Measurement of serum zinc improves prostate cancer detection efficiency in patients with PSA levels between 4 ng/mL and 10 ng/mL

Aim： To investigate whether the measurement of serum zinc may improve the detection of prostate cancer （PCa） in men who had total prostate-specific antigen （PSA） levels higher than 4.1 ng/mL. Methods： A mass screening for PCa of 3940 men over 50 years old was undertaken using total serum PSA. Of the 190 men （4.8 %） with elevated PSA, 143 （3.6 %） underwent a transrectal ultrasonography （TRUS）-guided biopsy of the prostate, and 42 men （1% of total and 29.3 % of men undergoing biopsy） were found to have cancer. The areas under the receiver operating characteristic curves （ROC-AUC） were used to compare the diagnostic power of cancer detection by means of serum zinc, and free PSA/total PSA ratio （fit）. Results： The men with levels of serum zinc that ranged from 40 ng/mL-60 ng/mL, had an age-adjusted odds ratios（OR） of 5.0. A cutoff value of 100 gg/mL for-serum zinc concentration provided a sensitivity of 90.5 % and a specificity of 32.7 % in elevated PSA range, and a sensitivity of 93.3 % and specificity of 27.1% in gray zone, respectively. In the gray zone ranges of 4.1 ng/mL-10.0 ng/mL, the ROC-AUC for zinc was 73.0 % higher than 62.7 % of f/t PSA ratio and 56.7 % of total PSA. Conclusion： PCa displays a lower serum zinc concentration. The measurement of zinc levels improves PCa detection in the gray zone compared with the f/t PSA ratio and total PSA. （Asian J Androl 2005 Sep; 7： 323-328）

AIM： To investigate the protective effect of rosuvastatin on ischemia-reperfusion （I-R）-induced small intestinal injury and inflammation in rats, and to determine the effect of this agent on the expression of endothelial nitric oxide synthase （eNOS） protein. METHODS： Intestinal damage was induced in male Sprague-Dawley rats by clamping both the superior mesenteric artery and the celiac trunk for 30 min, followed by reperfusion for 60 min. Rosuvastatin dissolved in physiological saline was administered intraperitoneally 60 min before ischemia. The severity of the intestinal mucosal injury and inflammation were evaluated by several biochemical markers, as well as by histological findings. The protein levels of eNOS were determined by Western blot. RESULTS： The levels of both intraluminal hemoglobin and protein, as indices of mucosal damage, were significantly increased in the I-R group compared with those in the sham-operated group. These increases, however, were significantly inhibited by treatment with rosuvastatin in a dose-dependent manner. The protective effects of rosuvastatin were also confirmed by histological findings. Exposure of the small intestine to I-R resulted in mucosal inflammation characterized by significant increases in thiobarbituric acid-reactive substances, tissueassociated myeloperoxidase activity, and the mucosal contents of rat cytokine-induced neutrophil chemoattracrant-1 （CINC-1） and tumor necrosis factor-α（TNF-α）. These increases in inflammatory parameters after I-R were significantly inhibited by pretreatment with rosuvo astatin at a dose of 10 mg/kg. Furthermore, mRNA expression of CINC-1 and TNF-α was increased after I-R, and this increase was also inhibited by rosuvastatin. The mucosal protein levels of eNOS decreased during I-R, but were preserved in rats treated with rosuvastatin. Luminescent Characteristic of Organic Compound-Containing Inorganic Crystal at Room Temperature

The luminescent functional crystal was produced by the organic guest molecules conformation control in the inorganic host crystal matrix. Inorganic host-organic guest crystals were successfully prepared and luminescence spectra were investigated. These crystals had a phosphorescent property in room temperature by 254 nm UV irradiation. In this system, the potassium sulfate host matrix was effectively inhibited the molecular vibration of organic guest molecules by interaction between K+ and π-electrons. In addition, control of the luminescence wavelength (both fluorescence and phosphorescence) was achieved by controlling the structure of organic molecules which were taken in anorganic compound-containing inorganic crystal. Specifically, theorganic compound-containing inorganic crystals with 2-aminobenzenesulfonic acid and 2-naptylamine-1-sulfonic acid as a guest had a phosphorescence band of 455 nm and 510 nm, respectively.

Tumor differentiation phenotype in gastric differentiated-type tumors and its relation to tumor invasion and genetic alterations

AIM： To clarify the relations between tumor differentiation phenotype and tumor invasion or genetic alterations in gastric differentiated-type tumors. METHODS： We examined the tumor differentiation phenotype, the presence of mutations in APC and p53, and the microsatellite instability （MSI） status in 48 gastric adenomas and 171 differentiated-type carcinomas, The tumor differentiation phenotype was determined by examining the expression of human gastric mucin （HGM）, NUC6, MUC2 and CD10, The tumors were then classified into gastric- （G-）, gastric and intestinal mixed （GI-）, or intestinal- （I-） phenotypes, according to the immunopositivity of the above markers, The presence of mutations in APC and p53 and the MSI status were also investigated in all the tumors, RESULTS： Gastric adenomas were significantly associated with CDIO expression, I-phenotype tumors and the presence of APC mutations, compared with carcinomas （66.7% vs 25.1%, P 〈 0.0001; 56.3% vs 14.6%, P 〈 0.0001; 39.6% vs 14.0%, P 〈 0.0001, respectively） and inversely associated with expressions of HGM and MUC6 and the presence of p53 mutations （10.4% vs 62.6%, P 〈 0.0001; 39.6% vs 64.3%, P = 0.003; 2.0% vs 26.3%, P = 0.001, respectively）. The frequency of APC mutations was significantly higher in HGM-negative tumors, MUC6-negative tumors, CD10-positive tumors and I-phenotype tumors than in HGM-positive tumors, MUC6- positive tumors, CD10-negative tumors and G-phenotype tumors （32.7% vs 7.1%, P 〈 0.0001; 27.8% vs 14.0%, P = 0.0182; 37.3% vs 10.4%, P 〈 0.0001; and 38.5% vs 9.5%, P = 0.0017, respectively）. The frequency of MSI was significantly higher in MUC6-positive tumors, CD10- negative tumors and G-phenotype tumors than in MUC6- negative tumors, CD10-positive tumors and I-phenotype tumors （24.8% vs 6.7%, P = 0.0009; 22.2% vs 8.0%, P = 0.0143; and 28.6% vs 9.6%, P = 0.0353, respectively）. Chronological changes in the liver after temporary partial portal venous occlusion

AIM:To investigate time-dependent changes caused by temporal portal vein obstruction and subsequent reperfusion in the lobe with or without an occluded portal ***:The portal vein(PV)of the anterior lobe of the liver of a male Wistar rat(8 wk-old)was obstructed(70%)for 12,24,36 and 48 h,respectively,and models were sacrificed at 48 h after reperfusion(each group:n=10).The histological changes and the status of liver regeneration were compared between a liver biopsy performed on each lobe after temporary obstruction of the portal vein in the same rat liver,and the liver extracted at the time of sacrifice(48 h after reperfusion).RESULTS:With regard to the obstructed lobe,the liver weight/body weight ratio significantly decreased according to obstruction *** the other hand,in thenon-obstructed lobe,there were no significant differences within each *** duration of PV occlusion did not seem to be strong enough to introduce liver weight *** of liver regeneration was brought about in the non-occluded lobe by 12-h occlusion,and was sustained even at 48 h after *** obstructed lobe atrophied with the passage of time in the obstructed ***,the proliferating-cell nuclear antigen labeling index also increased at 48 h after reperfusion,and a repair mechanism was ***:Temporary blood flow obstruction of the portal vein may become a significant trigger for liver regeneration,even with an obstruction of 12 h.

Gastric adenocarcinoma of the fundic gland(chief cell-predominant type): A review of endoscopic and clinicopathological features

Gastric adenocarcinoma of the fundic gland(chief cellpredominant type, GA-FG-CCP) is a rare variant of welldifferentiated adenocarcinoma, and has been proposed to be a novel disease entity. GA-FG-CCP originates from the gastric mucosa of the fundic gland region without chronic gastritis or intestinal metaplasia. The majority of GA-FG-CCPs exhibit either a submucosal tumor-like superficial elevated shape or a flat shape on macroscopic examination. Narrow-band imaging with endoscopic magnification may reveal a regular or an irregular microvascular pattern, depending on the degree of tumor exposure to the mucosal surface. Pathological analysis of GA-FG-CCPs is characterized by a high frequency of submucosal invasion, rare occurrences of lymphatic and venous invasion, and low-grade malignancy. Detection of diffuse positivity for pepsinogen-I by immunohistochemistry is specific for GA-FG-CCP. Careful endoscopic examination and detailed pathological evaluation are essential for early and accurate diagnosis of GA-FG-CCP. Nearly all GA-FG-CCPs are treated by endoscopic resection due to their small tumor size and low risk of recurrence or metastasis.

Gastric adenocarcinoma of fundic gland type: Five cases treated with endoscopic resection

A total of 2177 food samples collected from nine cities in northern China during 2005 to 2007 were screened for the presence of Listeria monocytogenes,a highly fatal food-borne *** isolates were subjected to serotyping,antimicrobial susceptibility,pulsed-field gel electrophoresis(PFGE) and PCR for tetracycline resistance(tet) gene,transpon Tn916,and class 1 *** pathogen occurred among 4.13%(90/2177) of samples,and the main sources were quick-frozen food(26/252,10.32%) and raw meats(46/733,6.28%).Among the 90 isolates five serotypes(1/2a,1/2b,1/2c,4b,and 3a) existed,serotype 1/2a was dominant(48.88%). Antimicrobial resistance was most frequently observed for ciprofloxacin(17.8%),tetracycline (15.6%) and streptomycin(12.2%).Overall,resistance was observed against 14 out of 18 antimicrobials tested while multiple resistance occurred among 18.9%(17/90) isolates. Interestingly,two isolates were resistant to more than five *** 14 tetracycline-resistant isolates,13 carried tet(M) gene including nine possessing Tn916,and one harbored tet(S) *** PFGE types could be distinguished but no clear-cut affiliation with specific serotype or food category was *** widespread distribution of epidemiologically important serotypes(1/2a,1/2b and 4b),their high genetic diversity and resistance to commonly used antibiotics indicate a potential public health risk.

跨肌浆网膜的钙离子ATP酶SERCA1a可在水解ATP的同时逆浓度梯度从胞浆转运钙离子入肌浆网,引发收缩的骨骼肌细胞舒张.目前对SERCA1a结构与功能的研究,要领先于对其他P型离子转运ATP酶的研究.为了解SERCA1a跨膜螺旋M7与M8膜内侧连接部Linker78(L78)的功能,我们评估了L78部分氨基酸残基突变对SERCA1a反应循环的影响,发现:除G864A之外的所有突变体均可不同程度地降低钙离子转运速率;G862或P863氨基酸残基的突变导致SERCA1a的ATP酶活性显著降低或丧失,并引起由ATP或无机磷酸Pi生成的磷酸化酶中间体EP量显著减少;A893被P取代对SERCA1a的影响与G862、P863突变相似;与野生型SERCA1a相比,G864A与FMQ873-875单突变体ATP酶活性及EP生成量无明显差别.上述结果表明,M7、M8膜内侧连接部L78不仅与跨膜区钙离子转运过程密切相关,而且L78在GPG862-864处及A893附近的正确转折对胞浆结构域P的磷酸化也具有关键的远距离调控作用,提示L78的结构及柔度对SERCA1a构象正常的周期性变化至关重要.
