Estimation of visceral fat is useful for the diagnosis of significant fibrosis in patients with non-alcoholic fatty liver disease

G protein-coupled receptor kinase 2 （GRK2） is an important serine/threonine-kinase regulating different membrane receptors and intraceUular proteins. Attenuation of Drosophila Gprk2 in embryos or adult flies induced a defective differentiation of somatic muscles, loss of fibers, and a flightless phenotype. In vertebrates, GRK2 hemizygous mice contained less but more hypertrophied skeletal muscle fibers than wild-type littermates. In C2C12 myoblasts, overexpression of a GRK2 kinase-deficient mutant （K220R） caused precocious differentiation of ceUs into immature myotubes, which were wider in size and contained more fused nuclei, while GRK2 overexpression blunted differentiation. Moreover, p38MAPK and Akt pathways were activated at an earlier stage and to a greater extent in K220R-expressing cells or upon kinase downregulation, while the activation of both kinases was impaired in GRK2-overexpressing cells. The impaired differentiation and fewer fusion events promoted by enhanced GRK2 levels were recapitulated by a p38MAPK mutant, which was able to mimic the inhibitory phosphorylation of p38MAPK by GRK2, whereas the blunted differentiation observed in GRK2-expressing clones was rescued in the presence of a constitutively active upstream stimulator of the p38MAPK pathway. These results suggest that balanced GRK2 function is necessary for a timely and complete myogenic process.

Comparison of clinical outcomes between unrelated single umbilical cord blood and"ex-vivo"T-cell depleted haploidentical transplantation in children with hematological malignancies

Background Over the last two decades,umbilical cord blood(UCB)and haploidentical transplantation(HaploHSCT)have emerged as alternative sources of hematopoietic stem cell for allogeneic *** are few retrospective studies and no prospective studies comparing both types of alternative transplantation in pediatric *** We analyzed the data of 134 children with hematological malignancies who received a hematopoietic stem cell transplantation from a single umbilical cord blood(UCB)(n=42)or an"tex-vivo"T-cell depleted transplant from a haploi-dentical-related donor(HaploHSCT)(n=92)between 1996 and *** recovery was faster after HaploHSCT than the UCB transplant group(median times to neutrophil and platelet recovery:13 vs.16 days,10 vs.57 days,respectively)(PloHSCT group had a significantly early immune reconstitution based on NK and CD8+T cells compared with the UCB ***,after the first year *** had a lower number of CD4+ T and B lymphocytes compared with the UCB transplant *** cumulative incidence of TRM was 29±8%in the HaploHSCT group versus 40±5%in the UCB *** incidence was 21±7%in the HaploHSCT group and 19±8%in the UCB *** of DFS was 58±8%in the HaploHSCT group versus 40±9%in the UCB group(P=0.051).Conclusions TCD haploidentical transplant is associated with advantages in terms of engraftment and early immune reconstitution *** haploidentical transplant was associated with lower incidence of infectious and non-infectious complications,especially in the early phases of the transplant compared with UCB transplant ***,there are no advantages in transplant outcomes compared with UCB transplant.

TTie Ultisols in the Rana de Canamero area in Southwest Spain showed aluminum(Al)phytotoxicity,and the clearance of natural vegetation and decades of intensive conventional agriculture caused the deplation of soil organic matter(SOM).Therefore,we studied the long-term effects of no tillage and liming using sugar beet foam(SF)and red gypsum(RG),alone or in combination,on the restoration of Ultisols affected by acidification,Al phytotoxicity,and SOM *** measured the main soil chemical properties,soil microbial biomass,soil enzyme activities involved in carbon,nitrogen,phosphorus,and sulfur mineralization,and the emission of volatile organic compounds(VOCs).The results indicated that liming effectively neutralized the soil acidity in the long term and,in combination with no tillage,significantly increased soil microbial biomass and enzyme ***-three VOCs were detected using the proton transfer reaction-time of flight(PTR-ToF)technique,and both liming and tillage changed the VOC emission *** greatest difference in VOC emission pattern was observed between no-tilled un-amended soils and tilled lime-amended soils,suggesting the activation of different metabolic pathways within the microbial communities of soils under different *** in VOC emission patterns could be attributed to the decomposition of carbohydrates,which were also sustained by the higher enzyme activities in the lime-amended soils.

Toxins isolated from the venom of the spider Phoneutria nigriventer either inhibit or activate a vast number of targets, such as ion channels, with high selectivity and affinity. PhTx3-1 toxin is a selective blocker of IA currents of K^＋ channels, and PhTx3-3 and PhTx3-4 are calcium channel blockers of the P/Q-type calcium channels. The spider neurotoxin PhTx3-6 reversibly and non-specifically inhibits high-voltage-activated Ca^2＋ channels with varying potency （N 〉 R 〉 P/Q 〉 l） in heterologous and native systems. There has been a huge increase in the range of medical disorders involving ionic channels, and blockers of these channels have potential as therapeutic agents. The mechanism of action of each toxin family is different; thus, each needs to be evaluated for its therapeutic potential. This review will focus on peptide toxins purified from the venom of the spider Phoneutria nigriventer that are currently being evaluated in preclinical tests as possible drugs for the treatment of pain, cardiac arrhythmias and ischemia.

BACKGROUND The gut microbiota plays a key role in the maintenance of intestinal homeostasis and the development and activation of the host immune system. It has been shown that commensal bacterial species can regulate the expression of host genes. 16 S rRNA gene sequencing has shown that the microbiota in inflammatory bowel disease(IBD) is abnormal and characterized by reduced diversity. Micro RNAs(miRNAs) have been explored as biomarkers and therapeutic targets, since they are able to regulate specific genes associated with Crohn's disease(CD). Faecal samples of newonset CD patients, free of treatment, and healthy controls were collected.
