Ocular flora in patients undergoing intravitreal injections: antibiotic resistance patterns and susceptibility to antiseptic picloxydine

AIM: To study antibiotic resistance patterns and susceptibility to eye antiseptic picloxydine of conjunctival flora in patients undergoing intravitreal injections(IVIs).METHODS: Conjunctival swabs were taken in 4 groups of patients, 20 patients in each group(n=80): without IVIs and ophthalmic operations in history(group n1;control group);with the first IVI and antibiotic eye drops Tobrex applied 3 d before IVI and 5 d after it(group n2);with 20 or more IVIs and repeated courses of antibiotic eye drops(group n3);with the first IVI and antiseptic eye drops Vitabact(picloxydine) applied 3 d before IVI and 5 d after it(group n4). In groups n2 and n4 swabs were taken at baseline and after the treatment. Efficacy of picloxydine in inhibition of growth of conjunctival isolates susceptible and resistant to antibiotic was studied in vitro. Minimal inhibition concentrations(MIC) were determined with microdilution ***: Two of the three patients who had to undergo the IVI procedure showed conjunctiva bacterial contamination. Along with few Staphylococcus aureus and Gram-negative isolates susceptible to most antibiotics, the majority(71%-77%) of causative agents were coagulase-negative Staphylococci(Co nS), 40%-50% of which were multidrug resistant(MDR). Eye disinfection in the operating room and peri-injection courses of Tobrex or Vitabact resulted in total elimination of isolates found at baseline. However, in 10% and 20% of patients, respectively, recolonization of the conjunctiva with differing strains occurred. In patients with repeated IVI and Tobrex/Maxitrol treatment, the conjunctival flora showed high resistance rates: 90% of Co nS were MDR. In the in vitro study, picloxydine showed bactericidal effect against Staphylococci isolates both antibiotic resistant and susceptible with MIC≥13.56 μg/m L. Effect of Growing Conditions of Rice Donor Plants on Anther Culture in Vitro

An experiment was conducted to find the effect of three types of donor plants growing conditions （growth chamber, open space ＋ growth chambers and open space） on callus induction and subsequent plant regeneration and productive shoot regeneration from the anthers culture in vitro of four rice hybrid （1-2, 2-1, 7-1, 13-3） developed by Primorsky Scientific Research Institute of Agriculture. Five variants of n6 medium （n6-I, n6-2, n6-3, Mix-l, n6-4） were used as basal medium. Mean value of callus induction frequency on three types of conditions ranged from 5.68% to 9.44% and the difference was non-significantly. In general, callus derived from donor plants grown on condition of open space ＋ growth chambers showed significantly better performances for plant regeneration （0.23 green regenerants on anther and 3.77 green regenerants on callus） and productive shoot regeneration （0.06 productive regenerants on anther and 0.56 productive regenerants on callus）. Favourable conditions for donor plant growth in open space positively affect on callus induction and regenaration. It is possible to get assured results on many hybrids, but not the highest. In growth chamber, frequency of callus induction can be the maximal only on some samples, few hybrids are resulted in deficiency of callus induction.

Kinetics of the H＋-ATPase from Dry and 5-Hours- Imbibed Maize Embryos in Its native, Solubilized, and Reconstituted Forms

Membranes undergo recovery upon rehydration in seed germination. Previous work has described that the plasma membrane H＋-ATPase from maize embryos adopts two different forms at 0 and 5 h of imbibition. We investigated how the kinetics of these two forms could be affected by alterations in the plasma membrane （PM）. In comparison to the O-h, PMs from the 5-h imbibed embryos showed changes in glycerophospholipid composition, decrease in leakage, and increase in fluidity. Kinetics of the PM H^-ATPase from 0 and 5-h imbibed embryos showed negative cooperativity, With the removal of the membrane environment, the activity of the enzymes shifted to a more complex kinetics, displaying two enzyme components. Lipid reconstitution produced one component with positive cooperativity. In all cases, enzymes from 0 and 5-h imbibed embryos presented similar kinetics with some quantitative differences. These results indicate that the two enzyme forms have the potential ability to respond to changes in the membrane enyiror^rpent, but the fact that they do not show differences in the native membranes at 0 or 5 h implies that modifications in the membrane are not drastic enough to alter their kinetics, or that they are able to preserve their boundary lipids or associated proteins and thus retain the same kinetic behavior.

A pilot study of the relative number of circulating tumor cells and leukocytes containing actin-binding proteins in head and neck cancer patients

Circulating tumor cells(CTCs)play an important role in tumor metastases,which is positively correlated with an increased risk of ***-binding proteins,including cofilin(CFL1),profilin 1(PFn1),and adenylate cyclase-associated protein 1(CAP1),are thought to be involved in tumor cell motility and metastasis,specifically in head and neck squamous cell carcinoma(HnSCC).However,currently,there are no published studies on CFL1,PFn1,and CAP1 in CTCs and leukocytes in HnSCC *** assessed serum levels of CFL1,PFn1,and CAP1 and the number of CTCs and leukocytes containing these proteins in blood from 31 HnSCC patients(T1-4n0-2M0).The analysis used flow cytometry and an enzyme-linked immunosorbent assay *** found that CAP1+CTCs and CAP1+leukocyte subpopulations were prevalent in these HnSCC patient samples,while the prevalence rates of CFL1+and PFn1+CTCs were relatively *** with stage T2-4n1-2M0 had CFL1+and PFn1+CTCs with an elevated PFn1 serum level,compared with the T1-3n0M0 *** summary,the PFn1 serum level and the relative number of PFn1+CD326+CTCs could be valuable prognostic markers for HnSCC *** current study is the first to obtain data regarding the contents of actin-binding proteins(ABPs)in CTCs,and leukocytes in blood from HnSCC *** is also the first to assess the relationship between the number of CTCs subgroups and disease characteristics.

Water scarcity threatens global food security and agricultural systems are challenged to achieve high yields while optimizing water *** deficit can be accentuated by soil physical degradation,which also triggers water losses through runoff and consequently soil *** soil health in cropping systems within the Brazilian Cerrado biome have been surveyed throughout the years,information about soil erosion impacts and its mitigation are still not well understood;especially concerning the role of cropping system diversification and its effects on crop ***,the aim of this study was to assess whether ecological intensification of cropping systems-inclusion of a consorted perennial grass and crop rotation-could promote soil coverage and consequently decrease water erosion and soil,water,and nutrient *** work studied the effects of crop rotation and consorted Brachiaria,along with different levels of investment in fertilization on soil physical quality and on soil,water,and nutrient losses,and crop *** proved that soybean monoculture(SS)is a system of low sustainability even under no-till in the Brazilian Cerrado *** exhibited high susceptibility to soil,water,and nutrient losses,causing low crop *** results showed that water losses in SS cropping system were approximately 10%of the total annual rainfall,and total K losses would require an additional 35%of K ***,ecological intensification of cropping systems resulted in enhanced soil environmental and agronomic functions,increased grain yield,and promoted soil and water conservation:high soil cover rate,and low soil,water and nutrient *** intensification proved to be an adequate practice to boost crop resilience to water deficit in the Brazilian Cerrado.

Genes associated with testicular germ cell tumors and testicular dysgenesis in patients with testicular microlithiasis

Testicular microlithiasis （TM） is one of the symptoms of testicular dysgenesis syndrome （TDS）. TM is particularly interesting as an informative marker of testicular germ cell tumors （TGCTs）. KIT ligand gene （KITLG）, BCL2 antagonist/killer 1 （BAK1）, and sprouty RTK signaling antagonist 4 （SPRY4） genes are associated with a high risk of TGCTs, whereas bone morphogenetic protein 7 gene （BMP7）, transforming growth factor beta receptor 3 gene （TGFBR3）, and homeobox D cluster genes （HOXD） are related to TDS. Using polymerase chain reaction-restriction fragment length polymorphism （PCR-RFLP） analysis, we investigated allele and genotype frequencies for KITLG （rs995030, rs1508595）, SPRY4 （rs4624820, rs6897876）, BAK1 （rs210138）, BMP7 （rs388286）, TGFBR3 （rs12082710）, and HOXD （rs17198432） in 142 TGCT patients, 137 TM patients, and 153 fertile men （control group）. We found significant differences in the KITLG GG_rs995030 genotype in TM （P= 0.01） and TGCT patients （P = 0.0005） compared with the control. We also revealed strong associations between KITLGrs1508595 and TM （G allele, P = 0.003; GG genotype, P= 0.01） and between KITLG_rs1508595 and TGCTs （G allele, P-- 0.0001; GG genotype, P = 0.0007）. Moreover, there was a significant difference in BMP7_rs388286 between the TGCT group and the control （T allele, P = 0.00004; TT genotype, P = 0.00006） and between the TM group and the control （T allele, P= 0.04）. HOXDalso demonstrated a strong association with TGCTs （rs17198432 A allele, P = 0.0001; AA genotype, P = 0.001）. Furthermore, significant differences were found between the TGCT group and the control in the BAK1_rs210138 G allele （P= 0.03） and the GG genotype （P= 0.01）. KITLG and BMP7genes, associated with the development of TGCTs, may also be related to TM. In summary, the KITLG GG_rs995030, GG_rs1508595, BMP7 TT_rs388286, HOXD AA_rs17198432, and BAK1 GG_rs210138 genotypes were associated with a high risk of TGCT development.
