Modelling the long-term geomorphic response to check dam failures in an alpine channel with CAESAR-Lisflood

Globally, between 1950 and 2011 nearly 80,000 debris flow fatalities occurred in densely populated regions in mountainous terrain. Mitigation of these hazards includes the construction of check dams,which limit coarse sediment transport and in the European Alps number in the 100,000s. Check dam functionality depends on periodic, costly maintenance, but maintenance is not always possible and check dams often fail. As such, there is a need to quantify the long-term(10-100 years) geomorphic response of rivers to check dam failures. Here, for the first time, a landscape evolution model(CAESARLisflood) driven by a weather generator is used to replicate check dam failures due to the lack of maintenance, check dam age, and flood occurrence. The model is applied to the Guerbe River,Switzerland, a pre-Alpine catchment containing 73 check dams that undergo simulated failure. Also presented is a novel method to calibrate CAESAR-Lisflood?s hydrological component on this ungauged catchment. Using 100-year scenarios of check dam failure, the model indicates that check dam failures can produce 8 m of channel erosion and a 322% increase in sediment yield. The model suggests that after check dam failure, channel erosion is the remobilization of deposits accumulated behind check dams, and, after a single check dam failure channel equilibrium occurs in five years, but after many check dam failures channel equilibrium may not occur until 15 years. Overall, these findings support the continued maintenance of check dams.

Y90-radioembolization via variant hepatic arteries: Is there a relevant risk for non-target embolization?

BACKGROUND The hepatic arterial anatomy is highly variable, with the two most common variants being a replaced right hepatic artery(RHA) originating from the superior mesenteric artery(SMA) and a left hepatic artery(LHA) originating from the left gastric artery(LGA). These anatomical variants could potentially increase the risk for non-target embolization during Y90-Radioembolization due to the close proximity between hepatic and enteric vessel *** To evaluate the safety of Yttrium-90radioembolization(90Y-RE) with resin microspheres in patients with a variant hepatic arterial *** In this retrospective single-center observational study, 11 patients who underwent RE with 90Y-resin microspheres via a LHA originating from the LGA,and 13 patients via a RHA originating from the SMA were included. Patient and treatment data were reviewed regarding clinical and imaging evidence of nontarget embolization of 90Y-resin microspheres to the GI tract. Positioning of the tip of the microcatheter in relationship to the last hepatoenteric side branch was retrospectively analyzed using angiographic images, cone-beam CT and preinterventional *** None of the 24 patients developed clinical symptoms indicating a potential nontarget embolization to the GI tract within the first month after 90Y-RE. On thepostinterventional 90Y-bremsstrahlung images and/or 90Y-positron emission tomographies, no evidence of extrahepatic 90Y-activity in the GI tract was noted in any of the patients. The mean distance between the tip of the microcatheter and the last enteric side branch during delivery of the 90Y microspheres was 3.2 cm(range: 1.9-5 cm) in patients with an aberrant LHA originating from a LGA. This was substantially shorter than the mean distance of 5.2 cm(range: 2.9-7.7 cm) in patients with an aberrant right hepatic originating from the ***90Y-RE via aberrant hepatic arteries appears to be safe;at least with positioning of the microcatheter tip

The imbalance between pathogenic and protective T cell subsets is a cardinal feature of autoimmune disorders such as multiple sclerosis(MS).Emerging evidence indicates that endogenous and dietary-induced changes in fatty acid metabolism have a major impact on both T cell fate and *** date,however,the molecular mechanisms that underlie the impact of fatty acid metabolism on T cell physiology and autoimmunity remain poorly ***,we report that stearoyl-CoA desaturase-1(SCD1),an enzyme essential for the desaturation of fatty acids and highly regulated by dietary factors,acts as an endogenous brake on regulatory T-cell(Treg)differentiation and augments autoimmunity in an animal model of MS in a T cell-dependent *** by RNA sequencing and lipidomics analysis,we found that the absence of Scd1 in T cells promotes the hydrolysis of triglycerides and phosphatidylcholine through adipose triglyceride lipase(ATGL).ATGL-dependent release of docosahexaenoic acid enhanced Treg differentiation by activating the nuclear receptor peroxisome proliferator-activated receptor *** findings identify fatty acid desaturation by SCD1 as an essential determinant of Treg differentiation and autoimmunity,with potentially broad implications for the development of novel therapeutic strategies and dietary interventions for autoimmune disorders such as MS.

Quantitative analysis of RNA levels from single hepatocytes in vivo: combined use of real-time RT-PCR and laser microdissection

AIM: The manner in which a cell responds to and influences its environment is ultimately determined by the genes that are expressed. To better understand cellular functions, the isolation of single cells and subsequent quantification of the expressed genes is essential. METHODS: Normal liver tissue was obtained from operation, snap-frozen in liquid nitrogen and sectioned in crystat. Individual hepatocytes were microdissected. RNA was extracted, then reverse transcribed and amplified using real-time quantitative polymerase chain reaction (PCR). RESULTS: Single hepatocytes were dissected by laser beam and catapulted to the microcentrifuge cap which was put above the slide. In this way, cells were collected, RNA was extracted, reverse transcribed to cDNA and used for analysis of RNA expression by real-time quantitative PCR. The amplification results showed that quantitation of the RNA inside the cell was compatible with the number of cells. CONCLUSION: The expression of RNA in single cells can be quantitated successfully by using laser microdissection and real-time PCR. These techniques provide an opportunity to monitor in vivo gene expression levels in single hepatocytes.

In the field of advanced driver assistance systems,researchers and developers make great efforts to reduce drivers' workload and to keep it on an ideal level *** the course of these efforts the physiological detection of workload,also in commercial vehicles,is coming more and more to the fore.A total of 44 driving situations have been identified in an exploratory survey of truck drivers which are relevant for the resulting driver *** includes various driving situations as well as driving maneuvers and traffic situations,such as high traffic density or different weather *** regard to objectively measurable parameters,these 44 situations were examined in a study with 37 professional truck drivers in the dynamic truck driving simulator of the Institute of Automotive *** addition to the changes in pupil dilation the electrodermal activity and a subjective assessment based on the Rating Scale of Mental Effort(RSME)were used in order to detect the mental *** results were validated in a field trial subsequent to this driving simulator *** the selection of the test course,particular attention was paid to a high degree of similarities between realworld and simulated roadway *** evaluation of the electrodermal activity measured in the real traffic study as an indicator for mental workload revealed the *** most demanding activity during the performed study was making phone calls,whereby the participants were confronted with a planning *** attention to an accessing car was also a high demanding task with anticipatory requirements with respect to the car driver's *** routine tasks such as crossing intersections,the evaluation of electrodermal activity does not lead to statistical differences between the investigated *** contrast,a self-report measurement of mental workload conducted in parallel shows sufficient sensitivity and therefore significant differences between th
