A bendamustine resistance gene signature in diffuse large b-cell lymphoma and multiple myeloma

Aim:bendamustine is primarily used for treatment of indolent lymphomas but has shown efficacy in some patients with diffuse large b-cell lymphoma(DLbCL)and multiple myeloma(MM).Molecular-based patient stratification for identification of resistant patients,who will benefit from alternative treatments,is *** aim of this study was to develop a resistance gene signature(REGS)from bendamustine dose-response assays in cultures of DLbCL and MM cell lines,enabling prediction of bendamustine response in DLbCL and MM ***:bendamustine response was determined in 14 DLbCL and 11 MM cell *** baseline gene expression profiles and degree of growth inhibition after bendamustine exposure,a bendamustine REGS was developed and examined for the risk stratification potential in DLbCL(n=971)and MM(n=1,126)patients divided into prognostic ***:bendamustine resistance significantly correlated with resistance to cyclophosphamide in DLbCL and melphalan in MM cell *** bendamustine REGS showed significantly lower bendamustine resistance probabilities in DLbCL patients with GCb subtype tumors and in tumors of the differentiation dependent centrocyte and plasmablast *** MM patients,pre-bII classified tumors displayed high bendamustine resistance probabilities and the plasma cell subtype had lower bendamustine resistance probability than memory ***,tumors belonging to the 4p14,MAF,and D2 TC subclasses consistently displayed high bendamustine resistance ***:Significant differences in predicted response to bendamustine were found in molecular subtypes of DLbCL and MM,encouraging validation in prospective bendamustine-treated cohorts with available gene expression profiles and follow-up data.

Emerging insights into the function of very long chain fatty acids at cerebellar synapses

Very long chain-saturated and-polyunsaturated fatty acids(VLC-SFA and VLC-PUFA, respectively) are a functionally important class of fatty acids containing 28 carbons or more in their acyl chain. They are synthesized by the elongation of very long fatty acids-4(ELOVL4) enzyme, expressed mainly in the brain, retina, skin, testes, and meibomian gland, where these fatty acids are found(Agbaga et al., 2008). Further, these organs exhibit tissuespecific VLC-PUFA and VLC-SFA biosynthesis and incorporation into complex lipids for specific functions. In the brain, skin, and Meibomian glands, the ELOVL4 mainly makes VLC-SFA, which are incorporated into complex sphingolipids. In the retina, the ELOVL4 makes VLC-PUFA that are incorporated into phosphatidylcholine, that are critical for visual function, while in testes and sperm, the VLC-PUFA are incorporated into sphingolipids that are critical for fertility(Yeboah et al., 2021).

The development of low-temperature solid oxide fuel cells(LT-SOFCs)is of significant importance for realizing the widespread application of *** has stimulated a substantial materials research effort in developing high oxide-ion conductivity in the electrolyte layer of *** this context,for the first time,a dielectric material,CaCu_(3)Ti_(4)O_(12)(CCTO)is designed for LT-SOFCs electrolyte application in this *** individual CCTO and its heterostructure materials with a p-type Ni_(0.8)Co_(0.15)Al_(0.05)LiO_(2−δ)(NCAL)semiconductor are evaluated as alternative electrolytes in LT-SOFC at 450–550℃.The single cell with the individual CCTO electrolyte exhibits a power output of approximately 263 mW cm^(-2) and an open-circuit voltage(OCV)of 0.95 V at 550℃,while the cell with the CCTO–NCAL heterostructure electrolyte capably delivers an improved power output of approximately 605 mW cm^(-2) along with a higher OCV over 1.0 V,which indicates the introduction of high hole-conducting NCAL into the CCTO could enhance the cell performance rather than inducing any potential short-circuiting *** is found that these promising outcomes are due to the interplay of the dielectric material,its structure,and overall properties that led to improve electrochemical mechanism in CCTO–***,density functional theory calculations provide the detailed information about the electronic and structural properties of the CCTO and NCAL and their heterostructure CCTO–*** study thus provides a new approach for developing new advanced electrolytes for LT-SOFCs.

Conditioned duality between supercontinental’assembly’and’breakup’LIPs

A compilation of 178 more precise ages on 10 potential Large Igneous Provinces(LIPs)across southern Africa,is compared to Earth’s supercontinental cycles,where 5 more prominent LIP-events all formed during the assembly of supercontinents,rather than during *** temporal bias is confirmed by a focused review of field relationships,where these syn-assembly LIPs formed behind active continental arcs;whereas,the remaining postassembly-and likely breakup-related-LIPs never share such *** the possibility of two radically different LIP-types,only the two younger breakup events(the Karoo LIP and Gannakouriep Suite)produced basalts with more enriched asthenospheric OIb-signatures;whereas,all assembly LIPs produced basalts with stronger lithospheric,as well as more or less primitive asthenospheric,signatures.A counterintuitive observation of Precambrian breakup LIPs outcropping as smaller fragments that are more peripherally located along craton margins,compared to assembly LIPs as well as the Phanerozoic Karoo breakup LIP,is explained by different preservation potentials during subsequent supercontinental ***,further accentuating radical differences between(1)breakup LIPs,preferentially intruding along what evolves to become volcanic rifted margins that are more susceptible to deformation within subsequent orogens,and(2)assembly LIPs,typically emplaced along backarc rifts within more protected cratonic interiors.A conditioned duality is proposed,where assembly LIPs are primarily sustained by thermal blanketing(as well as local arc hydration and rifting)below assembling supercontinents and breakup LIPs more typically form above impinging mantle *** a duality is further related to an overall dynamic Earth model whereby predominantly supercontinent-orientated ocean lithospheric subduction establishes/revitalizes large low shear velocity provinces(LLSVPs)during assembly LIP-activity,and heating of such LLSVPs by the Earth’s core subsequ

The benefits of regular physical activity are well ***,few studies have examined the effectiveness of integrating physical activity(PA)into curricula within a post-secondary *** investigate the incorporation of PA into medical curriculum,we developed a series of optional exercise-based review sessions designed to reinforce musculoskeletal(MSK)anatomy course *** synchronous sessions were co-taught by a group fitness instructor and an anatomy *** fitness instructor would lead students through both strength and yoga style exercises,while the anatomy instructor asked questions about relevant anatomical structures related to course material previously *** the sessions,participants were asked to evaluate the classes on their self-reported exam preparedness in improving MSK anatomy knowledge,PA levels,and mental *** participants completed surveys;a majority agreed that the classes increased understanding of MSK concepts(90.0%)and activity levels(97.7%).Many(70.0%)felt that the classes helped reduce *** majority of respondents(90.0%)agreed that the classes contributed to increased feelings of social ***,medical students saw benefit in PA based interventions to supplement MSK course *** with increasing activity levels and promoting health behaviours,integrating PA into medical curriculum may improve comprehension of learning material,alleviate stress and foster social connectivity among medical students.

Prediction of the Resonance Characteristics of Combustion Chambers on the basis of Large-Eddy Simulation

In the last few years intensive experimental investigations were performed at the University of Karlsruhe to develop an analytical model for the Helmholtz resonator-type combustion system. In the present work the resonance characteristics of a Helmholtz resonator-type combustion chamber were investigated using large-eddy simulations (LES), to understand better the flow effects in the chamber and to localize the dissipation. In this paper the results of the LES are presented, which show good agreement with the experiments. The comparison of the LES study with the experiments sheds light on the significant role of the wall roughness in the exhaust gas pipe.

Fat deposition and partitioning for meat production in cattle and sheep

In markets for beef and sheep meat, an appropriate level of intramuscular fat(IMF) is highly desirable for meat-eating quality, but strategies to improve it usually lead to an undesirable excess in carcase fat,presenting a major challenge to livestock producers. To solve this problem, we need to understand the partitioning of fat among the major fat depots: IMF, subcutaneous fat(SCF) and visceral fat(VF). In most genotypes of cattle and sheep, the rate of accretion is lower for IMF than for SCF and VF, so genetic selection for a high level of IMF, or the use of an increased dietary energy supply to promote IMF deposition, will increase overall fatness and feed costs. On the other hand, feeding postnatal calves with excessive concentrates promotes IMF deposition, so a nutritional strategy is feasible. With genetic strategies, several problems arise: 1) positive genetic correlations between IMF, SCF and VF differ among genotypes in both cattle and sheep; 2) genotypes appear to have specific, characteristic rates of accretion of IMF during periods of growth and fattening; 3) most breeds of cattle and sheep naturally produce meat with relatively low levels of IMF, but IMF does vary substantially among individuals and breeds so progress is possible through accurate measurement of IMF. Therefore, an essential prerequisite for selection will be knowledge of the genetic correlations and fat accretion rates for each genotype. Currently,selection for IMF is based on existing technology that directly measures IMF in the progeny or siblings, or estimates IMF in live animals. New technology is needed to permit the simultaneous measurement of SCF and IMF in the field, thus opening up the possibility of accurate selection, particularly for fat partitioning in live animals. Specifically, there would be great value in detecting individuals with an IMF advantage at an early age so the generation interval could be shortened and genetic gain accelerated. Acid-treated Fe-doped TiO2 as a high performance photocatalyst used for degradation of phenol under visible light irradiation

The photocatalytic activity of Fe-doped TiO2 nanoparticles is significantly increased by an acid-treatment process. The photocatalyst nanoparticles were prepared using sol–gel method with 0.5 mol% ratio of Fe:Ti in acidic pH of 3. The nanoparticles were structurally characterized by means of X-ray diffraction(XRD), high-resolution transmission electron microscope(HRTEM), energy-dispersive X-ray spectroscopy(EDX), X-ray photoelectron spectroscopy(XPS) and diffuse reflectance spectroscopy(DRS). It was observed that the photocatalytic activity suffered from an iron oxide contaminating layer deposited on the surface of the nanoparticles. This contamination layer was removed using an HCl acidtreatment process. The photocatalytic activity using 500 mg/L of Fe0.5-TiO2 in a 10 mg/L of phenol solution increased significantly from 33% to 57%(about 73% increase in the performance), within 90 min of reaction time under visible light irradiation. This significant improvement was achieved by removing the iron oxide contamination layer from the surface of the nanoparticles and adjusting p H to mild acidic and basic pHs.
