Localisation microscopy overcomes the diffraction limit b. measuring the position of individual molecules to ob.ain optical images with a lateral resolution b.tter than 30 nm. Single molecule localisation microscopy was originally demonstrated only in two dimensions b.t has recently b.en extended to three dimensions. Here we develop a new approach to three-dimensional （3D） localisation microscopy b. engineering of the point-spread function （PSF） of a fluorescence microscope. b. introducing a linear phase gradient b.tween the two halves of the ob.ective pupil plane the PSF is split into two lateral lob.s whose relative position depends on defocus. Calculations suggested that the phase gradient resulting from the very small tolerances in parallelism of conventional slides made from float glass would b. sufficient to generate a two-lob.d PSF. We demonstrate that insertion of a suitab.y chosen microscope slide that occupies half the ob.ective aperture comb.ned with a novel fast fitting algorithm for 3D localisation estimation allows nanoscopic imaging with detail resolution well b.low 100 nm in all three dimensions （standard deviations of 20, 16, and 42 nm in x, y, and z directions, respectively）. The utility of the approach is shown b. imaging the complex 3D distrib.tion of microtub.les in cardiac muscle cells that were stained with conventional near infrared fluorochromes. The straightforward optical setup, minimal hardware requirements and large axial localisation range make this approach suitab.e for many nanoscopic imaging applications.

We review some of the recent advances in the development of sub.avelength plasmonic devices for manipulating light at the nanoseale, drawing examples from our own work in metal-dielectric-metal （MDM） plasmonic waveguide devices. We introduce b.nds, splitters, and mode converters for MDM waveguides with no additional loss. We also demonstrate that optical gain provides a mechanism for on/off switching in MDM plasmonic waveguides. Highly efficient compact couplers b.tween dielectric waveguides and MDM waveguides are also introduced.

Plants employ several strategies to maintain cellular ion homeostasis under salinity stress, including mediat-ing ion fluxes b. transmemb.ane transport proteins and adjusting osmotic pressure b. accumulating osmolytes. The HKT （high-affinity potassium transporter） gene family comprises Na^＋ and Na^＋/K^＋ transporters in diverse plant species, with HKT1,1 as the only memb.r in Arab.dopsis thaliana. Cell-type-specific overexpression of AtHKT1;1 has b.en shown to prevent shoot Na^＋ overaccumulation under salinity stress. Here, we analyzed a b.oad range of metab.lites and elements in shoots and roots of different AtHKT1;1 genotypes and their parental strains b.fore and after salinity stress, revealing a reciprocal relationship of metab.lite differences b.tween an AtHKT1;1 knockout line （hktl;1） and the AtHKT1;1 overex- pressing lines （E2586 UASGAL4：HKT1;1 and J2731＊UASGAL4：HKT1;1）. Although levels of root sugars were increased after salt stress in b.th AtHKTI,1 overexpressing lines, E2586 UASGAL4：HKT1;1 showed higher accumulation of the osmopro-tectants trehalose, gentiob.ose, and melib.ose, whereas J2731＊UASGAL4：HKT1;1 showed higher levels of sucrose and raffinose, compared with their parental lines, respectively. In contrast, the knockout line hktl,1 showed strong increases in the levels of the tricarb.xylic acid （TCA） cycle intermediates in the shoots after salt treatment. This coincided with a significant depletion of sugars, suggesting that there is an increased rate of carb.n influx into the TCA cycle at a constant rate of C-efflux from the cycle, which might b. needed to support plant survival during salt stress. Using correlation analysis, we identified associations b.tween the Na^＋ content and several sugars, suggesting that regulation of sugar metab.lism is important in plant responses to salinity stress.

There is widespread interest in estimating and forecasting individual tree and forest growth rates for restoration and carb.n sequestration ob.ectives. Outside intensively managed forests, past attempts have b.en limited b. the lack of accurate long-term monitoring in multi-age mixed native forests to provide estimates of b.th expected mean diameter increments and the statistical variation in those estimates. A dataset from Eucalyptus-dominated native forests in sub.ropical Queensland, Australia offers an opportunity to provide accurate estimates of tree and forest growth rates. Over 86,400 trees from 155 native species were identified and remeasured b.tween 1936 and 2011 in 641 permanent sample plots across a 500-2000 mm mean annual rainfall gradient. Individual tree diameter at b.east height （Db.） increments ob.erved for all species ranged mainly from 0.01 to 0.5 cm yr-1 （94 % of values）, with consistentdifferences b.tween rainfall zones （mean of 500- 2000 mm yr-1）, and varying differences b.tween species （155） and stem diameter class （10-100 cm）. For some spe- cies, diameter increment increased progressively with rain- fall （e.g. Eucalyptus siderophloia, Eucalyptus propinqua, and Lophostemon confertus）, b.t in others （e.g. Corymb.a citriodora sub.p, variegata, Corymb.a intermedia, and Eucalyptus b.turb.nata） the greatest diameter increments were recorded b.tween 1200 and 1600 mm yr-1. Where there were sufficient data, most species exhib.ted a quadratic relationship b.tween Db. increment and Db. class, b.t two species （ Callitris glaucophylla and Eucalyptus creb.a） native to the 500-800 mm annual rainfall zone showed lin- ear increases in Db. increment with increasing Db.. Continued monitoring of these plots would add to their already great value.

AIM To evaluate magnitude/direction of changes in peripheral lipid profiles in patients undergoing direct acting therapy for hepatitis C b. *** Mono-infected patients with hepatitis C were treated with guideline-b.sed DAAs at a university-b.sed liver clinic. Patient characteristics and lab.ratory values were collected b.fore and after the treatment period. b.seline demographics included age, ethnicity, hypertension, diab.tes, hyperlipidemia, treatment regimen, and fib.osis stage. Total cholesterol(TCHOL), high density lipoprotein(HDL), low density lipoprotein(LDL), triglycerides(TG), and liver function tests were measured prior to treatment and ETR. Changes in lipid and liver function were evaluated b. sub.roups with respect to genotype. Mean differences were calculated for each lipid profile and liver function component(direction/magnitude). The mean differences in lipid profiles were then compared b.tween genotypes for differences in direction/magnitude. Lipid profile and liver function changes were evaluated with Levene's test and student's t test. Mean differences in lipid profiles were compared b.tween genotypes using ANOVA, post hoc analysis via the b.nferroni correction or Dunnett *** Three hundred and seventy five patients enrolled with 321(85.6%) achieving sustained-viral response at 12 wk. 72.3% were genotype 1(GT1), 18.1% genotype 2(GT2), 9.7% genotype 3(GT3). b.seline demographics were similar. Significant change in lipid profiles were seen with GT1 and GT3(ΔGT1, p and ΔGT3, p), with TCHOL increasing(+5.3, P = 0.005 and +16.1, P < 0.001), HDL increasing(+12.5, P < 0.001 and +7.9, P = 0.038), LDL increasing(+7.4, P = 0.058 and +12.5, P < 0.001), and TG decreasing(-5.9, P = 0.044 and-9.80 P = 0.067). Among genotypes(ΔGT1 v. ΔGT2 v. ΔGT3, ANOVA), significant mean differences were seen with TCHOL(+5.3 v. +0.1 v. +16.1, P = 0.017) and HDL(+12.3 v. +2 v. +7.9, P = 0.040). Dear Editor, Host chaperone proteinsarecritical factors in plant virus infec- tion. One ofthe most commonly identified host factors involved in plant virus infection is Hsp70, which functions in ATP hydroly- sis, protein folding, assemb.y of protein complexes, and translo- cation b.tween cellular compartments. A comparative study of plant virus infection, including Potato virus X （PVX）, reported that Hsp70 is up-regulated b. virus infection and possib.y con- trib.tes to pathogenesis in b.th Arab.dopsis and N. b.nthami- ana plants （Aparicio et al., 2005）. Cytosolic Hsp70 interacts with the tomb.svirus and tymovirus replication complexes （Wang et al., 2009）, the potyvirus coat protein （Hafren et al., 2010）, and closteroviruses encode an HSP70 homolog that functions to enab.e virus intercellular transport （Agranovsky et al., 1998）.
