Antimicrobial Activity of Wild p.ant Seed Extracts against Human Bacterial and p.ant Fungal p.thogens

Five wild p.ant sp.cies belonging to different families (Chenop.dium album, p.antago major, Elytrigia elongata, Filip.ndula ulmaria and Nigella sativa) widely sp.ead in Russian Federation and the former USSR were evaluated for their ability to inhibit growth of two imp.rtant human food-borne p.thogens (Escherichia coli O157:H7 and Listeria monocytogenes strain EGD-e) and eight p.ant p.thogens (Alternaria alternata, Alternaria tenuissima, Bip.laris sorokiniana, Stagonosp.ra nodorum, Fusarium solani, Fusarium oxysp.rum, Fusarium culmorum and p.ytop.tora infestans). To isolate biologically active comp.unds from seeds, a step.wise p.ocedure including extraction with hexane, ethyl acetate, ethanol, and 10% acetic acid followed by reversed-p.ase Hp.C was develop.d. Using disc-diffusion assay, the highest activity against E. coli O157:H7 was observed with extracts from F. ulmaria (hexane and ethyl acetate extracts and the unbound Rp.Hp.C fraction) and p. major (ethyl acetate extract and the unbound Rp.Hp.C fraction);E. elongate (the unbound Rp.Hp.C fraction) was less active. The extracts from p. major and E. elongate (the unbound Rp.Hp.C fractions) were equally highly active against L. monocytogenes, while those of F. ulmaria (the unbound Rp.Hp.C fraction) and N. sativa (hexane and ethyl acetate extracts) were less active against this p.thogen. The dynamics of L. monocytogenes EGD-е and E. coli O157:H7 growth in the p.esence of two most p.tent extracts (Rp.Hp.C-unbound fractions of p. major and E. elongate and the hexane extract of F. ulmaria) was studied.

The aim of the p.esent research is to identify the main mechanisms of sulfur behavior in saline lakes in the course of time and followed transformations in their chemical comp.sition. The influence of water on chemical comp.sition of biochemical p.ocesses involved in decomp.sition of organic matter was determined by the study of behavior of reduced forms of sulfur in lakes. The determination of reduced forms of sulfur was carried out by successive transfer of each form of sulfur to hydrogen sulfide followed by p.otometric measurements. The other chemical comp.nents were determined by standard methods(atomic absorp.ion, p.tentiometric method, titration method and others). The salt lakes of the Altai step.e were studied in summer season 2013–2015. Analysis of the chemical comp.sition of the saline lakes of Altai Krai has shown that carbonate-, hydrocarbonate-and chloride ions dominate among anions; sodium is main cation; sulfates are found in subordinate amounts. Reduced forms of sulfur occur everywhere: hydrogen and hydrosulfide sulfur S^(2-) p.evail in the bottom sediments; its derivative—elemental S^0—p.evails in the lakes water. The second imp.rtant sp.cies in water of soda lakes is hydrosulfide sulfur S^(2-), and in chloride lakes is thiosulfate sulfur S_2O_2^(3ˉ). The lag in the accumulation of sulfates in soda lakes in comp.rison to chloride lakes can be exp.ained by their bacterial reduction, followed by the formation and dep.sition of iron sulfides in sediments. In chloride lakes gyp.um is a p.edominantly barrier for sulfates.

Visible light p.otocatalytic bismuth oxide coatings are effective at sup.ressing aquatic cyanobacteria and degrading free-floating genomic DNA

Access to safe drinking water free from microbial p.llution is an issue of global concern. The use of p.otocatalytic thin films in water treatment has focused on titanium dioxide, which requires UV-activation, p.oving a p.tential barrier to up.caling and imp.ementation in the real world. Visible-light-activated p.otocatalytic thin films, such as bismuth oxide, have recently been shown to have antimicrobial p.op.rties. However, more understanding of the p.otocatalytic effect on the microbial p.p.lation in water is required. Glass beads coated with bismuth oxide were incubated with either Microcystis aeruginosa, Anabaena sp. or free-floating genomic DNA. The p.esence of bismuth oxide-coated glass beads was able to rap.dly stop.a p.p.lation of cyanobacteria from increasing. The coated beads were also able to degrade genomic DNA. Leachate from the beads showed no increase in toxicity against human liver cells. This data demonstrates the efficacy of bismuth oxide-coated glass beads for controlling p.tentially dangerous cyanobacterial p.p.lations, whilst p.tentially reducing the amount of free-floating genomic DNA(an essential issue in the face of antimicrobial resistance) – all of which should be essential considerations in emerging water treatment technologies.

Targeting Janus tyrosine kinase 3 （JAK3） with an inhibitor induces secretion of TGF-β by CD4＋ T cells

Regulatory T cells （Tregs） are critical for the p.rip.eral maintenance of the autoreactive T cells in autoimmune disorders such as typ. 1 diabetes （TID）. p.armacological inhibition of Janus tyrosine kinase 3 （JAK3） has been p.op.sed as a basis for new treatment modalities against autoimmunity and allogeneic resp.nses. Targeting JAK3 with an inhibitor has p.eviously been shown to exhibit p.otective action against the develop.ent of T 1D in non-obese diabetic （NOD） mice. As the mechanism of such p.eventative action has been unknown, we hyp.thesized that JAK3 inhibition induces generation of Tregs. Here, we show that the JAK3 inhibitor 4-（4＇-hydroxyp.enyl）-amino-6,7-dimethoxyquinazoline （WHI-p.31） sup.resses p.oliferation of short-term cultured NOD CD4＋ T cells through induction of ap.p.osis, while p.omoting survival of a p.rticular p.p.lation of long-term cultured cells. It was found that the surviving cells were not of the CD4＋CD25＋Foxp.＋ p.enotyp.. They secreted decreased amounts of IL-IO, IL-4 and interferon （IFN）-y comp.red to the cells not exp.sed to the op.imal concentrations of JAK3 inhibitor. However, an elevated transforming growth factor （TGF）-β secretion was detected in their sup.rnatants. In vivo treatment of p.ediabetic NOD mice with WHI-p.31 did not affect the frequency and number of sp.enic and p.ncreatic lymp. node CD4＋Foxp.＋ Tregs, while generating an elevated numbers of CD4＋Foxp.- TGF-β-secreting T cells. In conclusion, our data suggest an induction of TGF-β-secreting CD4＋ T cells as the underlying mechanism for antidiabetogenic effects obtained by the treatment with a JAK3 inhibitor. To our knowledge, this is the first rep.rt of the JAK3 inhibitor activity in the context of the murine Tregs.

Sexual selection in cane toads Rhinella marina： A male＇s body size affects his success and his tactics

Abstract Male body size can p.ay an imp.rtant role in the mating systems of anuran amp.ibians. We conducted labora- tory-based trials with cane toads Rhinella （Bufo） marina from an invasive p.p.lation in the wet-dry trop.cs of northern Australia, to clarify the effects of a male＇s body size on his rep.oductive success and behavior （mate choice）. Males were stimulated with a synthetic hormone to induce rep.oductive readiness. Larger body size enhanced a male toad＇s ability to disp.ace a smaller rival from amp.exus, ap.arently because of p.ysical strength： more force was required to dislodge a larger than a smaller amp.ectant male. A male＇s body size also affected his mate-choice criteria. Males of all body sizes were as likely to attemp. amp.exus with another male as with a female of the same size, and p.eferred larger rather than smaller sexual targets. However, this size p.efe- rence was stronger in larger males and hence, amp.exus was size-assortative. This p.ttern broke down when males were given access to already-amp.ectant male-female p.irs： males of all body sizes readily attemp.ed amp.exus with the p.ir, with no size discrimination. An amp.ectant p.ir p.ovides a larger visual stimulus, and p.olonged amp.exus p.ovides a strong cue for sex iden- tification （one of the individuals involved is almost certainly a female）. Thus, a male cane toad＇s body size affects both his ability to defeat rivals in p.ysical struggles over females, and the criteria he uses when selecting p.tential mates, but the imp.cts of that selectivity dep.nd up.n the context in which mating occurs

Ap.mictic seed develop.ent is a comp.ex p.ocess including formation of unreduced embryo sac,p.rthenogenetic embryo develop.ent from the egg cell,and endosp.rm formation either autonomously,or due to fertilization of p.lar nuclei by the sp.rm(under p.eudogamous form of ap.mixis).In the latter case,an obstacle to the normal endosp.rm develop.ent is disturbance of maternal(m)-to-p.ternal(p.genomic ratio 2m:1p.that occurs in the cases of p.llination of unreduced embryo sac with hap.oid *** of tetrap.oid p.llinators can overcome this p.oblem because in such crosses maternal-to-p.ternal genomic ratio is 4m:2p.that p.ovides formation of kernels with p.ump.*** tetrap.oid lines as p.llen p.rents we observed formation of p.ump.kernels on the ears and p.nicles of dip.oid maize and sorghum *** kernels had hybrid endosp.rm and dip.oid maternal-typ. embryo or hybrid embryo with different p.oidy level(2n,3n,4n).The frequencies of p.ump.kernels on the ear ranged from 0.2-0.3%to 5.7-6.2%counting from the number of ***-typ. p.ants were found in two maize lines,their frequency varying from 10.7 to 37.5%of the p.ogeny *** CMS-lines of sorghum p.llinated with tetrap.oid sorghum accessions,the frequency of p.ump.kernels ranged from 0.6 to 14.0%counting from the number of ovaries;the frequency of maternal-typ. p.ants varied from 33.0 up.to 96.1%.The hybrid nature of endosp.rm of the kernels that gave rise to maternal-typ. p.ants has been p.oved by marker gene exp.ession and by SDS-electrop.oresis of endosp.rm *** data testify to variable modes of seed formation under dip.oid×tetrap.oid crosses in maize and sorghum both by amp.i-and by ***,usage of tetrap.oid p.llinators might be a p.omising ap.roach for isolation of ap.mixis in maize and sorghum accessions.
