Oxygen is essential for plant growth and development.Hypoxia occurs in plants due to limited oxygen avail-abilityfollowing adverse environmental conditions as well inhypoxic niches in otherwisenormoxic environ-ments.However,the existence and functional integration of spatiotemporal oxygen dynamics with plant development remains unknown.In animal systems dynamic fluctuations in oxygen availability are known as cyclic hypoxia.In this study,we demonstrate that cyclic fluctuations in internal oxygen levels occur in young emerging leaves of Arabidopsis plants.Cyclic hypoxia in plants is based on a mechanism requiring the ETHYLENE RESPONSE FACTORS type VII(ERFVII)that are central components of the oxygen-sensing machinery in plants.The ERFVll-dependent mechanism allows precise adjustment of leaf growth in response to carbon status and oxygen availability within plant cells.This study thus establishes a functional connection between internal spatiotemporal oxygen dynamics and developmental processes of plants.

The cold-induced factor CBF3 mediates root stem cell activity,regeneration,and developmental responses to cold

Plant growth and development involve the specification and regeneration of stem cell niches(SCNs).Although plants are exposed to disparate environmental conditions,how environmental cues affect developmental programs and stem cells is not well understood.Root stem cells are accommodated in meristems in SCNs around the quiescent center(QC),which maintains their activity.Using a combination of genetics and confocal microscopy to trace morphological defects and correlate them with changes in gene expression and protein levels,we show that the cold-induced transcription factor(TF)C-REPEAT BINDING FACTOR 3(CBF3),which has previously been associated with cold acclimation,regulates root development,stem cell activity,and regeneration.CBF3 is integrated into the SHORT-ROOT(SHR)regulatory network,forming a feedback loop that maintains SHR expression.CBF3 is primarily expressed in the root endodermis,whereas the CBF3 protein is localized to other meristematic tissues,including root SCNs.Complementation of cbf3-1 using a wild-type CBF3 gene and a CBF3 fusion with reduced mobility show that CBF3 movement capacity is required for SCN patterning and regulates root growth.Notably,cold induces CBF3,affecting QC activity.Furthermore,exposure to moderate cold around 10℃–12℃promotes root regeneration and QC respecification in a CBF3-dependent manner during the recuperation period.By contrast,CBF3 does not appear to regulate stem cell survival,which has been associated with recuperation from more acute cold(-4℃).We propose a role for CBF3 in mediating the molecular interrelationships among the cold response,stem cell activity,and development.

In the present work,a new combination of Raman and ultraviolet and visible(UV/Vis)absorption spectroelectrochemistry in reflection mode is proposed.The new experimental setup allows obtaining the two kinds of spectroscopic data without interferences concomitantly with the electrochemical information.To the best of our knowledge,it is the first time to report the simultaneous obtention of electrochemical,electronic,and vibrational information in the same experiment.This new combination provides time-resolved information about the processes that are taking place on the electrode/solution interface which has significant implications in different fields of chemistry,such as modification of electrodes,studies of electrocatalytic reaction mechanisms,development of sensors,among others.Two different systems were used to demonstrate the advantages and capabilities of the brand-new technique,namely,the oxidation of potassium ferrocyanide,an out-sphere system that is usually employed in the validation of SEC techniques,and the electrochemical-surface enhanced Raman spectroscopy(EC-SERS)detection of crystal violet by in-situ formation of the silver SERS substrate,where the UV/Vis spectra were used to follow the formation of the SERS substrate,whereas the Raman response of a probe molecule was used to confirm either the formation of a nanostructured surface and to obtain the fingerprint of the molecule with a high time resolution.The brand-new experimental setup has shown to be useful,versatile,robust,compact,and easy to use for future applications.

Determining indoor air quality and identifying the origin of odour episodes in indoor environments

A methodology for identifying volatile organic compounds （VOCs） and determining air quality of indoor air has been developed. The air samples are collected using pump samplers by the inhabitants when they perceive odorous and/or discomfort episodes. Glass multi-sorbent tubes are connected to the pump samplers for the retention of VOC. The analysis is performed by automatic thermal desorption （ATD） coupled with gas chromatography-mass spectrometry （GC/MS）. This methodology can be applied in cases of sick building syndrome （SBS） evaluation, in which building occupants experience a series of varied symptoms that appear to be linked to time spent in the building. Chemical pollutants concentrations （e.g., VOC） have been described to contribute to SBS. To exemplify the methodology, a qualitative determination and an evaluation of existing VOC were performed in a dwelling where the occupants experienced the SBS symptoms. Higher total VOC （TVOC） levels were detected during episodes in indoor air （1.33 ±1.53 mg/m^3） compared to outdoor air （0.71± 0.46 mg/m^3）. The concentrations of individual VOCs, such as ethanol, acetone, isopropanol, 1-butanol, acetic acid, acetonitrile and 1-methoxy-2-propanol, were also higher than the expected for a standard dwelling. The external source of VOC was found to be an undeclared activity of storage and manipulation of solvents located at the bottom of a contiguous building.

Pancreatic cancer(PC)continues to pose a major clinical challenge.There has been little improvement in patient survival over the past few decades,and it is projected to become the second leading cause of cancer mortality by 2030.The dismal 5-year survival rate of less than 10%after the diagnosis is attributable to the lack of early symptoms,the absence of specific biomarkers for an early diagnosis,and the inadequacy of available chemotherapies.Most patients are diagnosed when the disease has already metastasized and cannot be treated.Cancer interception is vital,actively intervening in the malignization process before the development of a full-blown advanced tumor.An early diagnosis of PC has a dramatic impact on the survival of patients,and improved techniques are urgently needed to detect and evaluate this disease at an early stage.It is difficult to obtain tissue biopsies from the pancreas due to its anatomical position;however,liquid biopsies are readily available and can provide useful information for the diagnosis,prognosis,stratification,and follow-up of patients with PC and for the design of individually tailored treatments.The aim of this review was to provide an update of the latest advances in knowledge on the application of carbohydrates,proteins,cell-free nucleic acids,circulating tumor cells,metabo-lome compounds,exosomes,and platelets in blood as potential biomarkers for PC,focusing on their clinical relevance and potential for improving patient outcomes.

Cardiopulmonary Stability on a Patient with Hip Fracture and Severe Pulmonary Hypertension, Anesthetized with Lumbar-Sacral Plexus Block and Non-Invasive Ventilation: Case Report

Background: Pulmonary hypertension is defined as a mean arterial pressure in the pulmonary artery exceeding 20 mm Hg at rest, measured by means of right heart catheterization. Patients with pulmonary hypertension undergoing surgical procedures such as hemiarthroplasty present a high risk of fatal complications. Nonetheless, there are no widely accepted protocols explaining their perioperative care in great detail. Case presentation: We present a case with an 89-years-old patient, with comorbidities such as GOLD 4D chronic obstructive pulmonary disease (COPD) and, as a consequence of this, severe pulmonary hypertension with signs of right ventricular dysfunction, thus requiring of pulmonary vasodilator, that has suffered a subcapital hip fracture requiring urgent surgery. Surgery is carried out successfully, under regional lumbar-sacral plexus block and sedation assisted by non-invasive ventilation. Conclusion: Multidisciplinary specialized treatment, preoperative optimization, as well as the careful selection of both the surgical and anesthetic techniques to be used, are among the strategies that improve the perioperative outcome in patients with pulmonary hypertension with right ventricle systolic dysfunction. Regional lumbar-sacral plexus block plus sedation is a technique that maintains hemodynamic stability;however, these patients require advance measures and postoperative monitoring under intensive care.

AIM To develop a fast, low-cost diagnostic strategy to identify single point mutations in highly variable genomes such as hepatitis C virus(HCV).METHODS In patients with HCV infection, resistance-associated amino acid substitutions within the viral quasispecies prior to therapy can confer decreased susceptibility to direct-acting antiviral agents and lead to treatment failure and virological relapse. One such naturally occurring mutation is the Q80 K substitution in the HCV-NS3 protease gene, which confers resistance to PI inhibitors, particularly simeprevir. Low-cost, highly sensitive techniques enabling routine detection of these single point mutations would be useful to identify patients at a risk of treatment failure. Light Cycler methods, based on real-time PCR with sequencespecific probe hybridization, have been implemented in most diagnostic laboratories. However, this technique cannot identify single point mutations in highly variable genetic environments, such as the HCV genome. To circumvent this problem, we developed a new method to homogenize all nucleotides present in a region except the point mutation of interest. RESULTS Using nucleotide-specific probes Q, K, and R substitutions at position 80 were clearly identified at a sensitivity of 10%(mutations present at a frequency of at least 10% were detected). The technique was successfully applied to identify the Q80 K substitution in 240 HCV G1 serum samples, with performance comparable to that of direct Sanger sequencing, the current standard procedure for this purpose. The new method was then validated in a Catalonian population of 202 HCV G1-infected individuals. Q80 K was detected in 14.6% of G1 a patients and 0% of G1 b in our setting. CONCLUSION A fast, low-cost diagnostic strategy based on real-time PCR and fluorescence resonance energy transfer probe melting curve analysis has been successfully developed to identify single point mutations in highly variable genomes such as hepatitis C virus. Chronic Stress and Its Symptoms among Mexican Immigrants in Edmonton, Canada

Immigration-related stress is the result of an environment that imposes demands surpassing the psychological resources of those suffering from it with a destabilizing effect on their physical and psychological health. The purpose of this study is to determine what the chronic stress levels are and their association with psychophysiological symptoms and gender in Mexican immigrants living in Edmonton, Canada. A representative random sample was made of 237 immigrants in 2012. The stress symptoms inventory was used and a multiple regression and multivariate analysis was made. Results indicated that chronic stress levels were: 14% high, 46% medium and 40% low and psychophysiological predictor symptoms were: fatigue, constipation, frequent colds, cold hands and feet and insomnia, and the female gender. Therefore one may conclude that a precise identification of the predictor symptoms of chronic stress may help promote and implement programs aimed at timely prevention of stress before they affect the health and occupational safety of Mexican immigrants living in Edmonton, Canada.

Adsorption of Propazine, Simazine and Bisphenol A on the Surface of Nanoparticles of Iron Oxide Nanoparticles of Carbon and Metallic Oxides

Simazine and propazine are selective triazine herbicides currently in use to control broad-leaved weeds and annual grasses around the world. Bisphenol A (BPA) is an industrial chemical used in the production of polycarbonate plastics often found in consumer goods, such as plastic containers, baby bottles etc. These synthetic compounds are known to increase the risk of cancer, cause adverse reproductive effect in reptiles, mammals, birds, humans, and lead to other health problems. They have become some of the principal agents of contamination in water bodies around the world through herbicide runoff, industrial waste and leaching. Some triazines such as atrazine are banned in most European countries for over ten years due to their adverse reproductive effect in mammals, birds and humans;however propazine and simazine are still in use around the world. The removal of these compounds from contaminated water is an exigent challenge. In this study, we investigated their affinity for the surface of nanoparticles (NPS) and standard metallic oxides in an effort to exploit the unique potential applications of NPS for water purification systems. We studied the adsorption of the two triazines and BPA on the surface of NPS of iron (III) oxide, NPS of carbon, bulk iron (III) oxide and aluminum oxide at pH 6 and pH 8 using UV-Visible spectroscopy. Result indicates that these compounds have different affinity towards the surface of metallic oxides and carbon at various pHs. In general, there is relatively high adsorption of some of these compounds on the surface of NPS compared to bulk particles. NPS of carbon have shown the highest affinity for all the three compounds. The lower pH was found to be favorable for all of the compounds except for BPA. BPA have shown high adsorption at pH 8 than at pH 6.

Molecular Identification of Shiga-Toxin Producing and Enteropathogenic <i>Escherichia coli</i>(STEC and EPEC) in Diarrheic and Healthy Young Alpacas

Isolation and biochemical and molecular identification of 303 strains of Escherichia coli obtained from diarrheic and healthy young alpacas of Puno-Peru, were realized. PCR amplification for 7 virulence factor genes associated with STEC, STEC O157:H7, EPEC: sxt1, sxt2, rfbO157, fliCH7, hlyA, eae y bfp were determined. A total of 39 strains (12.88%) showed amplification for one or more of these genes. Twenty three strains (59%) were classified as STEC and 16 strains (41%) as EPEC. An 88.18% (34/39) of STEC and EPEC strains were obtained from healthy alpacas and only 11.82% (5/39) from diarrheic alpacas considering this specie as potential zoonotic reservoir of STEC and EPEC.
