DNA extraction method selection for agricultural soil using TOPSIS multiple criteria decision-making model

There is an increased interest in the extraction of nucleic acids from various environmental samples since culture-independent molecular techniques contribute to deepen and broaden the understanding of a greater portion of uncultivable microorganisms. Due to difficulties to select the optimum DNA extraction method in view of downstream molecular analyses, this article presents a straightforward mathematical framework for comparing some of the most commonly used methods. Four commercial DNA extraction kits and two physical-chemical methods (bead-beating and freeze-thaw) were compared for the extraction of DNA under several quantitative DNA analysis criteria: yield of extraction, purity of extracted DNA (A260/280 and A260/230 ratios), degradation degree of DNA, easiness of PCR amplification, duration of extraction, and cost per extraction. From a practical point of view, it is unlikely that a single DNA extraction strategy can be optimum for all selected criteria. Hence, a systematic Technique for Order Preference by Similarity to Ideal Solution (TOPSIS) was employed to compare the methods. The PowerSoil? DNA Isolation Kit was systematically defined as the best performing method for extracting DNA from soil samples. More specifically, for soil:manure and soil:manure:biochar mixtures, the PowerSoil?DNA Isolation Kit method performed best, while for neat soil samples its alternative version gained the first rank.

Mandibular cementoblastoma:Case report

Cementoblastoma is a rare benign lesion that represents less than 1% of all odontogenic tumours. It’s characterized by proliferation of cementum-like tissue and in almost all cases tends to be associated with an erupted permanent tooth, most often the first mo- lar. We present an unusual case of a large cemento-blastoma that affected the right mandibular body, extending from the first premolar to the second molar, of a 19-years-old male. In this case an initial surgery was attempted under local anaesthesia, resulting in incomplete tumor removal. A second surgical procedure was performed under general anaesthesia, ensuring the complete excision of the lesion. The patient was monitored for 1 year after surgery and did not show any signs of recurrence.

It seems that defects on micro-minerals levels have an etiologic role involved in type 1 diabetes mellitus pathogenicity. The aim of our study were to evaluate the serum levels of zinc and iron and investigate their possible relationship between these biochemical parameters with demographic conditions and glycemic control in patients with type 1 diabetes mellitus disorder. In this case-control based study, 68 female with type 1 diabetes mellitus with a mean age of 52.2 ±2 as case group and 122 healthy women as a control group with a mean age of 49/4 ±3/2 were investigated .for biochemical analysis ,10 mL fasting venous blood sample were obtained from each subjects. FBS （fasting blood glucose）, fructosamine （glycemic control parameter） were determined （spectrophotometry method, （pars azmoon, Iran）, nitroblue tetrazoline method respectively）.serum zinc level with colorimetric method （Biorex-UK） and serum iron with photometric method （pars azmoon, Iran） were determined. Statistical analysis using SPSS software performed. Significant levels considered as P 〈 0.05. According to this study there is statistically significant difference between serum levels of iron and zinc in patients with type 1 diabetes compared to controls .indeed serum level of iron and zinc had lower level in patient group toward controls. In patients group, there are a positive correlation between age and decreased level of serum zinc （P 〈 0.05）. Also there was a significant negative correlation between the glycemic control status and serum zinc. Other studied parameters concluded BMI （body mass index）, Weight and height have not significant difference between groups. The decrease in serum iron and zinc level in women with type 1 diabetes may be related to low dietary intake or increased excretion of micro minerals or the presence of confounding factors that require more extensive intervention studies to confirm it.

Evaluation of the prognostic value of liver stiffness in patients with hepatitis C virus treated with triple or dual antiviral therapy:A prospective pilot study

AIM:To evaluate the association between liver stiffness(LS) prior to the initiation of dual/triple therapy and viral ***:LS was measured in all patients before treatment was *** therapeutic approach was based on hepatic,virological,and immunological evaluations and considered the fact that patients with severe fibrosis(F3)or compensated cirrhosis(F4)in Child-Pugh class A are the primary candidates for triple *** total,65 hepatitis C virus(HCV)patients were treated with Peg-interferon/ribavirin(Peg-IFN/RBV);24patients were classified as genotypes 1/4(36.92%),and41 patients were classified as genotypes 2/3(63.08%)(dual therapy).In addition,20 HCV treatment-experienced genotype 1 patients were treated with Peg IFN-RBV and boceprevir(triple therapy).Wilcoxon rank-sum tests were used to compare the ***:LS significantly differed between dual therapy and triple therapy(P=0.002).The mean LS value before dual therapy treatment was 8.61±5.79k Pa and was significantly different between patients achieving a sustained virologic response(SVR)24weeks after therapy and those who did not(7.23±5.18 k Pa vs 11.72±5.99 k Pa,respectively,P=0.0003).The relative risk of non-response to therapy was 4.45(95%CI:2.32-8.55).The attributable risk of non-response to therapy was 49%.The mean LSvalue before triple therapy treatment was 13.29±8.57k Pa and was significantly different between patients achieving and not achieving SVR24(9.41±5.05 vs19.11±9.74,respectively;P=0.008).The relative risk of non-response to therapy was 5.57%(95%CI:1.50-20.65).The attributable risk of non-response to therapy(70%)was increased compared with dual therapy ***-treatment stiffness>12 k Pa was significantly associated with non-SVR(P<0.025)in both ***:Pre-treatment liver stiffness may be useful for predicting the response to treatment in patients treated with either dual or triple anti-HCV therapy.
