Necropsies and extensive histological evaluation for clinical and sub-clinical disease of approximately three hundred Portuguese Water dogs are available as part of an ongoing study to assess their state of health at the end of life. Throughout life these dogs enjoyed a variety of lifestyles and environments. Here we carry out retrospective quantitative assessments of life-time dietary input and physical activity for each dog. To do this, collagens from skull vault bone and from dentine have been analyzed for ratios of stable isotopes to determine differences in diet that individual dogs experienced during late or early life respectively. Robustness of skull bone (weight/unit of skull size) was used as a relative indicator of the amount of physical activity experienced during a dog’s lifetime. These environmental parameters were correlated with the frequency and severity of specific disease processes determined at necropsy. Both measures were shown to exert significant low-level

目前已经分离到两个在烟草去甲基烟碱形成过程中起作用的基因。阻断这两个基因可使烟草植株中的去甲基烟碱含量较低，从而使得一种在烟叶调制过程中经亚硝化形成的TSNA—N′-亚硝基去甲基烟碱（NNN）的含量降低。我们通过乙基甲烷磺酸盐（EMS）处理创造了一个深色晾烟的孟德尔遗传近等基因系突变群体。本研究中，我们报道了一个新的深色烟多突变体的发现，这一突变体能够消除主效和微效脱甲基酶的活性。第一代突变群体在大田环境下种植，第二代突变群体在大田和温室两种环境下种植。

阻断烟草中去甲基烟碱的形成可以降低一种在烟叶调制过程中经亚硝化形成的TSNA——N’-亚硝基-去甲基烟碱（NNN）的含量。在之前的研究中，我们报道了烟碱去甲基化酶基因的发现及其使用GMO（转基因生物，RNAi干扰方法）和非GMO（诱变）策略降低烟碱去甲基酶活性。本研究报道了在高代和育成品系中这种策略的有效性和稳定性，并且描述了将RNAi和突变位点成功转入不同品种的育种过程和进展。在多重试验中实验材料种植在大田环境下，包括4代RNAi转基因系和相应的R2代雄性不育系副本。

A crossing family of segments is a collection of segments each pair of which crosses. Given positive integers j and k,a(j,k) grid is the union of two pairwise-disjoint collections of segments (with j and k members, respectively) such that each segment in the first collection crosses all members of the other. Let c(k) be the least integer such that any planar set of c(k) points in general position generates a crossing family of k segments. Also let #(j,k) be the least integer such that any planar set of #(j,k) points in general position generates a (j,k)-grid. We establish here the facts 9≤c(3)≤16 and #(1,2)=8.

Introduction Fecal microbiota transplantation(FMT)is highly effective and potentially life-saving for recurrent Clostridioides difficile infection(CDI)[1].Recent pilot trials also reported the use of FMT to revert hepatic encephalopathy(HE)in patients with decompensated liver cirrhosis without infectious colitis[2,3].Intestinal microbiota-driven hyperammonemia is central to HE development and most treatments aim to reduce ammonia levels by modulating the intestinal microbiota.However,FMT might lead to increased intestinal ammonia production,as we describe here.

Objectives: To describe the communication behaviors of patients and physicians and patient par-ticipation in communication about treatment decision-making during consultation visits for local-ized prostate cancer (LPCa). Methods: This is a secondary analysis of data from 52 men enrolled in the usual care control group of a randomized trial that focused on decision-making for newly diagnosed men with LPCa. We analyzed the patient-physician communication using the transcribed audio-recordings of real-time treatment consultations and a researcher-developed coding tool, including codes for communication behaviors (information giving, seeking, and clarifying/ verifying) and contents of clinical consultations (health histories, survival/mortality, treatment options, treatment impact, and treatment preferences). After qualitative content analysis, we categorized patient participation in communication about treatment-related clinical content, including “none” (content not discussed);“low” (patient listening only);“moderate” (patient providing information or asking questions);and “high” (patient providing information and asking questions). Results: Physicians mainly provided information during treatment decision consultations and patients frequently were not active participants in communication. The participation of patients with low and moderate cancer risk typically was: 1) “moderate and high” in discussing health histories;2) “low” in discussing survival/mortality;3) “low and moderate” in discussing treatment options;4) “none and low” in discussing treatment impacts;and 5) “low” in discussing treatment preferences. Conclusions: Findings suggest opportunities for increasing patient participation in communication about treatment decision-making for LPCa during clinical consultations.

3-Arylisothiazoloquinols as potent ligands for the benzodiazepine site of GABA_(A) receptors

3-Arylisothiazolo[5,4-b]quinolin-4(9H)-ones and 3-arylisoxazolo[5,4-b]quinolin-4(9H)-ones were synthesized and assayed for affinity for the benzodiazepine binding site of the GABAA receptors. While the 3-arylisothiazoloquinolin-4-ones were found to be potent ligands, with affinities (expressed as the affinity Ki value) down to 1 nM, the 3-arylisoxazoloquinolin-4-ones are less potent. This is suggested to depend on sterical repulsive interaction of the 3-arylisoxazoloquinolin-4-ones with the receptor essential volume of the binding site, and a higher electron density at the nitrogen in the azole ring (N-2) as well as the carbonyl oxygen in the isothiazoloquinolin-4-ones enabling them to interact stronger with hydrogen bond donor sites at the binding site.

目的目前还没有治疗青年男性腹股沟疝病人的最佳修补方法的大宗病例报道。该文分析了丹麦全国青年男性病人接受Lichtenstein术或传统非补片修补术后复发再手术的风险。方法在丹麦国家疝数据库内前瞻性地记录了1998-01-01至2005-12-31所有接受手术治疗的腹股沟疝病例,以复发再次接受手术作为判定复发的标准。该研究的入组标准为数据库中年龄在18～30岁之间的原发性腹股沟斜疝并在研究阶段内接受了第一次修补手术的病例。结果在纳入该研究的3181例病人中,1120例(中位年龄23岁,范围18～30岁)接受了单纯的缝合修补术,2061例(中位年龄24岁,范围18～30岁)接受了Lichtenstein术。单纯缝合组的中位随访时间为62(0～96)个月,Lichtenstein组的中位随访时间为41(0～96)个月。两组的5年累积复发再手术率分别为1.6%(Lichtenstein组)和3.9%(单纯缝合组),单纯缝合组的术后总复发再手术率(39例,3.5%)为Lichtenstein组(24例,1.2%)的将近3倍(P=0.003)。结论与传统单纯缝合修补术相比,在18～30岁的青年腹股沟斜疝病人中应用Lichtenstein术可以大大降低术后复发的风险。但是,在治疗青年男性病人中应用补片时必须衡量术后可能发生的慢性疼痛的风险。
