Comprehensive Transcriptome Analysis of Auxin Responses in Arabidopsis

In plants, the hormone auxin shapes gene expression to regulate growth and development. Despite the detailed characterization of auxin-inducible genes, a comprehensive overview of the temporal and spatial dynamics of auxinregulated gene expression is lacking. Here, we analyze transcriptome data from many publicly available Arabidopsis profiling experiments and assess tissue-specific gene expression both in response to auxin concentration and exposure time and in relation to other plant growth regulators. Our analysis shows that the primary response to auxin over a wide range of auxin application conditions and in specific tissues comprises almost exclusively the up-regulation of genes and iden- tifies the most robust auxin marker genes. Tissue-specific auxin responses correlate with differential expression of Aux/ IAA genes and the subsequent regulation of context- and sequence-specific patterns of gene expression. Changes in transcript levels were consistent with a distinct sequence of conjugation, increased transport capacity and down-regulation of biosynthesis in the temperance of high cellular auxin concentrations. Our data show that auxin regulates genes associated with the biosynthesis, catabolism and signaling pathways of other phytohormones. We present a transcriptional overview of the auxin response. Specific interactions between auxin and other phytohormones are highlighted, particularly the regulation of their metabolism. Our analysis provides a roadmap for auxin-dependent processes that underpins the concept of an ＇auxin code＇-a tissue-specific fingerprint of gene expression that initiates specific developmental processes.

Promising new treatment targets in patients with fibrosing lung disorders

The processes of lung fibrogenesis and fibrotic healing are common to a number of conditions with different etiologies. The lungs are the only affected organ in some cases, whereas in others, several organ systems are involved. Therapeutic options can be discussed from various perspectives. In this review, we address the localization of therapeutic targets with regard to cell compartments, including secreted ligands, cell surface, plasma membrane-cytosol interplay, cytosol and nucleus. Complex approach using stem cell therapy is also discussed. As the prognosis of patients with these disorders remains grim, treatment combinations targeting different molecules within the cell should sometimes be considered. It is reasonable to assume that blocking specific pathways will more likely lead to disease stabilization, while stem cell-based treatments could potentially restore lung architecture. Gene therapy could be a candidate for preventive care in families with proven specific gene polymorphisms and documented familial lung fibrosis. Chronobiology, that takes into account effect of circadian rhythm on cell biology, has demonstrated that timed drug administration can improve treatment outcomes. However, the specificrecommendations for optimal approaches are still under debate. A multifaceted approach to interstitial lung disorders, including cooperation between those doing basic research and clinical doctors as well as tailoring research and treatment strategies toward(until now) unmet medical needs, could improve our understanding of the diseases and, above all, provide benefits for our patients.

The rheological properties in question are influenced by many factors, ranging from the characteristics of the given polymer or solvent to the flowing conditions. The primary focus of this study is to analyse the rheological behaviour of poly（vinyl butyral）—Mowital B 60 H—（PVB） solutions dissolved in methanol and a blend of these with fumed silica nanoparticles. The preparation of the nanofibrous web and the quality of nanofibres were correlated with the rheology of the polymer solution. It was discerned that drastically intensifying shear viscosity and the elasticity of the solution exerted a negligible effect on the formation of fibres, a finding which has rarely been discussed in the literature. The morphologies and structures of the PVB/silica nanofibrous membranes were investigated by scanning electron microscopy, thermogravimetric analysis and Fourier transform infrared spectroscopy.

Conodont elements are calcium phosphate(apatite structure)mineralized remains of the cephalic feeding apparatus of an extinct marine *** to the high affinity of apatite for rare earth elements(REE)and other high field strength elements(HFSE),conodont elements were frequently assumed to be a reliable archive of sea-water composition and changes that had occurred during ***,the crystallinity index of bioapatite,i.e.,the rate of crystallinity of biologically mediated apatite,should be generally linearly dependent on diagenetic alteration as the greater(and longer)the pressure and temperature to which a crystal is exposed,the greater the resulting *** this study,we detected the uptake of HFSE in conodont elements recovered from a single stratigraphic horizon in the Upper Ordovician of Normandy(France).Assuming therefore that all the specimens have undergone an identical diagenetic history,we have assessed whether conodont taxonomy(and morphology)impacts HFSE uptake and crystallinity *** found that all conodont elements are characterized by a clear diagenetic signature,with minor but significant differences among *** distinctions are evidenced also by the crystallinity index values which show positive correlations with some elements and,accordingly,with diagenesis;however,correlations with the crystallinity index strongly depend on the method adopted for its calculation.

A strategy for uncovering germline variants altering anti-tumor CD8 T cell response

Among many factors known to alter the outcomes of T cell receptor(TCR)-induced proximal signaling,the role of human germline variants in dictating the individuality of the anti-tumor CD8 T cell response has remained challenging to ***,we describe a convenient strategy for molecular and functional characterization of phosphotyrosine-altering non-synonymous single nucleotide variations(pTyr-SNVs)that directly impact TCR-induced proximal phosphotyrosine motif-based signaling *** devise an experimental co-cultivation set-up comprising a C57BL/6 mouse-derived metastatic melanoma cell line engineered to constitutively present ovalbumin(OVA)antigens and retrovirally engineered syngeneic major histocompatibility complex(MHC)Class I restricted OVA TCR-transgenic CD8 T cells(OT-I).Using the synthetic version of pTyr-SNV rs1178800678-G/T-encoding integrin alpha 4(ITGA4)p.S1027I variant as a prototype,we show that under identical TCR stimulation conditions,genetically determined membrane-proximal immunoreceptor tyrosin activation motif(ITAM)results in increased tyrosine phosphorylation of 70 kDa zeta-chain-associated protein(ZAP70)and the levels of cytotoxic effector molecule granzyme B(GZMB),which in turn result in enhanced cytotoxic activity against metastatic melanoma cell *** strategy paves the way for rapid molecular and functional characterization of anti-tumor immune response-linked germline pTyr-SNVs so as to improve our understanding of the genetic basis of individual-to-individual differences in anti-tumor CD8 T cell response.
