Ultrastructural changes in hepatocytes after taurinetreatment in CCl_4 induced liver injury

AIM: To search the organelle based changes in hepatocytes after taurine treatment in experimental liver fi brosis induced by CCl_4 administration. METHodS: Thirty rats were divided into two groups. Group 1 (n = 15) was injected with CCl_4 plus taurine and Group 2 (n = 15) with CCl_4 plus saline for 12 wk. At the end of 12th wk, mitochondria, rough and smooth endoplasmic reticulum, and nuclei of hepatocytes were evaluated using a scoring system. The results were compared with histopathological f indings, as well. RESULTS: Taurine treatment reduced fi brosis scores signifi cantly as compared to placebo. Organelle injury scores decreased signifi cantly with taurine treatment. Ultrastructural and histopathological scores in both groups were in strong correlation (r = 0.931 for CCl_4 plus taurine and r = 0.899 for CCl_4 plus saline group). CONCLUSION: Organelle based transmission electron microscopy fi ndings can reflect successfully histological results as well as tissue healing in hepatocytes from hepatotoxin-induced liver fi brosis.

AIM: To examine the ultrastructural changes after ursodeoxycholic acid (UDCA) treatment in hepatocytes from experimentally induced fibrotic ***: Liver fibrosis was induced in male Sprague-Dawley rats with CCl4 for 12 wk, and the rats were divided into two groups. Group I was treated with saline and group Ⅱ with UDCA (25 mg/kg per day) for 4 wk. All the rats were killed at wk 16. Mitochondria, nuclei, rough endoplasmic reticulum (RER) and smooth endoplasmic reticulum (SER) of hepatocytes were evaluated according to a scoring ***: Mitochondria, nuclei, RER and SER injury scores in group Ⅱ were significantly lower than those in groupⅠ(P < 0.001). CONCLUSION: UDCA alleviates hepatocyte organelle injury in CCl4-induced liver fibrosis.

The circadian clock temporally coordinates plant growth and metabolism in close synchronization with the diurnal and seasonal environmental changes. Research over the last decade has identified a number of clock components and a variety of regulatory mechanisms responsible for the rhythmic oscillations in metabolic and physiological activities. At the core of the clock, transcriptional/translational feedback loops modulate the expression of a significant proportion of the genome. In this article, we briefly describe some of the very recent advances that have improved our understanding of clock organization and function in Arabidopsis thaliana. The new studies illustrate the role of clock protein complex for- mation on circadian gating of plant growth and identify alternative splicing as a new regulatory mechanism for clock function. Examination of key clock properties such as temperature compensation has also opened new avenues for func- tional research within the plant clockwork. The emerging connections between the circadian clock and metabolism, hor- mone signaling and response to biotic and abiotic stress also add new layers of complexity to the clock network and underscore the significance of the circadian clock regulating the daily life of plants.

Extracorporeal liver support in severe alcoholic hepatitis

The severity of alcoholic hepatitis(AH) which may coexist with cirrhosis varies greatly, from asymptomatic forms which are detected in alcoholic patients without any sign of liver disease, except laboratory abnormalities, to severe forms characterised by deep jaundice, ascites, hepatic encephalopathy and low prothrombin index. In hospitalized patients the mortality could be as high as 75%. The elevated number of therapeutic proposals reported for more than forty years reveals the lack of efficacy of a particular modality. Even in the most favorable trials, the survival is already very poor and in some cases related to the development of renal failure or hepatorenal syndrome. There are some motivating reports concerning albumin dialysis as a support treatment in patients with severe AH, either alone or in combination with other pharmacological therapies. The favorable effects of albumin dialysis in patients with severe AH suggest that the procedure used alone or in combination with other therapies may have a role in this clinical condition. This will be particularly relevant to offer an alternative therapy in these patients, thus being a potential bridge to recovery or to be listed for liver transplantation.

Given a compact symmetric space, M, we obtain the mean exit time function from a principal orbit, for a Brownian particle starting and moving in a generalized ball whose boundary is the principal orbit. We also obtain the mean exit time flmction of a tube of radius r around special totally geodesic submanifolds P of M. Finally we give a comparison result for the mean exit time function of tubes around submanifolds in Riemannian manifolds, using these totally geodesic submanifolds in compact symmetric spaces as a model.

Novel elastomer biocomposites based on straw fibers (raw or chemically modified) as reinforcing elements ofnatural rubber (NR) were reported and studied. Oat straw fibres with different average lengths were *** materials were incorporated into the elastomer, before and after chemical surface modificationinvolving sodium hydroxide. Fourier transform infrared spectroscopy (FTIR) and microscopy techniques wereemployed for characterization of fillers. The kinetics of rubber mixtures, as well as rheometric properties ofcompounds were determined. The cross-linking density was executed on the basis of equilibrium solventswellingmeasurements applying the modified Flory–Rehner equation. The morphology of biocompositessamples was analyzed by a scanning electron microscopy. Mechanical characteristic: the values of hardness,tensile strength and attenuation coefficient were also determined. The physical and chemical investigations haveproved the reinforcing effect of treated oat straw on natural rubber vulcanizates.

AIM:To evaluate ursodeoxycholic acid (UDCA) therapy on the in vitro contraction of gallbladder smooth muscle strips from cholesterol gallstone patients. METHodS:The contraction forces of gallbladder smooth muscle strips from 28 patients with cholesterol gallstones treated with UDCA were compared with contraction forces from 14 untreated patients. The strips were stimulated with increasing concentrations of cholecystokinin-8 (CCK-8). RESULTS:Although the contraction forces that developed in response to CCK-8 were higher in strips from specimens of UDCA treated patients compared to untreated patients,longer treatment periods (6-wk) caused more contraction responses than the short treatment period of 3-wk (F = 19.297,1.85 ± 0.22 g vs 1.70 ± 0.10 g,P ods may cause more effective contractions in the gallbladder,and thereby increase the rate of response to treatment.
