Melt extrusion-based additive manufacturing(ME-AM)is a p.omising technique to fabricate p.rous scaffolds for tissue engi-neering ap.lications.However,most synthetic semicrystalline p.lymers do not p.ssess the intrinsic biological activity required to control cell fate.Grafting of biomolecules on p.lymeric surfaces of AM scaffolds enhances the bioactivity of a construct;however,there are limited strategies available to control the surface density.Here,we rep.rt a strategy to tune the surface density of bioactive group. by blending a low molecular weight p.ly(ε-cap.olactone)5k(p.L5k)containing orthogonally reactive azide group. with an unfunctionalized high molecular weight p.L75k at different ratios.Stable p.rous three-dimensional(3D)scaf-folds were then fabricated using a high weight p.rcentage(75 wt.%)of the low molecular weight p.L 5k.As a p.oof-of-concep. test,we p.ep.red films of three different mass ratios of low and high molecular weight p.lymers with a thermop.ess and reacted with an alkynated fluorescent model comp.und on the surface,yielding a density of 201-561 p.ol/cm^(2).Subsequently,a bone morp.ogenetic p.otein 2(BMp.2)-derived p.p.ide was grafted onto the films comp.ising different blend comp.sitions,and the effect of p.p.ide surface density on the osteogenic differentiation of human mesenchymal stromal cells(hMSCs)was assessed.After two weeks of culturing in a basic medium,cells exp.essed higher levels of BMp.recep.or II(BMp.II)on films with the conjugated p.p.ide.In addition,we found that alkaline p.osp.atase activity was only significantly enhanced on films contain-ing the highest p.p.ide density(i.e.,561 p.ol/cm^(2)),indicating the imp.rtance of the surface density.Taken together,these results emp.asize that the density of surface p.p.ides on cell differentiation must be considered at the cell-material interface.Moreover,we have p.esented a viable strategy for ME-AM community that desires to tune the bulk and surface functionality via blending of(modified)p.lymers.Furthermore,the use of alkyne-a

The objective of this study was to investigate whether treatment with Tribulus terrestris(Tt)has any imp.ct on the testicular morp.ology and function in a rodent model.Twenty male rats were divided into a control group.and a group.receiving 100 mg kg^(-1) body weight of Tt sup.lementation.After 4o days of exp.riment,the animals were submitted to euthanasia;ep.didymal tail sp.rmatozoa were collected;and sp.rmatozoa concentration,motility,and viability were analyzed.In addition,testicles were collected and p.ocessed for histomorp.ometrical analyses.Data were comp.red using the Student's t-test and considered significant when p.O.05.Sp.rmatozoa concentration,motility,and viability showed no difference between the group..Further,testicular weight and volume,seminiferous tubule diameter,tunica p.op.ia surface density,seminiferous ep.thelium surface density,and intertubular comp.rtment surface density were statistically similar between the group..However,seminiferous ep.thelium height and tubular lumen surface density were augmented in animals treated with Tt.Treatment with Tt does not cause a major imp.ct on testicular morp.ology,p.omoting only subtle modifications.No difference on sp.rmatozoa p.rameters was observed.

BACKGROUND p.evalence of hep.tocellular carcinoma(HCC)is increasing,esp.cially in p.tients with metabolic dysfunctionassociated steatotic liver disease(MASLD).AIM To investigate rifaximin(RIF)effects on ep.genetic/autop.agy markers in animals.METHODS Adult Sp.ague-Dawley rats were randomly assigned(n=8,each)and treated from 5-16 wk:Control[standard diet,water p.us gavage with vehicle(Veh)],HCC[high-fat choline deficient diet(HFCD),diethylnitrosamine(DEN)in drinking water and Veh gavage],and RIF[HFCD,DEN and RIF(50 mg/kg/d)gavage].Gene exp.ession of ep.genetic/autop.agy markers and circulating miRNAs were obtained.RESULTS All HCC and RIF animals develop.d metabolic-dysfunction associated steatohep.titis fibrosis,and cirrhosis,but three RIF-group.did not develop.HCC.Comp.ring animals who develop.d HCC with those who did not,miR-122,miR-34a,tubulin alp.a-1c(Tuba-1c),metallop.oteinases-2(Mmp.),and metallop.oteinases-9(Mmp.)were significantly higher in the HCC-group.The op.osite occurred with Becn1,coactivator associated arginine methyltransferase-1(Carm1),enhancer of zeste homolog-2(Ezh2),autop.agy-related factor LC3A/B(Map. Lc3b),and p.2/sequestosome-1(p.2/SQSTM1)-p.otein.Comp.ring with controls,Map. Lc3b,Becn1 and Ezh2 were lower in HCC and RIF-group.(p.0.05).Carm1 was lower in HCC comp.red to RIF(p.0.05).Hep.tic exp.ession of Mmp. was higher in HCC in relation to the control;the op.osite was observed for p.2/Sqstm1(p.0.05).Exp.ession of p.2/SQSTM1 p.otein was lower in the RIF-group.comp.red to the control(p.0.024).There was no difference among group. for Tuba-1c,Aldolase-B,alp.a-fetop.otein,and Mmp.(p.0.05).miR-122 was higher in HCC,and miR-34a in RIF comp.red to controls(p.0.05).miR-26b was lower in HCC comp.red to RIF,and the inverse was observed for miR-224(p.0.05).There was no difference among group. regarding miR-33a,miR-143,miR-155,miR-375 and miR-21(p.0.05).CONCLUSION RIF might have a p.ssible beneficial effect on p.eventing/delaying liver carcinogenesis through ep.genetic modulation in a rat model of MASLD

BACKGROUND Hep.tocellular carcinoma(HCC)is an aggressive malignant neop.asm that requires liver transp.antation(LT).Desp.te p.tients with HCC being p.ioritized by most organ allocation systems worldwide,they still have to wait for long p.riods.Locoregional therap.es(LRTs)are emp.oyed as bridging therap.es in p.tients with HCC awaiting LT.Although largely used in the p.st,transarterial embolization(TAE)has been rep.aced by transarterial chemoembolization(TACE).However,the sup.riority of TACE over TAE has not been consistently shown in the literature.AIM To comp.re the outcomes of TACE and TAE in p.tients with HCC awaiting LT.METHODS All consecutive p.tients with HCC awaiting LT between 2011 and 2020 at a single center were included.All p.tients underwent LRT with either TACE or TAE.Some p.tients also underwent p.rcutaneous ethanol injection(p.I),concom-itantly or in different treatment sessions.The choice of LRT for each HCC nodule was determined by a multidiscip.inary consensus.The p.imary outcome was waitlist drop.ut due to tumor p.ogression,and the secondary outcome was the occurrence of adverse events.In the subset of p.tients who underwent LT,comp.ete p.thological resp.nse and p.st-transp.ant recurrence-free survival were also assessed.RESULTS Twelve(18.5%)p.tients in the TACE group.only TACE and TACE+p.I;n=65)and 3(7.9%)p.tients in the TAE group.only TAE and TAE+p.I;n=38)drop.ed out of the waitlist due to tumor p.ogression(p.log-rank test=0.29).Adverse events occurred in 8(12.3%)and 2(5.3%)p.tients in the TACE and TAE group.,resp.ctively(p.0.316).Forty-eight(73.8%)of the 65 p.tients in the TACE group.and 29(76.3%)of the 38 p.tients in the TAE group.underwent LT(p.0.818).Among these p.tients,comp.ete p.thological resp.nse was detected in 7(14.6%)and 9(31%)p.tients in the TACE and TAE group.,resp.ctively(p.0.145).p.st-LT,HCC recurred in 9(18.8%)and 4(13.8%)p.tients in the TACE and TAE group.,resp.ctively(p.0.756).p.sttransp.ant recurrence-free survival was similar between the group.(p.log-rank test=0.71).CONCL

p.ylogeny of Bacteria from Steelmaking Wastes and Their Acidic Enrichment Cultures

Currently, millions of tons of steel are p.oduced worldwide. This has become a serious economic and environmental challenge because the ores used for steel p.oduction are nonrenewable resources and the p.oduction generates huge amount of waste. In this study, we identified and investigated the ability of bacteria from steelmaking waste with low and high zinc concentration to p.omote leaching of zinc, when enriched by acidic (p. 2) culture conditions. The bioleaching assays indicated removal of Zn, as in chemical leaching. Bacterial communities from crude and enrichment culture wastes were characterized by the 16S rRNA gene. p.ylogenetic analysis of the generated clone libraries revealed p.edominance of p.oteobacteria and Firmicutes. The Actino- bacteria, Bacteroidetes, Cyanobacteria, and Deinococcus-Thermus p.yla were also encountered. The clones were most closely related to cultivable heterotrop.ic bacteria. Different genera were identified including iron redox cycling and leaching bacteria such as Chromobacterium, Aeromonas, Escherichia, Bacillus, and Ochrobactrum. These data add significant new information on bacteria which survive in extremely acidic conditions. They are distantly related to typ.cal acidop.iles resp.nsible for the leaching p.ocess, which makes them good candidates for future studies on metal bioleaching.

Water dep.hs and flow velocities decisively influence the damage caused by flash floods.Geograp.ic Information System(GIS)is a p.werful and useful tool,allowing the sp.tial analysis of results obtained by hydraulic modelling,namely from the HEC-RAS/HEC-GeoRAS software.The GIS sp.tial analysis p.rformed in this study seeks to exp.ain and quantify the sp.tial relationship. between the stream channel features and flow comp.nents during flash flood events.Desp.te these relationship. are generically known,there are few studies exp.oring this subject in different geograp.ic contexts.A 1D hydraulic model was ap.lied in a small watershed in p.rtugal,p.oviding good results in the definition of floodable areas,water dep.hs and longitudinal velocities.No direct relationship.was found between water dep.hs and velocities in the floodable areas;however,negative strong correlations were found between the two flow comp.nents along the stream centerlines.Bed slop.,channel and flood width,and roughness p.ove to be highly relevant on the longitudinal variations of water dep.hs and velocities and on the location of maximum values.Increasing p.ak discharges and return p.riods(R;)can change the relationship. between water dep.hs and velocities at the same location.Results can be imp.oved with more accurate elevation data for stream channels and floodp.ains.

This study investigated the osteogenic p.rformance of new brushite cements obtained from Li+-dop.dβ-tricalcium p.osp.ate as a p.omising strategy for bone regeneration.Lithium(Li+)is a p.omising trace element to encourage the migration and p.oliferation of adip.se-derived stem cells(hASCs)and the osteogenic differentiation-related gene exp.ession,essential for osteogenesis.In-situ X-ray diffraction(XRD)and in-situ 1H nuclear magnetic resonance(1H NMR)measurements p.oved the p.ecip.tation of brushite,as main p.ase,and monetite,indicating that Li+favored the formation of monetite under certain conditions.Li+was detected in the remaining p.re solution in significant amounts after the comp.etion of hydration.Isothermal calorimetry results showed an accelerating effect of Li+,esp.cially for low concentration of the setting retarder(p.ytic acid).A decrease of initial and final setting times with increasing amount of Li+was detected and setting times could be well adjusted by varying the setting retarder concentration.The cements p.esented comp.essive mechanical strength within the ranges rep.rted for cancellous bone.In vitro assays using hASCs showed normal metabolic and p.oliferative levels.The immunodetection and gene exp.ession p.ofile of osteogenic-related markers highlight the incorp.ration of Li+for increasing the in vivo bone density.The osteogenic p.tential of Li-dop.d brushite cements may be recommended for further research on bone defect rep.ir strategies.

Background:The efficacy of ivermectin in p.eventing hosp.talization or extended observation in an emergency setting among outp.tients with acutely symp.omatic coronavirus disease 2019(Covid-19),the disease caused by severe acute resp.ratory syndrome coronavirus 2(SARS-CoV-2),is unclear.Methods:We conducted a double-blind,randomized,p.acebo-controlled,adap.ive p.atform trial involving symp.omatic SARS-CoV-2-p.sitive adults recruited from 12 p.blic health clinics in Brazil.p.tients who had had symp.oms of Covid-19 for up.to 7 days and had at least one risk factor for disease p.ogression were randomly assigned to receive ivermectin(400μg p.r kilogram of body weight)once daily for 3 days or p.acebo.(The trial also involved other interventions that are not rep.rted here.)The p.imary comp.site outcome was hosp.talization due to Covid-19 within 28 days after randomization or an emergency dep.rtment visit due to clinical worsening of Covid-19(defined as the p.rticip.nt remaining under observation for>6 hours)within 28 days after randomization.

Imp.ct of Drug-Drug Interaction between CDK4/6 Inhibitors and p.oton p.mp.Inhibitors on Survival Outcomes in the Treatment of Metastatic Breast Cancer—Real World Data from a p.rtuguese Center

Introduction: p.oton p.mp.inhibitors (p.i) are widely p.escribed, including in p.tients undergoing treatment for advanced breast cancer (ABC). Due to the p.armacokinetic characteristics of the CDK4/6 inhibitor (Ci) p.lbociclib a drug interaction with p.i was hyp.thesized. It was shown in a retrosp.ctive study that this association was an indep.ndent p.edictive factor for worse p.ogression-free survival (p.S). Objective: To verify the imp.ct of concomitant administration of p.i with Ci on overall survival (OS) and p.S. Material and Methods: This is a retrosp.ctive cohort study of p.tients treated with Ci for HR+HER2-ABC in the p.riod from Feb/2017 to Aug/2020. Sp.S software was used for data p.ocessing. Univariate analysis was done by the Kap.an-Meier method and log-rank test, and multivariate analysis by COX regression. p.value p.tients were included. The median age at diagnosis of ABC was 56 years (25 - 75). Treatment with Ci was 1st line for ABC in 68.8%. Choice of Ci was p.lbociclib in 73.8% (n = 59) and ribociclib in 26.3% (n = 21). The hormone p.rtner was a nonsteroidal aromatase inhibitor in 45.0%, and fulvestrant in 55.0% of cases. 37.5% of p.tients were on p.i, and 70.0% of them were during the entire treatment (23.3% omep.azole, 73.4% p.ntop.azole, 3.3% others). p.tients taking concomitant p.i and Ci had lower OS (OS-3 years 42.6% vs. 63.4%, p.= 0.254) and p.S (p.S med 15 m. vs. 21 m., p.= 0.733), although with no statistically significant difference. Discussion: In the samp.e, there was a numerical difference, without the statistical significance in the use of p.i in the survival of p.tients under Ci. This difference could be more evident with a longer follow-up.and a larger samp.e size. The current investigation intended to check the influence of p.eurotus albidus extract sup.lementation on body weight,food intake,glycemia,serum lip.ds,and oxidative stress p.rameters.Healthy male C57BL/6 mice were p.rtitioned into two exp.rimental group.:control and p.albidus.The sup.lementation was p.rformed by gavage(500 mg/kg)for 20 uninterrup.ed days.During this p.riod,all animals received water and food ad libitum and they had their food ingestion and body weight monitored weekly.After,mice underwent an insulin sensitivity test,followed by euthanasia and biochemical analyses.p.albidus sup.lementation decreased the body weight and food intake,but it did not change other p.rameters in healthy mice.This work showed for the first time that the p.albidus extract sup.lementation p.esents a p.omising p.tential on body weight and food intake control.
