A novel diagnostic gene region for distinguishing between two pest fruit flies:Bactrocera tryoni(Froggatt)and Bactrocera neohumeralis(Hardy)(Diptera:Tephritidae)

Bactrocera tryoni and Bactrocera neohumeralis are morphologically similar sibling pest fruit fly species that possess different biological attributes,geographic distributions,and host *** need to differentiate between the two species is critical for accurate pest status assessment,management,biosecurity,and maintenance of reference *** morphologically similar,adults may be separated based on subtle characters;however,some characters exhibit intraspecific variability,creating overlap between the two ***,there is currently no single molecular marker or rapid diagnostic assay that can reliably distinguish between *** and ***;therefore,ambiguous samples remain *** we report the first molecular marker that can consistently distinguish between *** and *** diagnostic region consists of two adjacent single nucleotide polymorphisms(SNPs)within the pangolin(pan)gene *** confirmed the genotypes of each species are consistent across their distributional range,then developed a tetra-primer amplification refractory mutation system(ARMS)PCR assay for rapid diagnosis of the *** assay utilizes four primers in multiplex,with two outer universal primers,and two internal primers:one designed to target two adjacent SNPs(AA)present in *** and the other targeting adjacent SNPs present in ***(GG).The assay accurately discriminates between the two species,but their SNP genotypes are shared with other nontarget tephritid fruit fly ***,this assay is most suited to adult diagnostics where species confirmation is necessary in determining ambiguous surveillance trap catches;maintaining pure colony lines;and in Sterile Insect Technique management responses.

Development of internal COI primers to improve and extend barcoding of fruit flies (Diptera: Tephritidae: Dacini)

Accurate species-level identifications underpin many aspects of basic and applied biology;however,identifications can be hampered by a lack of discriminating morphological characters,taxonomic expertise or *** approaches,such as DNA"barcoding"of the cytochrome c oxidase(COI)gene,are argued to overcome these ***,nuclear encoding of mitochondrial genes(numts)and poor amplification success of suboptimally preserved specimens can lead to erroneous *** insect group for which these molecular and morphological problems are significant are the dacine fruit flies(Diptera:Tephritidae:Dacini).We addressed these issues associated with COI barcoding in the dacines by first assessing several"universal"COI primers against public mitochondrial genome and numt sequences for dacine *** then modified a set of four primers that more closely matched true dacine COI sequence and amplified two overlapping portions of the COI barcode *** new primers were tested alongside universal primers on a selection of dacine species,including both fresh preserved and decades-old dry ***,Bactrocera tiyoni mitochondrial and nuclear genomes were compared to identify putative *** numt clades were identified,three of which were amplified using existing universal *** contrast,our new primers preferentially amplified the"true"mitochondrial COI barcode in all dacine species *** new primers also successfully amplified partial barcodes from dry specimens for which full length barcodes were *** we recommend these new primers be incorporated into the suites of primers used by diagnosticians and quarantine labs for the accurate identification of dacine species.

橘小实蝇Bactrocera dorsalis,又名东方果实蝇,是全球重要农业入侵性害虫并严重阻碍果蔬产品国际贸易。近年来的相关报道显示,木瓜实蝇***、菲律宾实蝇***、入侵实蝇***是橘小实蝇的同物异名,该虫分布范围从亚太地区扩展到了非洲大多数国家,其全球种群结构和入侵路径亟待研究。本研究在收集橘小实蝇全球63个种群样品的基础上,利用微卫星、线粒体基因cox1、nad6两种分子标记和翅形态测量学数据,探究橘小实蝇的宏观地理种群结构和入侵路径。结果表明,橘小实蝇亚洲种群表现出较高的遗传多样性且遗传结构不明显,南亚可能是其他亚洲种群的来源地,非洲和夏威夷的种群推断为南亚起源的单一入侵种群,我国中部地区的橘小实蝇为多次重复入侵的结果,很可能与果蔬贸易有关。因此,应大力加强针对橘小实蝇的全球入侵防控工作,特别是严防该虫通过果蔬贸易和人员往来传入亚洲中北部地区、欧洲、大洋洲及美洲大部分国家和地区。

Laboratory study of a hydronic concrete deck heated externally in a controlled sub-freezing environment

Geothermal heating of bridge decks is a reliable and sustainable method for bridge de-icing that has been in-creasing in demand since conventional de-icing methods were proved to be environmentally *** research on geothermal heating of bridge decks relied on hydronic pipes embedded inside of bridge decks,which are confined to newly constructed *** existing bridges,a newly devised method for external heating has been recently tested under limited laboratory conditions to determine its overall *** study explores laboratory heating tests of a concrete slab with a thickness representing a typical concrete bridge *** slab was equipped with a simulated geothermal bridge de-icing system and tested inside a freezer subjected to sub-freezing controlled *** winter scenarios were applied to the system to determine its heat-ing performance and how feasible it will be for the system to be transferred to the field.A prediction equation was developed to estimate the total energy reserves required to permit de-icing,and statistical analysis was performed and validated with test *** slab surface heat flux was estimated to range from 27 W/m^(2) k to 73 W/m^(2) k from the heating *** externally-heated deck can be designed with the developed prediction equation for snow melting.

Obesity has been linked to a range of pathologies,including *** contrast,regular physical activity is associated with the prevention or reduced progression of ***,physical activity can improve memory and spatial cognition,reduce age-related cognitive decline,and preserve brain volume,but the mechanisms are not fully ***,we investigated whether any detrimental effects of high-fat diet(HFD)-induced obesity on cognition,motor behavior,adult hippocampal neurogenesis,and brain-derived neurotrophic factor(BDNF)could be mitigated by voluntary exercise training in male C57Bl/6 ***-induced impairment of motor function was not reversed by ***,voluntary wheel running improved long-term memory and increased hippocampal neurogenesis,suggesting that regular physical activity may prevent cognitive decline in obesity.

The mating system of the true fruit fly Bactrocera tryoni and its sister species, Bactrocera neohumeralis

The frugivorous ＂true＂ fruit fly, Bactrocera tryoni （Queensland fruit fly）, is presumed to have a nonresourced-based lek mating system. This is largely untested, and contrary data exists to suggest Bactrocera tryoni may have a resource-based mating system focused on fruiting host plants. We tested the mating system ofBactrocera tryoni, and its close sibling Bactrocera neohumeralis, in large field cages using laboratory reared flies. We used observational experiments that allowed us to determine if：（i） mating pairs were aggregated or nonaggregated; （ii） mating system was resource or nonresource based; （iii） flies utilized possible landmarks （tall trees over short） as mate-rendezvous sites; and （iv） males called females from male-dominated leks. We recorded nearly 250 Bactrocera tryoni mating pairs across all experiments, revealing that：（i） mating pairs were aggregated; （ii） mating nearly always occurred in tall trees over short; （iii） mating was nonresource based; and （iv） that males and females arrived at the mate-rendezvous site together with no evidence that males preceded females. Bactrocera neohumeralis copulations were much more infrequent （only 30 mating pairs in total）, but for those pairs there was a similar preference for tall trees and no evidence of a resource-based mating system. Some aspects of Bactrocera tryoni mating behavior align with theoretical expectations of a lekking system, but others do not. Until evidence for unequivocal female choice can be provided （as predicted under a true lek）, the mating system ofBactrocera tryoni is best described as a nonresource based, aggregation system for which we also have evidence that land-marking may be involved.
