Novel Spectrophotometric Method of Determination of Cefadroxil in Dosage Form

A simple and new spectrophotometric method have been developed for the determination of cefadroxil in pure and in its pharmaceutical formulations. The method is based on formation of blood red coloured complex with ferric nitrate and 1,10-phenanthroline having λmax 515 nm. The method obeys Beer’s law in the concentration range of 0.22 μg/mL to 2.97 μg/mL. The method is statistically evaluated for accuracy and precision.

Co-Localization of Alpha1-Adrenoceptors and gPR55: A Novel Prostate Cancer Paradigm?

α1-adrenoceptors (α1-ARs) and “cannabinoid-like” g Protein Coupled Receptor 55 (gPR55) belong to the g-protein coupled receptor (gPCR) family and play a crucial role in regulating prostate function. Although physical and functional interactions between the cannabinoid and adrenergic systems have been reported, analysis of functional interactions between α1-AR and gPR55 in normal and neoplastic prostate has not been reported. Since gPR55 levels are high in rodent adrenal gland, we propose a function link between the adrenergic system and gPR55 receptor. Confocal Laser Scanning Microscopy (CLSM) was employed to examine the endogenous α1-AR and gPR55 expression and their co-localization, expressed as fluorescence, in vitro in human andro-gen-insensitive PC-3 and androgen-sensitive LNCaP prostatic carcinoma cell lines, using the fluo-rescent ligands—Syto 62 (nuclear stain), BODIPY FL-Prazosin (QAPB;fluorescent quinazoline α1-AR ligand) and Tocriflour (T1117;a novel fluorescent diarylpyrazole cannabinoid/gPR55 ligand). Fluorescent ligand binding in untreated PC-3 cells and LNCaP cells and spheroids showed hetero-geneous expression of both α1-ARs and gPR55. A small proportion of cells had both α1-ARs and gPR55 in relatively equal numbers indicating a degree of co-localization. Co-localization of fluo-rescent ligand binding exhibited a stronger correlation in LNCaP (0.87) as compared to PC-3 (0.63) cells. Upregulation of α1-AR was observed in PC-3 cells following chronic doxazosin incubation. Robust T1117 binding, suggestive of gPR55 upregulation, was also observed in these cells. The presence of subtype-rich cells with a degree of co-localization between α1-ARs and gPR55 indicates a possibility for dimerisation or functional interaction and a new paradigm for functional synergism in which interactions may be either between cells or involve converging intracellular signaling processes.

Incidence of Urinary Tract Infections and Its Aetiological Agents among Pregnant Women in Karnataka Region

Urinary tract infection (UTI) is the most common medical complications of pregnancy together with anaemia and hypertension and it occurs approximately in 5%-10% of all pregnancies. The aim of this study was to determine the incidence of UTI and prevalence of uropathogens among pregnant women. During the study period from December 2009 to August 2010, 417 urine samples were analyzed. UTI was diagnosed by growth of at least 105 CFU/ml of a urinary tract pathogen in a culture of a midstream urine sample. The isolated bacteria were identified by biochemical tests. The results showed that 49.4% of pregnant women have UTI. E. coli which was the most frequently isolated organism (56.79%), followed by Klebsiella sps (19.9%), Pseudomonas sps (6.3%), and Proteus sps (5.8%). Other pathogens isolated were Enterobacter (3.8%), Citrobacter (1.4%) and Enterococcus sps (0.9%). Of the variables examined, the highest prevalence rate was observed where, 53.3% of the infected women were in the age group 36 to 40 years, 54.15% were in their 3rd trimester;also the highest infection rate (70.2%) was observed in 7th month of pregnancy, concluding that old age pregnancy increased parity prone for UTI apart from individual hygiene and economical status.

An Overview of Extended Spectrum Beta Lactamases and Metallo Beta Lactamases

Antibiotic resistant β-lactamases are diverse and complex enzymes produced by most of the gram-negative bacteria that are mediated by number of plasmids. The impact of these enzymes has posed a major threat to the health sectors and has challenged the available treatment options for both community and hospital acquired infections. These include the uncomplicated most severe life-threatening infections. Moreover, with resistance to the cephalosporin drugs these MDR strains exhibit co-resistance patterns with different class of antibiotics which is a cause of concern that leads to narrow the limited treatment options. It is alarming situation since there is a steep rise in MDR—Beta lactamase pathogens mainly in Escherichia coli, Klebsiella pneumoniae, Acinetobacter baumanii and Pseudomonas aeruginosa. Currently, the clinical detection of Extended Spectrum of β-Lactamases (ESβL) and MβL producing pathogens are carried out by antibiotic sensitivity test on the guidelines of Clinical and Laboratory Standards Institute (formerly the National Committee for Clinical Laboratory Standards) since, the other methods being too expensive. The choice of antimicrobial treatment for infections should rely on the clinical data and the tests (AST) in asymptomatic and mild cases. However, this does not imply for critical infections. The last resorts of treatment for ESβL pathogens are carbapenem and nevertheless, resistances have also been reported for the same. With increasing resistance rate to the antibiotics, it’s very essential to follow the guidelines for detection, implementation of antibiotic rotation to reduce these pathogens, followed by the efficient infection control practices and strategies to avoid such outbreaks.

Novel 1,3,4-(thiadiazol-2-ylamino) methyl-5-(pyridin-4-yl)-1,3,4-oxadiazol-2-thiones: synthesis, docking and antimycobacterial testing

In the present study, a novel series of Mannich bases of 3-substituted 5-(pyridin-4-yl)-1,3, 4-oxadiazol-2-thione derivatives were synthesized. Docking study was performed to rationalize the possible interactions between the synthesized com-pounds and active site of 14DM. The test compounds were screened for antimycobacterial activity using Middlebrook 7H9 medium against M. tuberculosis H37Rv (ATCC 27294) as well as Isoniazid (INH) resistant clinical strain. Among the series 5c and 5a are found to be most potent (susceptible) while the compound 5f did not show activity against M. tuberculosis H37Rv (resistant). The SAR study reveals the importance of substitutions at para position for good activity.

Second Harmonic generation in Some Donor-Acceptor Substituted Chalcone Derivatives

A series of five chalcone derivatives with different substituents in para and meta posions have been synthesized, and single crystals were successfully grown in aceton solution by slow evaporation solution growth technique (SESgT). Single crystal X-ray studies revealed that all the crystals crystallized in noncentrosymmetric space group with their molecular dipoles perfectly aligned in a direction-favorable for large nonlinear optical effects. Kurtz powder tests revealed that all five materials have second-harmonic-generating properties with maximum efficiencies of approximately 14 times that of urea standard. UV-vis-NIR spectroscopy and thermogravimetric analyses are also presented for all of the reported materials. Among the five chalcones, high quality single crystals of 4-Methoxy-4'-chlorochalcone were grown by SESgT, and its crystalline perection were studied by using a high resolution X-ray diffractometry (HRXRD).

CdS nanocrystals as fluorescent probe for detection of dolasetron mesylate in aqueous solution:Application to biomedical analysis

A simple and straightforward method for the determination of dolasetron mesylate(DM) in aqueous solution was developed based on the fluorescence quenching of 3-Mercaptopropionic acid(MPA) capped Cd S quantum dots(QDs).The structure,morphology,and optical properties of synthesized QDs were characterized by using UV-Vis absorption spectroscopy,fluorescence spectroscopy,transmission electron microscopy(TEM) and dynamic light scattering(DLS) *** the optimum conditions,the MPA-Cd S QDs fluorescence probe offered good sensitivity and selectivity for detecting *** probe provided a highly specific selectivity and a linear detection of DM in the range of 2–40 μg/m L with detection limit(LOD) 1.512 μg/m *** common excipients did not interfere in the proposed *** fluorescence quenching mechanism of Cd S QDs is also *** developed sensor was applied to the quantification of DM in urine and human serum sample with satisfactory results.

Surgical obturator duplicating original tissue-form restores esthetics and function in oral cancer
