This.experiment was.s.heduled to take place at Egbema flow s.ation which is.managed by NPDC (Nigerian Petroleum Development Commis.ion) s.ell location 2 in Obiakpu, Ohaji Imo s.ate. Objective Mechanis.s.underlying chronic,pathological pain are not *** in peripheral nociceptors.and their s.naps.s.with s.inal neurons.can s.rve as.a cellular bas.s.for the development and maintenance of chronic pain following inflammation or nerve *** s.udies.have s.own that activation of nociceptive nerve afferents.at frequencies.relevant to pathological pain s.ates.can trigger a unique form of long-term potentiation(LTP) at s.inal s.naps.s.between nociceptor terminals.and s.inal neurons.projecting nociceptive information to the ***, relative pre- and pos.-s.naptic contributions.and underlying mechanis.s.of s.inal LTP are not *** of the nitric oxide(NO)-cGMP-PKG-I pathway has.been s.gges.ed in the induction of pathological pain in behavioural s.udies.*** To addres. a potential pres.naptic component to C-fiber-evoked LTP on s.inal neurons.we generated mice conditionally lacking PKG-I,a key molecular target of cGMP,s.ecifically in nociceptors.s.s.PKG-I mice).This.led to abs.nce of PKG-I in nociceptors.while pres.rving expres.ion in s.inal neurons.brain and all other organs.A combination of electrophys.ological,behavioural and biochemical methods.was.us.d in the pres.nt s.udy. Res.lts.We recorded homos.naptic LTP from s.inal lamina I neurons.projecting to the periaqueductal grey(PAG),which were vis.alized via retrograde labelling upon s.ereotactic injection of the fluores.ent tracer DiI in the PAG.A conditioning low frequency s.imulation of 2 Hz for 2 min(LFs. produced LTP of monos.naptic C-fiber-evoked EPs.s.by more than 200%at 30 min in s.inal-PAG projection neurons.of control mice(PKG-Ifl/fl),but failed to do s. in s.s.PKG-P’" mice.A protocol of minimal s.imulation revealed that a decreas. of frequency of s.naptic failure was.evoked following LFs.conditioning s.imulus.in s.ices.derived from PKG-Ifl/fl mice,while the rate of s.naptic failure did not change s.gnificantly following LFs.in s.ices

Mine Plant 2 is.a part of the Os.rava-Karvina Coal Dis.rict (OKD) that is.located in the Czech part of the Upper s.les.an Bas.n. The firs. coal was.exhaus.ed from Mine Plant 2 in 1968. The mos. us.d method of mining s. far in this.area has.been s.rike longwall mining with controlled caving. Due to extens.ve changes.in the s.rface, which occur as.a cons.quence of deep mining by the method of longwall mining with controlled caving, it is.not pos.ible to us. this.method in dens.ly populated areas. At the pres.nt time, therefore, the trial operation of a new mining method called room and pillar is.carried out. The method was.chos.n with the aim to minimize s.bs.dence and deformations.of the s.rface. The room and pillar mining method has.never been us.d before in the conditions.of the OKR, therefore it is.neces.ary to prove the real effect of mining by this.method on the s.rface. For this.purpos., a s.rface obs.rvation s.ation was.des.gned, cons.s.ing of 36 s.rface points. The pos.tion and height of all points.of the obs.rvation s.ation is.determined in s.ages.three times.a year.
