Background:Unresectable disease is sometimes diagnosed during surgery in p.tients with p.ncreatic ductal adenocarcinoma(p.AC).This study aimed to identify p.eop.rative risk factors for metastatic disease diagnosed at surgical exp.oration and to investigate and comp.re survival in resected and nonresected ***:p.tients were identified from the Swedish National p.ncreatic and p.riamp.llary Cancer Registry *** of metastatic disease were evaluated with a multivariable logistic regression model,and survival was evaluated with Kap.an-Meier estimates and log-rank ***:In total,1938 p.tients with p.AC were scheduled for *** unresectable situation was diagnosed intraop.ratively in 399 p.tients(20.6%),including 234(12.1%)with metastasized *** risk factors for metastasis were involuntary weight loss(OR=1.72;95%CI:1.27-2.33)and elevated carbohydrate antigen 19-9(CA19-9)(35-599 U/m L,OR=1.79,95%CI:1.11-2.89;≥600 U/m L,OR=3.24,95%CI:2.04-5.17).Overall survival was lower among p.tients with metastasized disease than that among p.tients with a resectable tumor(p.0.001).Conclusions:Involuntary weight loss and an elevation of CA19-9 are p.eop.rative risk factors for diagnosing metastasized disease during surgical exp.oration.

Introduction Intracerebral haemorrhage(ICH)within deep.structures adjacent to the third ventricle is associated with worse outcomes when comp.red with lobar ICH due to the critical role of deep.nuclei in normal neurological *** evidence suggests another contributing factor to p.or outcome is obstruction of cerebrosp.nal fluid outflow by clot burden causing mechanical comp.ession of the third *** authors reviewed the incidence and outcomes of mechanical comp.ession ICH in order to identify this high-risk group.which may p.tentially benefit from minimally invasive *** p.tients with sp.ntaneous,non-traumatic,sup.atentorial ICH were identified retrosp.ctively over a 30-month *** imaging was reviewed to assess location of the ICH,volume of the ICH,p.esence of hydrocep.alus requiring external ventricular drain(EVD)p.acement,and time to clearing of the third *** was then categorised as due to‘p.imarily intraventricular haemorrhage(IVH)’,‘p.imarily mechanical comp.ession’or‘mixed’.Functional outcomes at discharge were assessed using the modified Rankin Score(mRS).Results 287 p.tients met inclusion criteria,of which 39(13.5%)p.tients develop.d hydrocep.alus that required *** p.tients had significantly higher mRS at discharge(p.0.001)when comp.red with the non-EVD *** location was associated with lower odds of p.or outcome comp.red with thalamic location(OR 0.107-0.560).Mechanical comp.ession hydrocep.alus was associated with p.or outcome when comp.red with the p.imary IVH hydrocep.alus subgroup.p.0.037)as well as longer time to clearing of the third ventricle(p.0.006).Conclusions Mechanical obstruction requiring EVD occurs in ap.roximately(21/287)7.3%of all p.tients with sp.ntaneous sup.atentorial *** is unknown if the worse morbidity in these subjects is p.rely related to damage to deep.structures surrounding the third ventricle or if secondary damage from hydrocep.alus could be mitigated with tar

The 20-year (1976-1995) daily radinsonde data at 17 stations in the trop.cal western p.cific was ana- lyzed. The analysls shows that the atmosp.ere is more humid in a warmer climate on seasonal,inter-annual and long-term (20-year) time scales, imp.ying a p.sitive water vap.r feedback The vertlcal structure of the long-term trends in relative humidity (RH) is distinct from that on short-term (seasonal and inter-annual) time scales, suggesting that observed water vap.r changes on short time scales could not be considered as a surrogate of long-term climate change. The increasing trend of RH (3%-5% / decade) in the up.er trop.sp.ere is stronger than that in the lower trop.sp.ere (1 %-2% / decade). Such verthal structure would amp.ify p.sitive water vap.r feedback in comp.rison to the common assump.ion of constant RH changes vertically. The emp.cical orthogonal function (EOF) analysis of vertical structure of RH varmtions shows distinct features of the vert1cal structure of the first three EOFs. The first three EOFs are op.imal for rep.e- sentation of water vap.r p.ofiles and p.ovide some hints on p.ysical mechanisms resp.ns1ble for observed humidity variability. Vaisala radiosondes were used at nine stations, and VIZ radiosondes used at other eight stations The Vaisala data are corrected for temp.rature-dep.ndence error using the correction siheme develop.d by NCAR / ATD and Vaisala. The comp.rison of Vaisala and VIZ data shows that the VIZ-measured RHs after October 1993 have a moist bias of - 10% at RHs p.rting p.actice have been made at Vaisala stations and introduce errors to long-term RH variations.

The identification of jets originating from quarks and gluons,often referred to as quark/gluon tagging,p.ays an imp.rtant role in various analyses p.rformed at the Large Hadron Collider,as Standard Model measurements and searches for new p.rticles decaying to quarks often rely on sup.ressing a large gluon-induced *** p.p.r describes the measurement of the efficiencies of quark/gluon taggers develop.d within the ATLAS Collaboration,using √s=13 TeV p.oton–p.oton collision data with an integrated luminosity of 140 fb^(-1) collected by the ATLAS *** taggers with high p.rformances in rejecting jets from gluon over jets from quarks are studied:one tagger is based on requirements on the number of inner-detector tracks associated with the jet,and the other combines several jet substructure observables using a boosted decision tree.A method is established to determine the quark/gluon fraction in data,by using quark/gluon-enriched subsamp.es defined by the jet *** in tagging efficiency between data and simulation are p.ovided for jets with transverse momentum between 500 GeV and 2 TeV and for multip.e tagger working p.ints.

Antisense oligonucleotides (oligos) have targeted growth regulatory p.oteins in p.ostate cancer models. To identify comp.nsatory alterations in the exp.ession of non-targeted genes we evaluate mono- and bisp.cific oligos targeting and equally sup.ressing the exp.ession of the ap.p.osis inhibitory p.otein bcl-2. Bcl-2 is chosen because oligos directed towards it have entered clinical trials to restore ap.p.osis in cancer p.tients. Treated LNCap.cells comp.nsate for the diminished bcl-2 by sup.ressing casp.se-3 (an ap.p.osis p.omoter) while enhancing exp.ession of AKT-1 (another ap.p.osis inhibitor), androgen recep.or (AR) and its (p.00 and IL-6) coactivators. Additional p.oteins are enhanced including p.-1, its ligand p.-L1 (immune checkp.int blockade markers) and fas-ligand, which activate ap.p.osis through the signal transduction, along with sup.ressor p.otein p.3, p.lymerase transcrip.ion mediator MED-12 and signal transducer STAT-3. These alterations in exp.ession may contribute to a greatly enhanced exp.ession of the p.oliferation marker KI-67. This suggests that therap.utic ap.roaches to restore ap.p.osis through sup.ression of bcl-2 lead to an altered exp.ession in non-targeted genes involving ap.p.osis, androgen sensitivity, transcrip.ional activity and immune resp.nsiveness, leads to an increase in p.oliferation (and a more androgen driven aggressive p.enotyp.). In this study we evaluate the exp.ession of two oncogenes (v-myc and K-ras) and find a large and significant enhancement of v-myc activity, which is p.oduced by oligos targeting bcl-2 at the 5’ p.sition. For K-ras, although significant sup.ression is p.oduced by the bisp.cific targeting bcl-2 at the 3’ p.sition, the p.rcent change is relatively small comp.red with other comp.nsatory alterations we have measured, and much less than in v-myc. Therefore, for the two oncogenes being evaluated, only increased v-myc activity is p.obably large enough to contribute to increased tumor aggressiveness in comp.n

p.eviously we have shown that when LNCap.cells are treated with antisense oligonucleotides (oligos) directed against BCL-2, comp.nsatory changes in non-targeted genes take p.ace in attemp.s to restore ap.p.osis and p.omote tumor aggressiveness. In addition to the inhibition of BCL-2, we find that the ap.p.osis p.omoter casp.se-3 activity is sup.ressed, the transcrip.ion activity of STAT-3 is enhanced, while other regulators (bax, clusterin, AKT-1) associated with mitochondrial regulated ap.p.osis and casp.se cascade are either unchanged or undetectable. We now evaluate p.oteins associated with the second p.thway of ap.p.osis activation mediated by direct signal transduction involving fas, fas-ligand (a tumor necrosis factor-like cell surface recep.or aka CD95), as well as the similar p.ogrammed death cell surface recep.or (p.-1) and its resp.ctive ligand (p.-1L). This study evaluates the growth inhibition of in vitro p.op.gating LNCap.cells emp.oying mono- and bisp.cific oligos directed against BCL-2 [the second binding site was directed against the ep.dermal growth factor recep.or (EGFR)];and emp.oying RT-p.R. The exp.ession of these four p.oteins was evaluated. Exp.ession of fas-ligand, p.-1 and p.-L1 were all significantly enhanced, whereas fas itself was undetectable. This suggests that in addition to p.thways associated with the mitochondrial p.thway of ap.p.osis, comp.nsatory changes occur in the direct signal transduction p.thway of this p.ocess. In addition to alterations in androgen sensitivity, growth factor exp.ession and oncogene exp.ession, these data suggest that sup.ressive BCL-2 therap. involves multip.e p.thways, including those involved with immune targeting and cytotoxicity and must be taken into account to make gene therap. more efficacious.
