Expression and modulation of neuroligin and neurexin in the olfactory organ of the cotton leaf worm Spodoptera fittoralis

Carboxylesterases are enzymes widely distributed within living organisms. In insects, they have been mainly involved in dietary metabolism and detoxification function. Interestingly, several members of this family called carboxylesterase-like ad- hesion molecules （CLAMs） have lost their catalytic properties and are mainly involved in neuro/developmental functions. CLAMs include gliotactins, neurotactins, glutactins, and neuroligins. The latter have for binding partner the neurexin. In insects, the function of these proteins has been mainly studied in Drosophila central nervous system or neuro- muscular junction. Some studies suggested a role of neuroligins and neurexin in sensory processing but CLAM expression within sensory systems has not been investigated. Here, we reported the identification of 5 putative CLAMs expressed in the olfactory system of the model pest insect Spodoptera littoralis. One neuroligin, Slnlg4-yll and its putative binding partner neurexin Slnrxl were the most expressed in the antennae and were surprisingly associated with olfactory sensilla. In addition, both transcripts were upregulated in male antennae after mating, known to modulate the sensitivity of the peripheral olfactory system in S. littoralis, suggesting that these molecules could be involved in sensory plasticity.

Objectives: To use a novel in vitro model of three-dimensional(3D) neurosphere cultures to assess neurotoxic or neuroprotective effects with harmane as a model ***: A reproducible model of 3D spheroids was developed from embryonic mouse cortical neurons,using molded agarose micro-wells;this method seems particularly practical as it is customizable and widely available and does not require specific cell treatments or assay components different from 2D cultures, allowing for the easy transposition of routine protocols. To assess the neurotoxic effects of harmane, a resazurin assay was performed to measure cell viability, and a highly sensitive fluorometric method, based on the oxidation of dichlorodihydrofluorescein, was applied to measure eventually induced reactive oxygen species(ROS) after exposure to harmane at increasing concentrations of 50 100,and 250 μ***: Hydrogel microwells facilitated the assembly of spheroids containing neurons and glial cells into a complex 3D structure and prevented the agglomeration of spheroids. Exposure to harmane induced cytotoxicity in 3D neural spheroids, which was correlated with harmane concentrations, with a 27%reduction in viability at 250 μm. Harmane that did not induce significant levels of oxidative stress was detected for all tested ***: This 3D neurosphere model mimics a neuronal microenvironment, allowing a fine study of neurodegenerative disorders and the effects of chemicals on the brain. This model opens novel opportunities, not only from a pathogenetic point of view but also from a therapeutic perspective.

分子电子学的首要目标是研究通过分子结或微/纳米尺度器件中的电荷输运机制。我们依靠理论进行了实验方案的设计和实施,对输运机制进行了详细分析。单态模型(SLM)是一种采用隧穿输运机制分析电流-电压(I-V)特性的理论方法。单个分子轨道的存在证明了其模型在基本电荷传输研究中用于识别系统物理信息的合理性。在本论文中,我们基于SLM的模型对分子结中的隧穿输运进行了以下分析研究。(1)我们利用胺作为锚定基团来调节耦合,探索了隧道结的结构和传输特性以及寡苯烯和寡乙酰骨架,以支持单能级状态。由于条件的完善,在金表面形成了一层由不同长度的胺锚定分子组成的紧密、均匀的自组装单分子膜。通过使用共晶Ga In(EGa In)作为顶部电极构建分子结来进行电学表征,并揭示了隧穿机制。然后,我们通过拟合实验I-V曲线并应用SLM模型来确定结的相关参数,即耦合强度和势垒高度。低聚联苯胺和低聚并苯胺的隧穿势垒高度(能量偏移)和耦合强度相当。电子耦合的效率依赖于锚定基团,而对分子能级排列的贡献较小。(2)我们对三种不同的单态隧穿模型的性能进行了详细的模拟研究,并应用Landauer公式和两个近似解析公式分析了分子结中电荷输运机制。我们对模拟的I-V曲线和从模型拟合提取的物理信息与文献中实验数据进行对比,并对其实用性和局限性进行评价。这两种近似解析方法既不适合小耦合强度也不适合低能量偏移;高偏压下也偏离了准确的结果。这意味着如果模型没有在其内在约束下正确部署,则两种近似解析方法所提取的物理参数可能是错误的。因此,我们认为它们可能传递了关于系统的错误物理信息。最后,我们构建了一个实用性的图作为指导,分别指出了两种近似分析模型有效分析分子结的工作条件。

Agriculture has increased the release of reactive nitrogen to the environment due to crops’low nitrogen-use efficiency(NUE)after the application of *** like the use of stabilized-fertilisers with nitrification inhibitors such as DMPP(3,4-dimethylpyrazole phosphate)have been adopted to reduce nitrogen ***,cover crops can be used in crop-rotation-strategies to reduce soil nitrogen pollution and benefit the following ***(Sorghum bicolor)could be a good candidate as it is drought tolerant and its culture can reduce nitrogen losses derived from nitrification because it exudates biological nitrification inhibitors(BNIs).This work aimed to evaluate the effect of fallow-wheat and sorghum cover crop-wheat rotations on N_(2)O emissions and the grain yield of winter wheat *** addition,the suitability of DMPP addition was also *** use of sorghum as a cover crop might not be a suitable option to mitigate nitrogen losses in the subsequent *** sorghum–wheat rotation was able to reduce 22%the abundance of amoA,it presented an increment of 77%in cumulative N_(2)O emissions compared to fallow–wheat rotation,which was probably related to a greater abundance of heterotrophic-denitrification *** the other hand,the application of DMPP avoided the growth of ammonia-oxidizing bacteria and maintained the N_(2)O emissions at the levels of unfertilized-soils in both *** a conclusion,the use of DMPP would be recommendable regardless of the rotation since it maintains NH_(4)^(+)in the soil for longer and mitigates the impact of the crop residues on nitrogen soil dynamics.

The sowing pattern has an important impact on light interception efficiency in maize by determining the spatial distribution of leaves within the *** orientation is an important architectural trait determining maize canopies light *** studies have indicated how maize genotypes may adapt leaves orientation to avoid mutual shading with neighboring plants as a plastic response to intraspecific *** goal of the present study is 2-fold:firstly,to propose and validate an automatic algorithm(Automatic Leaf Azimuth Estimation from Midrib detection[ALAEM])based on leaves midrib detection in vertical red green blue(RGB)images to describe leaves orientation at the canopy level;and secondly,to describe genotypic and environmental differences in leaves orientation in a panel of 5 maize hybrids sowing at 2 densities(6 and 12 plants.m^(−2))and 2 row spacing(0.4 and 0.8 m)over 2 different sites in southern *** ALAEM algorithm was validated against in situ annotations of leaves orientation,showing a satisfactory agreement(root mean square[RMSE]error=0.1,R^(2)=0.35)in the proportion of leaves oriented perpendicular to rows direction across sowing patterns,genotypes,and *** results from ALAEM permitted to identify significant differences in leaves orientation associated to leaves intraspecific *** both experiments,a progressive increase in the proportion of leaves oriented perpendicular to the row is observed when the rectangularity of the sowing pattern increases from 1(6 plants.m^(−2),0.4 m row spacing)towards 8(12 plants.m^(−2),0.8 m row spacing).Significant differences among the 5 cultivars were found,with 2 hybrids exhibiting,systematically,a more plastic behavior with a significantly higher proportion of leaves oriented perpendicularly to avoid overlapping with neighbor plants at high *** in leaves orientation were also found between experiments in a squared sowing pattern(6 plants.m^(−2),0.4 m

The ability to accurately characterize projects novel project characteristic is developed characteristic, called project regularity, is essential to good project management. Therefore, a reflects the value accrue within a project. This expressed in terms of the newly introduced regular/irregular-indicator RI. The widely accepted management system of earned value management （EVM） forms the basis for evaluation of the new characteristic. More concretely, the influence of project regularity on EVMforecasting accuracy is assessed, and is shown to be significant for both time and cost forecasting. Moreover, this effect appears to be stronger than that of the widely used characteristic of project seriality expressed by the serial/parallel-indicator SP. Therefore, project regularity could also be useful as an input parameter for project network generators. Furthermore, the introduction of project regularity can provide project managers with a more accurate indication of the time and cost forecasting accuracy that is to be expected for a certain project and, correspondingly, of how a project should be built up in order to obtain more reliable forecasts during project control.

BACKGROUND The lack of space,as an indication for a native unilateral nephrectomy for positioning a future kidney graft in the absence of other autosomal dominant polycystic kidney disease-related symptoms,remains *** To evaluate the surgical comorbidity and the impact on graft survival of an associated ipsilateral native nephrectomy during isolated kidney transplantation in patients with autosomal dominant polycystic kidney *** One hundred and fifty-four kidney transplantations performed between January 2007 and January 2019 of which 77 without(kidney transplant alone(KTA)group)and 77 with associated ipsilateral nephrectomy(KTIN group),were retrospectively *** and surgical variables were analyzed and their respective impact on surgical comorbidity and graft *** Creation of space for future graft positioning was the main reason(n=74,96.1%)for associated ipsilateral *** significant difference in surgical comorbidity(lymphocele,wound infection,incisional hernia,wound hematoma,urinary infection,need for blood transfusion,hospitalization stay,Dindo Clavien classification and readmission rate)was observed between the two study *** incidence of primary nonfunction and delayed graft function was comparable in both groups[0%and 2.6%(P=0.497)and 9.1%and 16.9%(P=0.230),respectively,in the KTA and KTIN group].The 1-and 5-year graft survival were 94.8%and 90.3%,and 100%and 93.8%,respectively,in the KTA and KTIN group(P=0.774).The 1-and 5-year patient survival were 96.1%and 92.9%,and 100%and 100%,respectively,in the KTA and KTIN group(P=0.168).CONCLUSION Simultaneous ipsilateral native nephrectomy to create space for graft positioning during kidney transplantation in patients with autosomal dominant polycystic kidney disease does not negatively impact surgical comorbidity and short-and long-term graft survival.

Bite force is often associated with speciﬁc morphological features,such as sagittal *** presence of a pro-nounced sagittal crest in some tapirs(Perissodactyla:Tapiridae)was recently shown to be negatively correlated with hard-object feeding,in contrast with similar cranial structures in *** aim of this study was to investi-gate bite forces and sagittal crest heights across a wide range of modern and extinct tapirs and apply a comparative investigation to establish whether these features are correlated across a broad phylogenetic *** examined a sample of 71 specimens representing 15 tapir species(5 extant,10 extinct)using the dry-skull method,linear measurements of cranial features,phylogenetic reconstruction,and comparative *** were found to ex-hibit variation in bite force and sagittal crest height across their phylogeny and between different biogeographical realms,with high-crested morphologies occurring mostly in Neotropical *** highest bite forces within tapirs appear to be driven by estimates for the masseter–pterygoid muscle complex,rather than predicted forces for the temporalis *** results demonstrate that relative sagittal crest height is poorly correlated with relative cranial bite force,suggesting high force application is not a driver for pronounced sagittal crests in this *** divergent biomechanical capabilities of different contemporaneous tapirids may have allowed multiple species to occupy overlapping territories and partition resources to avoid excess *** forces in tapirs peak in Pleistocene species,independent of body size,suggesting possible dietary shifts as a potential result of climatic changes during this epoch.
