Interferon-λ polymorphisms and response to pegylated interferon in Iranian hepatitis C patients

AIM: To evaluate the efficacy of pegylated interferon in Iranian chronic hepatitis C patients in relation to interferon-λ(IFNL) polymorphisms. METHODS: This study enrolled patients with chronic hepatitis C referred to the Tehran Blood Transfusion Hepatitis Clinic in 2011. Patients were included in the study if they had no concomitant hepatic illness, were negative for human immunodeficiency virus antibodies, and had no prior history of treatment with any type ofpegylated interferon. Patients were treated with 180 μg pegylated interferon alpha-2a(Pegaferon®) weekly and 800-1200 mg ribavirin daily for 24 or 48 wk depending on weight and hepatitis C virus(HCV) genotype. Blood samples were collected from patients to obtain DNA for determination of IFNL rs12979860 and rs8099917 polymorphisms. The virologic response in patients was then evaluated and compared between the different IFNL ***: A total of 152 patients with a mean age of 41.9 ± 10.0 years were included in the study, of which 141/152 were men(92.8%). The most frequent HCV genotype was type-1, infecting 93/152(61.2%) patients. Sustained virologic response(SVR) was achieved in 81.9% of patients with HCV genotype-1 and 91.1% of patients with HCV genotype-3. Treatment success was achieved in 91.2%(52/57) of patients with the IFNL rs12979860 CC genotype and 82.1%(78/95) in those with other genotypes. Similar treatment response rates were also observed in patients with rs8099917 TT(39/45; 86.7%) and non-TT(61/68; 89.7%) genotypes. Univariate analyses identified the following factors which influenced treatment response for inclusion in a multivariate analysis: age, HCV RNA level, stage of liver fibrosis, rs12979860 CC genotype, and aspartate transaminase level. A logistic regression analysis revealed that only the rs12979860 CC genotype was significantly associated with achievement of SVR(OR = 6.2; 95%CI: 1.2-31.9; P = 0.03).CONCLUSION: The rs12979860 CC genotype was associated with SVR in patients

A selective, sensitive and label-free visual assay of fructose using anti-aggregation of gold nanoparticles as a colorimetric probe

A new convenient colorimetric sensor for fructose based on anti-aggregation of citrate-capped gold nanoparticles（Au NPs） is presented. 4-Mercaptophenylboronic acid（MPBA） induces the aggregation of Au NPs, leading to a color change from red to blue. Fructose as a potent competitor has strong affinity for MPBA and a borate ester is formed between MPBA and fructose. There is an obvious color change from blue to red with increasing the concentration of fructose. The anti-aggregation effect of fructose on Au NPs was seen by the naked eye and monitored by UV–vis spectra. Our results showed that the absorbance ratio（A（519）/A（640）） was linear with fructose concentration in the range of 0.032–0.96 μmol/L（R^2= 0.996）, with a low detection limit of 0.01 μmol/L（S/N = 3）. Notably, a highly selective recognition of fructose was shown against other monosaccharide and disaccharide（glucose, mannose, galactose,lactose and saccharose）. With anti-aggregation assays higher selectivity is achievable. The results of this work provide a rapid method for evaluating the quantitative analysis of fructose in human plasma at physiologically meaningful concentrations and at neutral pH. The proposed procedure can be used as an efficient method for the precise and accurate determination of fructose.

Application of sulfuric acid immobilized on silica gel as an efficient and benign catalyst has been explored in the syn-thesis of 2,4,5-Triaryl-1H-imidazoles via condensation reaction of benzil or benzoin, aldehyde and ammonium acetate. The key advantages of this process are high yields, cost effectiveness of catalyst, easy work-up, purification of products by non-chromatographic method and the reusability of the ***2 catalyst.

Background:Exosome administration is a novel medical approach that promises excellent immunomodulatory properties without the conventional side effects of current antitumor necrosis factor drugs and *** study aimed to assess the safety and efficacy of usingmesenchymal stemcell(MSC)exosomes to treat refractory fistulas in patients with inflammatory bowel ***:MSCs were derived from the umbilical cords and their exosomes were *** patients with refractory perianal Crohn’s disease fistulas with amedian age of 35 years(range 31–47 years)were enrolled in the *** injections were administered in the operating roomto patients with refractory fistula(fistulas that are irresponsive to anti-tumor necrosis factor-αadministration within 6months).Sixmonths later,a physical examination,face-to-face interviews,andmagnetic resonance imaging were employed to evaluate the therapy responses of ***:The outcomes within 6 months after initiation of therapy showed that four patients had responded to *** patients who received exosome injections exhibited complete healing,while one reported no improvement and active discharge from the fistula *** addition,five patients(100%)reported neither systemic nor local adverse ***:Injection of exosomes extracted from MSCs demonstrates safety and a satisfactory therapeutic effect,as evidenced in this and other studies,and may play a significant role in the future treatment of gastrointestinal fistulas.
