Alternative polyadenylation events in epithelial cells sense endometritis progression in dairy cows

Endometritis(inflammation of the endometrial lining) is one of the most devastating reproductive diseases in dairy cattle, resulting in substantial production loss and causing more than $650 million in lost revenue annually in the *** hypothesize that alternative polyadenylation(APA) sites serve as decisive sensors for endometrium health and disease in dairy cows. Endometrial cells collected from 18 cows with purulent vaginal discharge scored 0 to 2 were used for APA profiling with our whole transcriptome termini site sequencing(WTTS-seq) method. Overall, pathogens trigger hosts to use more differentially expressed APA(DE-APA), more intronic DE-APA, more DE-APA sites per gene and more DE-genes associated with inflammation. Host CD59 molecule(CD59), Fc fragment of IgG receptor IIa(FCGR2A), lymphocyte antigen 75(LY75) and plasminogen(PLG) may serve as initial contacts or combats with pathogens on cell surface, followed by activation of nuclear receptor subfamily 1 group H member 4(NR1H4) to regulate AXL receptor tyrosine kinase(AXL), FGR proto-oncogene, Src family tyrosine kinase(FGR), HCK protooncogene, Src family tyrosine kinase(HCK) and integrin subunit beta 2(ITGB2) for anti-inflammation. This study is the first to show significance of cilium pathways in endometrium health and animal reproduction. MIR21 and MIR30A would be perfect antagonistic biomarkers for diagnosis of either inflammation or anti-inflammation. These novel findings will set precedent for future genomic studies to aid the dairy industry develop new strategies to reduce endometritis incidence and improve fertility.

Thermal analysis of olive tree pruning and the by-products obtained by its gasification and pyrolysis: The effect of some heavy metals on their devolatilization behavior

In this work, the effect of the presence of nickel and lead in thermal decomposition of olive tree pruning (OTP), OTP-char and OTP-ashes was studied by thermogravimetry. Experiments were conducted in two kinds of atmosphere (nitrogen atmosphere and oxidizing atmosphere with 20% of O2) at a heating rate of 10 ℃/min. This investigation describes the chemical, physical and fuel properties of the OTP, which shows a similar composition to other lignocellulosic materials such as hazelnut husk and wood sawdust. In addition, SEM analysis indicated that OTP-char surface is higher than OTP surface with plenty of holes and channels. It makes the char an ideal support for metal retention specially for Pb metal (OTP retained 8.55 mg/g whereas OTP char retained 11.57 mg/g). On the other hand, metal retention occurred by adsorption or ion exchange, according to the IR spectrum of the samples. The results of thermogravimetric tests proved that the presence of lead did not have a strong effect on the decomposition of the samples, since TG and DTG curves were very similar. However, nickel increased the mass loss rate, accelerating the decomposition process, showing higher peaks in DTG curves. Additionally, for temperatures higher than 360 ℃, the volatilization of the samples was improved in the nickel-polluted sample, achieving a higher mass loss, getting more energy from the biomass and reducing the quantity of residues left after the process.

Interacting Light Paths Attract KELEA (Kinetic Energy Limiting Electrostatic Attraction) and Can Lead to the Activation of Water

Water can acquire a kinetic activity, which is attributed to the absorption of an environmental force termed KELEA (kinetic energy limiting electrostatic attraction). This activity can lead to increased volatility of the water that can be measured as the progressive weight loss in closed but not completely sealed containers. It has been proposed that KELEA is a natural force required to prevent the fusion and possible annihilation of electrostatically attracted opposite electrical charges. As such, it may be especially available where there is a convergence of force fields of opposing electrical charges. At least conceptually, this may arise with facing light sources with centrally directed light beams. An oscillating attraction and release of KELEA may be facilitated by repetitively disrupting the light paths using an overhead flashing light source. This paper reports on preliminary studies that are based on this premise. Although not proving the premise, the reported experiments do indicate a simple method for activating water. Moreover, the described procedure should allow for further exploration of the underlying mechanism of water activation. The procedure involves the use of four, diagonally placed regular LED traffic lights, with an overhead strobe light. The volatility of water samples in closed but not completely sealed glass vials placed within the lighted area significantly increases in a manner that persists well beyond the periods of light exposure. The paper further shows that activated water can indirectly lead to the activation of nearby water. The reported observations are of both practical and theoretical importance.

Electromagnetic Radiation Causes Weight loss and Weight Destabilization of Objects with Presumed Elevated Levels of KELEA (Kinetic Energy Limiting Electrostatic Attraction), Relevance to Human Health and to Global Warming

A natural force has been proposed, which is required to prevent the fusion and disappearance of the discrete electrical charges that are present on electrostatically attached opposing electrical charges. This force may also explain the repulsion between objects with either matching positive or negative electrical charges. The energy of this force is referred to as KELEA (kinetic energy limiting electrostatic attraction). KELEA is especially attracted to dipolar compounds and to other materials with spatially separated opposite electrical charges. These compounds can be used to increase the level of KELEA in water. KELEA activated water can become an added source of KELEA for objects that are placed in close proximity to the water. It is generally held that the weight of an object is solely determined by its mass in relation to that of the earth. Yet, it was previously reported that the measured weight of certain KELEA attracting objects can undergo considerable variability over time. This observation is consistent with the concept that KELEA can contribute to the measured weight of certain objects. The present study strengthens this concept by demonstrating that the weight of cellulose containing materials, including paper, cotton fabrics, and wood, is increased if the materials are placed close to containers of KELEA activated water. It is further shown that electromagnetic radiation can significantly reduce the added weight of the KELEA exposed cellulose containing materials. Moreover, the previously added weight of the materials can be regained by replacing the materials back into the KELEA enhanced environment. It is proposed that the electrical charges that accompany electromagnetic radiation are able to competitively withdraw some of the KELEA from certain KELEA-enhanced objects. Although vascular complications(VCs) following orthotopic liver transplantation(OLT) seldom occur, they are the most feared complications with a high incidence of both graft loss and mortality, as they compromise the blood flow of the transplant(either inflow or outflow). Diagnosis and therapeutic management of VCs constitute a major challenge in terms of increasing the success rate of liver transplantation. While surgical treatment used to be considered the first choice for management, advances in endovascular intervention have increased to make this a viable therapeutic option. Considering VC as a rare but a major and dreadful issue in OLT history, and in view of the continuing and rapid progress in recent years, an update on these uncommon conditions seemed necessary. In this sense, this review comprehensively discusses the important features(epidemiological, clinical, paraclinical, prognostic and therapeutic) of VCs following OLT.

Jejunitis and brown bowel syndrome with multifocal carcinogenesis of the small bowel

This is the first report describing a case where prolonged,severe malabsorption from brown bowel syndrome progressed to multifocally spread small bowel adenocarcinoma. This case involves a female patient who was initially diagnosed with chronic jejunitis associated with primary diffuse lymphangiectasia at the age of 26 years. The course of the disease was clinically,endoscopically,and histologically followed for 21 years until her death at the age 47 due to multifocal,metastasizing adenocarcinoma of the small bowel. Multiple lipofuscin deposits(so-called brown bowel syndrome) and severe jejunitis were observed microscopically,and sections of the small bowel showed dense lymphoplasmacytic infiltration of the lamina propria as well as blocked lymphatic vessels. After several decades,multifocal nests of adenocarcinoma cells and extensive,flat,neoplastic mucosal proliferations were found only in the small bowel,along with a loss of the mismatch repair protein MLH1 as a long-term consequence of chronic jejunitis with malabsorption. No evidence was found for hereditary nonpolyposis colon carcinoma syndrome. This article demonstrates for the first time multifocal carcinogenesis in the small bowel in a malabsorption syndrome in an enteritis-dysplasia-carcinoma sequence.

The heterodimeric structure of heterogeneous nuclear ribonucleoprotein C1/C2 dictates 1,25-dihydroxyvitamin D-directed transcriptional events in osteoblasts

Heterogeneous nuclear ribonucleoprotein （hnRNP） C plays a key role in RNA processing but also exerts a dominant negative effect on responses to 1,25-dihydroxyvitamin D （1,25（OH）2D） by functioning as a vitamin D response element-binding protein （VDRE-BP）. hnRNPC acts a tetramer of hnRNPC1 （huC1） and hnRNPC2 （huC2）, and organization of these subunits is critical to in vivo nucleic acid-binding. Overexpression of either huC1 or huC2 in human osteoblasts is sufficient to confer VDRE-BP suppression of 1,25（OH）2D-mediated transcription. However, huC1 or huC2 alone did not suppress 1,25（OH）2D-induced transcription in mouse osteoblastic cells. By contrast, overexpression of huC1 and huC2 in combination or transfection with a bone-specific polycistronic vector using a ＂self-cleaving＂ 2A peptide to co-express huC1/C2 suppressed 1,25D-mediated induction of osteoblast target gene expression. Structural diversity of hnRNPC between human/NWPs and mouse/rat/rabbit/dog was investigated by analysis of sequence variations within the hnRNP CLZ domain. The predicted loss of distal helical function in hnRNPC from lower species provides an explanation for the altered interaction between huC1/C2 and their mouse counterparts. These data provide new evidence of a role for hnRNPC1/C2 in 1,25（OH）2D-driven gene expression, and further suggest that species-specific tetramerization is a crucial determinant of its actions as a regulator of VDR-directed transactivation.

Influence of bladder neck suspension stitches on early continence after radical prostatectomy： a prospective randomized study of 180 patients

Several techniques have been introduced to improve early postoperative continence. In this study, we evaluated the impact of bladder neck （vesicourethral anastomosis） suspension on the outcome of extraperitoneal endoscopic radical prostatectomy （EERPE）. In this research, a total of 180 patients underwent EERPE. Group 1 included patients who underwent nerve-sparing EERPE （nsEERPE）（n=45）, and Group 2 included patients who underwent nsEERPE with bladder neck suspension （BNS, n=45）. Groups 3 （n=45） and 4 （n=45） included patients who received EERPE and EERPE with BNS, respectively. Patients were randomly assigned to receive BNS with their nsEERPE or EERPE procedure. Perioperative parameters were recorded, and continence was evaluated by determining the number and weight of absorbent pads （pad weighing test） on the second day after catheter removal and by a questionnaire 3 months postoperatively. Two days after catheter removal, 11.1% of Group 1, 11.1% of Group 2, 4.4% of Group 3 and 8.9% of Group 4 were conti nent. The average urine loss was 80.4, 70.1, 325.0 and 291.3 g for the each of these groups, respectively. At 3 months, 76.5% of Group I and 81.3% of Group 2 were continent. The continence figures for Group 3 and 4 were 48.5% and 43.8%, respectively. Similar overall rates were observed in all groups. In conclusion, although there are controversial reports in the literature, early continence was never observed to be significantly higher in the BNS groups when compared with the non-BNS groups, regardless of the EERPE technique performed.
