The biosynthesis of prosta.oids is involved in both physiologica. a.d pa.hologica. processes. The expression of prosta.la.din-endoperoxide syntha.e 2(PTGS2; a.so known a. COX-2) ha. been tra.itiona.ly a.socia.ed to the onset of severa. pa.hologies, from infla.ma.ion to ca.diova.cula., ga.trointestina. a.d oncologic events. For this rea.on, the sea.ch of selective PTGS2 inhibitors ha. been a.focus for thera.eutic interventions. In a.dition to the cla.sic non-steroida. a.ti-infla.ma.ory drugs, selective a.d specific PTGS2 inhibitors, termed coxibs, ha.e been genera.ed a.d widely used. PTGS2 a.tivity is less restrictive in terms of substra.e specificity tha. the homeosta.ic counterpa.t PTGS1, a.d it a.counts for the eleva.ed prosta.oid synthesis tha. a.compa.ies severa. pa.hologies. The ma.n regula.ion of PTGS2 occurs a. the tra.scription level. In a.dition to this, the sta.ility of the mRna.is finely regula.ed through the intera.tion with severa. cytopla.mic elements, ra.ging from specificmicroR nas to proteins tha. control mR na.degra.a.ion. Moreover, the protein ha. been recognized to be the substra.e for severa. post-tra.sla.iona. modifica.ions tha. a.fect both the enzyme a.tivity a.d the ta.geting for degra.a.ion via.protea.oma. a.d non-protea.oma. mecha.isms. a.ong these modifica.ions, phosphoryla.ion, glycosyla.ion a.d cova.ent modifica.ions by rea.tive lipidic intermedia.es a.d by free ra.ica.s a.socia.ed to the proinfla.ma.ory condition a.pea. to be the ma.n cha.ges. Identifica.ion of these post-tra.sla.iona. modifica.ions is releva.t to better understa.d the role of PTGS2 in severa. pa.hologies a.d to esta.lish a.correct a.a.ysis of the potentia. function of this protein in disea.es progress. Fina.ly, these modifica.ions ca. be used a. bioma.kers to esta.lish correla.ions with other pa.a.eters, including the immunomodula.ion dependent on molecula. pa.hologica. epidemiology determina.ts, which ma. provide a.better fra.e for potentia. thera.eutic interventions.
