Neurodegenerative and neuropsychiatric diseases are increasingly affecting individuals’quality of life,thus increasing their cost to social and health *** diseases have overlapping mechanisms,such as oxidative stress,protein aggregation,neuroinflammation,neurotransmission impairment,mitochondrial dysfunction,and ***,there is no cure for neurodegenerative diseases,and the available therapies have adverse effects and low *** neuropsychiatric disorders,such as depression,the current therapies are not adequate to one-third of the patients,the so-called treatmentresistant ***,searching for new treatments is *** plants appear as a strong alternative and complement towards new treatment protocols,as they have been used for health purposes for thousands of ***,the main goal of this review is to revisit the neuroprotective potential of some of the most predominant medicinal plants(and one fungus)used in traditional Chinese medicine(TCM),focusing on their proven mechanisms of action and their chemical compositions,to give clues on how they can be useful against neurodegeneration progression.

目的探究黄花菜醇提物(Hemerocallis citrina Baroni,HCE)在利血平(reserpine)诱导的斑马鱼幼鱼(Danio rerio)模型中对抑郁行为的改善作用。方法将斑马鱼幼鱼分为对照(Con)组、利血平组、氟西汀组、黄花菜低剂量组(1.5 mg/L)、黄花菜中剂量组(3 mg/L)、黄花菜高剂量组(4.5 mg/L)。采用斑马鱼声音刺激和光刺激进行抑郁样行为学分析。利用RT-qPCR研究黄花菜醇提物对抑郁症有关的星状胶质细胞标志物及其衍生物基因(GFAP、C3、C4B、EMP-1和S100α-10)以及神经营养因子(BDNF)及其自身受体转录基因(P75、TrkB)的调节功能。结果与对照组相比,模型组在440 Hz声音刺激与8000 Lux光照刺激下运动距离缩短、运动时间减少(P<0.05,P<0.001,P<0.0001),呈现运动的不敏感性。而经过黄花菜醇提物给药后的斑马鱼幼鱼对于光与声音的刺激敏感度相较于模型组有明显升高(P<0.05,P<0.0001)。与对照组相比,利血平组斑马鱼脑中的星形胶质细胞标志物基因表达上调(P<0.0001),而各给药组相较于利血平组,星形胶质细胞标志物表达显著下调(P<0.0001),神经营养因子及其受体基因表达与对照组相比,模型组斑马鱼脑中的BDNF、P75、TrkB下调(P<0.0001),而黄花菜醇提物给药组相较于模型组BDNF、P75、TrkB表达显著上调(P<0.01,P<0.0001)。表明黄花菜醇提物抑制了星形胶质细胞激活诱发的炎症表现,同时促进了神经营养因子及其受体基因的生成,对抑郁起到了改善作用。结论黄花菜醇提物可以改善利血平诱导的斑马鱼幼鱼的抑郁样行为变化,斑马鱼脑中星形胶质细胞标志物的表达量减少,同时促进神经营养因子及其受体基因的生成,起到抗抑郁作用。

目的通过网络药理学技术预测酸枣仁复方(Ziziphi spinosae semen formula, ZSSF)对于抑郁症的关键作用靶点,并通过利血平诱导的抑郁斑马鱼模型验证酸枣仁复方治疗抑郁症的作用机制。方法通过TCMSP数据库检索酸枣仁复方的药物作用靶点,利用UniProt数据库对靶点名称进行校正。采用GeneCards、OMIM、NCBI数据库检索抑郁症相关靶点,并对3个数据库的靶点结果进行汇总去比重。利用STRING数据库预测了交集靶点的蛋白质-蛋白质相互作用(protein-protein interaction, PPI)信息,利用Metascape数据库构建进行富集研究,并利用微生信对KEGG和GO的富集结果实现了可视化。通过利血平诱导的斑马鱼抑郁模型进行行为学实验和RT-qPCR实验,验证酸枣仁复方对抑郁症的治疗作用。结果筛选出抑郁症与酸枣仁复方的交集靶点188个,蛋白质-蛋白质相互作用结果显示,酸枣仁复方抗抑郁主要作用于肿瘤坏死因子-α(TNF-α)、血清白细胞介素2(IL-2)、血清白细胞介素6(IL-6)、血清白细胞介素1β(IL-1β)、血清白细胞介素10(IL-10)等靶点,KEGG通路富集分析表明,酸枣仁复方通过TNF信号通路、PI3K-Akt信号通路、cGMP-PKG信号通路等多种信号通路发挥其治疗抑郁症的作用。动物实验结果显示,与利血平组相比,酸枣仁复方高、中、低剂量组斑马鱼在声光刺激下的运动距离显著延长(P<0.05),运动速度显著增快(P<0.01)。RT-qPCR结果显示,与利血平组相比,酸枣仁复方高、中、低剂量给药组斑马鱼脑组织中TNF-α、IL-2、IL-6、IL-1β、IL-10 mRNA表达水平上调(P<0.001)。结论酸枣仁复方通过多成分、多靶点发挥抗抑郁作用,且其抗抑郁作用可能与抑制炎症因子的表达有关。

Tunnel face reliability analysis using active learning Kriging model——Case of a two-layer soils

This paper is devoted to the probabilistic stability analysis of a tunnel face excavated in a two-layer soil. The interface of the soil layers is assumed to be positioned above the tunnel roof. In the framework of limit analysis, a rotational failure mechanism is adopted to describe the face failure considering different shear strength parameters in the two layers. The surrogate Kriging model is introduced to replace the actual performance function to perform a Monte Carlo simulation. An active learning function is used to train the Kriging model which can ensure an efficient tunnel face failure probability prediction without loss of accuracy. The deterministic stability analysis is given to validate the proposed tunnel face failure model. Subsequently, the number of initial sampling points, the correlation coefficient, the distribution type and the coefficient of variability of random variables are discussed to show their influences on the failure probability. The proposed approach is an advisable alternative for the tunnel face stability assessment and can provide guidance for tunnel design.

Background: Osteoarthritis (OA) is a disabling disease that can affect 6% to 12% of the adult population and more than a third of people over 65 years of age. Purpose: To assess whether a group of people with hand osteoarthritis (hOA) who received different types of treatment improved their function after two years of follow-up. Method: The entire sample (n = 97) underwent three follow-up assessments regarding anthropometric parameters of the upper limbs and ability to perform functional activities. Subsequently, the sample was divided into two groups for the intervention periods, called the First Period (n = 73) and the Second Period (n = 24);the First Period kept the same protocol with orientations, and the Second Period went to an intervention with orientation strength exercises and use of orthosis. Findings: In the separate analysis of the three questions of the DASH pain module, no differences were found between the assessment moments for groups of guidelines, treatment, or symptoms. Significant effects were observed for F(2, 162) = 3.5, p = 0.033, η2 = 0.04, and interaction for moments and intervention F(2, 162) = 4.3, p = 0.016, η2 = 0.05. Implications: It can be concluded that only guidance treatment does not benefit patients with hand osteoarthritis. In contrast, guidance, exercise, and orthosis treatment can significantly improve the disease.
