Dear Editor,Drug repurposing is a convenient alternative when the need for new drugs in an unexpected medical scenario is urgent,as is the case of emerging *** recent years,approaches based on network biology have demonstrated to be superior to genecentric ones.1 Here,we use an innovative methodology that combines mechanistic modeling of the signal transduction circuits related to SARS-CoV-2 infection(the COVID-19 disease map)with a machine-learning algorithm that learns potential causal interactions between proteins,already targets of drugs,and specific signaling circuits in the COVID-19 disease map,to suggest potentially repurposable drugs.

AIM:To describe and evaluate the efficacy of Ahmed glaucoma valve implantation(AGV)combined with pars plana vitrectomy(PPV)in a single surgical act for the treatment of advanced neovascular glaucoma(nVG).METHODS:Retrospective observational case series included 51 eyes from 50 patients with severe nVG treated with PPV,AGV,and panretinal photocoagulation and/or cryotherapy in a single surgical act during a 13-year period(2005-2018).Preoperative,intraoperative and postoperative data at day 1 and months 1,3,6,21,and 24 were systematically *** of surgical success was stablished at IOP between 6 and 21 mm Hg with or without topical ***:Main indications for surgery were nVG secondary to proliferative diabetic retinopathy(39.2%)and central retinal vein occlusion(37.3%).Mean(±SD)preoperative IOP was 42.0±11.2 mm Hg decreasing to 15.5±7.1 mm Hg at 12 mo and 15.8±9.1 mm Hg at 24 mo of follow *** incidence of success of IOP control was 76.0%at first postoperative month,reaching 88.3%at 6 *** of successful IOP control at long term was 74.4%at 12 mo and 71.4%at 24 *** evisceration for unsuccessful nVG management was required in 1 case(2.0%).COnCLUSIOn:Combination of AGV implantation and PPV in a single act may be a suitable option for severe forms of nVG in a case-by-case basis for effective IOP control and a complete panretinal photocoagulation.

Secondary metabolites and synthetic iminosugars that structurally resemble monosaccharides are potent inhibitors of a-glucosidase activity. The enzyme is core in cleaving sucrose in phloem feeding insects and it also plays a crucial role of reducing osmotic stress via the formation of oligosaccharides. Inhibition of hydrolysis by iminosug- ars should result in nutritional deficiencies and/or disruption of normal osmoregulation. Deoxynojirimycin （DnJ） and 2 n-alkylated analogs [n-butyl DnJ （nB-DnJ） and n-nonyl DnJ （nn-DnJ）] were the major iminosugars used throughout the study. The extensive experiments conducted with a-glucosidase of the whitefly Bemisia tabaci indicated the competitive nature of inhibition and that the hydrophilic DnJ is a potent inhibitor in com- parison to the more hydrophobic nB-DnJ and nn-DnJ compounds. The same inhibitory pattern was observed with the psyllid Cacopsylla bidens a-glucosidase. In contrast to the above pattern, enzymes of the aphids, Myzus persicae and Aphis gossypii were more sen- sitive to the hydrophobic iminosugars as compared to DnJ. In vivo experiments in which adult B. tabaci were fed dietary iminosugars, show that the hydrophilic DnJ was far less toxic than the lipophilic nB-DnJ and nn-DnJ. It is proposed that this pattern is attributed to the better accessibility of the hydrophobic nn-DnJ to the a-glucosidase membrane- bound compartment in the midgut. Based on the inhibitory effects of certain polyhydroxy n-alkylated iminosugars, a-glucosidase of phloem feeding hemipterans could serve as an attractive target site for developing novel pest control agents.

Experimental Analysis of the Aerodynamic Performance of an Innovative Low Pressure Turbine Rotor

In the present work the aerodynamic performances of an innovative rotor blade row have been experimentally investigated. Measurements have been carried out in a large scale low speed single stage cold flow facility at a Reynolds number typical of aeroengine cruise, under nominal and off-design conditions. The time-mean blade aerodynamic loadings have been measured at three radial positions along the blade height through a pressure transducer installed inside the hollow shaft, by delivering the signal to the stationary frame with a slip ring. The time mean aerodynamic flow fields upstream and downstream of the rotor have been measured by means of a five-hole probe to investigate the losses associated with the rotor. The investigations in the single stage research turbine allow the reproduction of both wake-boundary layer interaction as well as vortex-vortex interaction. The detail of the present results clearly highlights the strong dissipative effects induced by the blade tip vortex and by the momentum defect as well as the turbulence production, which is generated during the migration of the stator wake in the rotor passage. Phase-locked hot-wire investigations have been also performed to analyze the time-varying flow during the wake passing period. In particular the interaction between stator and rotor structures has been investigated also under off-design conditions to further explain the mechanisms contributing to the loss generation for the different conditions.
