hepatocellular carcinoma (HCC). METHODS: We used 16 microsatellite markers informative in Japanese patients, which were selected from 61 pub- lished markers, on 8p, to analyze the frequency of loss of heterozygosity (LOH) in each region in 33 cases (56 lesions) of HCC. RESULTS: The frequency of LOH at 8p23.2-21 with at least one marker was 63% (20/32) in the informative cases. More specifically, the frequency of LOH at 8p23.2, 8p23.1, 8p22, and 8p21 was 6%, 52%, 47%, and 13% in HCC cases. The LOH was significantly more frequent at 8p23.1 and 8p22 than the average (52% vs 22%, P = 0.0008; and 47% vs 22%, P = 0.004, respectively) or others sites, such as 8p23.2 (52% vs 6%, P = 0.003; 47% vs 22%, P = 0.004) and 8p21 (52% vs 13%, P = 0.001; 47% vs 13%, P = 0.005) in liver cancer on the basis of cases. Notably, LOH frequency was significantly higher at D8S277, D8S503, D8S1130, D8S552, D8S254 and D8S258 than at the other sites. However, no allelic loss was detected at any marker on 8p in the lesions of nontumor liver tissues. CONCLUSION: Deletion of 8p, especially the loss of 8p23.1-22, is an important event in the initiation or promotion of HCC. Our results should be useful in identi- fying critical genes that might lie at 8p23.1-22.

Earthquake engineering research and development have received much attention since the first half of the twentieth century. This valuable research presented a huge step forward in understanding earthquake hazard mitigation,which resulted in appreciable reduction of the effects of past earthquakes. Nevertheless,the 2011 Tohoku earthquake and the subsequent tsunami resulted in major damage. This paper presents the timeline of earthquake mitigation and recovery,as seen by the authors. Possible research directions where the authors think that many open questions still remain are identified. These are primarily based on the important lessons learned from the 2011 Tohoku earthquake.

Low-frequency repetitive transcranial magnetic stimulation（LF-r TMS） to the contralesional hemisphere and intensive occupational therapy（i OT） have been shown to contribute to a significant improvement in upper limb hemiparesis in patients with chronic stroke. However, the effect of the combined intervention program of LF-r TMS and i OT on cognitive function is unknown. We retrospectively investigated whether the combined treatment influence patient＇s Trail-Making Test part B（TMT-B） performance, which is a group of easy and inexpensive neuropsychological tests that evaluate several cognitive functions. Twenty-five patients received 11 sessions of LF-r TMS to the contralesional hemisphere and 2 sessions of i OT per day over 15 successive days. Patients with right- and left-sided hemiparesis demonstrated significant improvements in upper limb motor function following the combined intervention program. Only patients with right-sided hemiparesis exhibited improved TMT-B performance following the combined intervention program, and there was a significant negative correlation between Fugl-Meyer Assessment scale total score change and TMT-B performance. The results indicate the possibility that LF-r TMS to the contralesional hemisphere combined with i OT improves the upper limb motor function and cognitive function of patients with right-sided hemiparesis. However, further studies are necessary to elucidate the mechanism of improved cognitive function.

To date, many studies related to robots have been performed around the world. Many of these studies have assumed operation at locations where entry is difficult, such as disaster sites, and have focused on various terrestrial robots, such as snake-like, humanoid, spider-type, and wheeled units. Another area of active research in recent years has been aerial robots with small helicopters for operation indoors and outdoors. However,less research has been performed on robots that operate both on the ground and in the air. Accordingly, in this paper, we propose a hybrid aerial/terrestrial robot system. The proposed robot system was developed by equipping a quadcopter with a mechanism for ground movement. It does not use power dedicated to ground movement, and instead uses the flight mechanism of the quadcopter to achieve ground movement as well. Furthermore, we addressed the issue of obstacle avoidance as part of studies on autonomous control. Thus, we found that autonomous control of ground movement and flight was possible for the hybrid aerial/terrestrial robot system, as was autonomous obstacle avoidance by flight when an obstacle appeared during ground movement.

The human CCR4-NOT deadenylase complex consists of at least nine enzymatic and non-enzymatic subunits.Accumulating evidence suggests that the non-enzymatic subunits are involved in the regulation of mRNA deadenylation,although their precise roles remain to be established.In this study,we addressed the function of the CNOT1 subunit by depleting its expression in HeLa cells.Flow cytometric analysis revealed that the sub G1 fraction was increased in CNOT1-depleted cells.Virtually,the same level of the sub G1 fraction was seen when cells were treated with a mixture of siRNAs targeted against all enzymatic subunits,suggesting that CNOT1 depletion induces apoptosis by destroying the CCR4-NOT-associated deadenylase activity.Further analysis revealed that CNOT1 depletion leads to a reduction in the amount of other CCR4-NOT subunits.Importantly,the specific activity of the CNOT6L immunoprecipitates-associated deadenylase from CNOT1-depleted cells was less than that from control cells.The formation of P-bodies,where mRNA decay is reported to take place,was largely suppressed in CNOT1-depleted cells.Therefore,CNOT1 has an important role in exhibiting enzymatic activity of the CCR4-NOT complex,and thus is critical in control of mRNA deadenylation and mRNA decay.We further showed that CNOT1 depletion enhanced CHOP mRNA levels and activated caspase-4,which is associated with endoplasmic reticulum ER stress-induced apoptosis.Taken together,CNOT1 depletion structurally and functionally deteriorates the CCR4-NOTcomplex and induces stabilization of mRNAs,which results in the increment of translation causing ER stress-mediated apoptosis.We conclude that CNOT1 contributes to cell viability by securing the activity of the CCR4-NOT deadenylase.
