Activation of hypoxia-inducible factor 1 attenuates periapical inflammation and bone loss

Hypoxia（low oxygen level） is an important feature during infections and affects the host defence mechanisms. The host has evolved specific responses to address hypoxia, which are strongly dependent on the activation of hypoxia-inducible factor 1（HIF-1）.Hypoxia interferes degradation of HIF-1 alpha subunit（HIF-1α）, leading to stabilisation of HIF-1α, heterodimerization with HIF-1 beta subunit（HIF-1β） and subsequent activation of HIF-1 pathway. Apical periodontitis（periapical lesion） is a consequence of endodontic infection and ultimately results in destruction of tooth-supporting tissue, including alveolar bone. Thus far, the role of HIF-1 in periapical lesions has not been systematically examined. In the present study, we determined the role of HIF-1 in a wellcharacterised mouse periapical lesion model using two HIF-1α-activating strategies, dimethyloxalylglycine（DMOG） and adenovirusinduced constitutively active HIF-1α（CA-HIF1 A）. Both DMOG and CA-HIF1 A attenuated periapical inflammation and tissue destruction. The attenuation in vivo was associated with downregulation of nuclear factor-κappa B（NF-κB） and osteoclastic gene expressions. These two agents also suppressed NF-κB activation and subsequent production of proinflammatory cytokines by macrophages. Furthermore, activation of HIF-1α by DMOG specifically suppressed lipopolysaccharide-stimulated macrophage differentiation into M1 cells, increasing the ratio of M2 macrophages against M1 cells. Taken together, our data indicated that activation of HIF-1 plays a protective role in the development of apical periodontitis via downregulation of NF-κB, proinflammatory cytokines, M1 macrophages and osteoclastogenesis.

In the process of coal mine drilling,controlling the rotary speed is important as it determines the efficiency and safety of *** this paper,a linear extended state observer(LESO)based backstepping controller for rotary speed is proposed,which can overcome the impact of changes in coal seam hardness on rotary ***,the influence of coal seam hardness on the drilling rig’s rotary system is considered for the first time,which is reflected in the numerical variation of load torque,and a dynamic model for the design of rotary speed controller is *** an LESO is designed to observe the load torque,and feedforward compensation is carried out to overcome the influence of coal seam *** on the model of the compensated system,a backstepping method is used to design a controller to achieve tracking control of the rotary ***,the effectiveness of the controller designed in this paper is demonstrated through simulation and field experiments,the steady-state error of the rotary speed in field is 1 r/min,and the overshoot is reduced to 5.8%.This greatly improves the stability and security,which is exactly what the drilling process requires.

A population randomization-based multi-objective genetic algorithm for gesture adaptation in human-robot interaction

In recent years, vision-based gesture adaptation has attracted great attention from many experts in the field of human-robot interaction, and many methods have been proposed and successfully applied, such as particle swarm optimization and genetic algorithm. However, the reduction of the error and energy consumption of a robot while paying attention to more subtle attitude changes is very important and *** view of these problems, we propose a population randomization-based multi-objective genetic *** gesture signal is processed with a slight change by imitating the biological evolution mechanisms. In the proposed algorithm, a random out-of-order matrix is added in the process of population evolution synthesis to prevent the premature grouping convergence of the new population. The weights of the objective function and the elite retention strategy are adopted, and the most adaptable individuals in each generation are inherited directly in the next generation without any recombination or mutation. To verify the effectiveness of the algorithm, preliminary application experiments are performed on the gesture adaptation of a robotic arm. The results are compared with the original signal, and the comparison shows that by using the proposed method, the energy consumption is reduced, and the end error is decreased to less than 3 mm while ensuring the tracking effect of the robotic arm. These obtained results meet the communication requirements for human-robot interactions such as handshakes. Moreover, the proposed method has better performance, uses less energy, and has a smaller tracking error than the particle swarm optimization, the single-objective genetic algorithm, and the traditional multi-objective genetic algorithm. A preliminary application experiment indicates that the robotic arm can adapt to human gestures in real time.

Reflection of ultraviolet （UV） light by the wings of the female Eurasian cabbage butterfly, Pieris rapae, shows a large geographic variation. The wings of the female of the European subspecies, P. rapae rapae, reflect little UV light, while butterflies of the Asian subspecies, P. rapae crucivora, may reflect it strongly or at only intermediate levels. The geographic region where P. rapae originated remains to be determined, Moreover, it is not clear if females with wings that reflect little UV light are ancestral to females with wings that reflect UV strongly or vice versa. In the present study, we aimed to determine the geographic origin and ancestral UV pattern of cabbage butterflies through mitochondrial DNA （mtDNA） sequence analysis and amplified fragment length polymorphism （AFLP） analysis. The results of these investigations suggest that P. rapae is of European origin and that it has expanded its distribution eastward to Asia. It follows that the ancestral subspecies is the type with UV-absorbing wings. Lower nucleotide diversities and haplotype network patterns ofmtDNA derived from East Asian populations suggest that population expansion from Europe to East Asia probably occurred fairly recently and at a rapid rate.

Evaluation of Reactive Oxygen Species (ROS) Generated on the Surface of Copper Using Chemiluminesence

The antibacterial activity of copper is well-known from an ancient civilization, however, its biocidal mechanism has not been necessarily elucidated. Notwithstanding up to now, mainly 4 processes have been proposed. Among them, it is cleared that 4 kinds of reactive oxygen species (ROS): hydroxyl radical ·OH, hydrogen per oxide H2O2, superoxide anion ·O-2 and singlet oxygen 1O2, play an important role for contact-killing of bacteria, viruses and fungi. In this paper, generation of ROS on the surfaces of copper plates heated from room temperature to 673 K for 4.2 × 102 s in air, was investigated using the chemiluminescence. ROS have been evaluated by selecting the most suitable scavengers, such as 2-propanol for ·OH, sodium pyruvate for H2O2, nitro blue tetrazolium for ·O-2, and sodium azide NaN3 for 1O2. At the same time the outermost surface of copper, on which thin film of cuprous oxide Cu2O was first formed and then cupric oxide CuO was laminated on Cu2O, was examined by thin-film XRD and TEM analysis to estimate the amounts and kinds of copper oxides. It was found that the most amounts of ROS were obtained for the 573 K-heated Cu plate and they were composed of ·OH, H2O2, and ·O-2..

Gemcitabine is a cytidine analogue frequently used in the treatment of various ***,the development of chemoresistance limits its *** resistance is regulated by various factors,including aberrant genetic and epigenetic controls,metabolism of gemcitabine,the microenvironment,epithelial-to-mesenchymal transition,and acquisition of cancer stem cell *** many situations,results using cell lines offer valuable lessons leading to the first steps of important *** this review,we mainly discuss the factors involved in gemcitabine metabolism in association with chemoresistance,including nucleoside transporters,deoxycytidine kinase,cytidine deaminase,and ATP-binding cassette transporters,and outline new perspectives for enhancing the efficacy of gemcitabine to overcome acquired chemoresistance.

Bacterial communities in different locations, seasons and segments of a dairy wastewater treatment system consisting of six segments

A dairy wastewater treatment system composed of the 1st segment（no aeration） equipped with a facility for the destruction of milk fat particles, four successive aerobic treatment segments with activated sludge and a final sludge settlement segment was developed. The activated sludge is circulated through the six segments by settling sediments（activated sludge） in the 6th segment and sending the sediments beck to the 1st and 2nd *** was examined using samples from the non-aerated 1st and aerated 2nd segments obtained from two farms using the same system in summer or winter. Principal component analysis showed that the change in microbiota from the 1st to 2nd segments concomitant with effective wastewater treatment is affected by the concentrations of activated sludge and organic matter（biological oxygen demand [BOD]）, and dissolved oxygen（DO） content. Microbiota from five segments（1st and four successive aerobic segments） in one location was also examined. Although the activated sludge is circulating throughout all the segments, microbiota fluctuation was observed. The observed successive changes in microbiota reflected the changes in the concentrations of organic matter and other physicochemical conditions（such as DO）, suggesting that the microbiota is flexibly changeable depending on the environmental condition in the segments. The genera Dechloromonas, Zoogloea and Leptothrix are frequently observed in this wastewater treatment system throughout the analyses of microbiota in this study.

The obvious grain refinement of the primary MnSb phase has been observed in the Mn-89.7 wt%Sb alloy directionally solidified under a high-gradient magnetic *** the application of a high-gradient magnetic field,the morphology of the primary MnSb phase transformed from developed dendritic-like to equiaxed-like,and the grain size decreased by approximately 93%.Refinement of the primary MnSb phase can be attributed to the constituent supercooling in front of the solidification interface,which promoted nucleation of the primary MnSb *** constituent supercooling can be linked to the enrichment of the Mn solute induced by the magnetic force and the Lorentz force that drove Mn solute migration and suppressed convection.
