Ascorbic acid and prunasin,two candidate biomarkers for endodormancy release in almond flower buds identified by a nontargeted metabolomic study

Temperate fruit trees belonging to Prunus species have the ability to suspend(induce dormancy)and resume growth periodically in response to environmental and seasonal *** release requires the long-term accumulation of *** accumulation of cultivar-specific chill requirements,plants enter the state of ecodormancy,which means the ability to grow has been restored,depending on the fulfilment of heat *** many different metabolic pathways are implicated in endodormancy release,we have performed a metabolomic analysis,using the ultra-high-performance liquid chromatography–quadrupole time-of-flying(UPLC–QToF)*** assayed flower buds in different stages of endodormancy in four almond cultivars with different flowering times:the extra-early Desmayo Largueta,the late Antoñeta,the extra-late Penta,and the ultra-late *** orthogonal projection to latent-structure discriminant-analysis model was created to observe differences between endodormant and ecodormant flower *** metabolites showing the most significant variation were searched against the Metlin,HMDB,and KEGG libraries,which allowed us to identify 87 *** metabolites were subsequently assigned to specific pathways,such as abscisic acid biosynthesis,phenylpropanoid biosynthesis,and D-sorbitol metabolism,among *** two metabolites that exhibited the most significant variations in all the cultivars studied with fold changes of up to 6.49 were ascorbic acid and *** the first time,these two metabolites have been proposed as potential biomarkers for endodormancy release in *** the high synteny present between the Rosaceae species,these results could be extrapolated to other important crops like peach,plum,cherry,or apricot,among others.

Tillage practices on sloping ground often result in unsustainable soil losses impairing soil functions such as crop productivity,water and nutrients storage,and soil organic carbon(sOC)sequestration.A sloping olive grove(10%)was planted in shallow gypsiferous soils in *** was managed by minimum tillage;the most frequent management practice in central *** consequences of erosion were studied in soil samples(at 0-10,10-20,and 20-30 cm depths)by analyzing sOC,available water and gypsum content,and by detecting spectral signatures using an AsD FieldspecPro(R)VIs/*** Brightness index(BI),shape index(FI),and Normalized Difference Vegetation Index(NDVI)were derived from the AsD spectral signatures and from remote sensing(sentinel-2 image)*** development of these young olive trees was estimated from the measured diameter of the trunks(17±18 cm diameter).In 20-30 cm of the soil,the carbon stock(38±18 Mg ha-1)as well as the available water content(12±6%)was scarce,affecting the productivity of the olive *** above-mentioned indices obtained from the laboratory samples and the pixels of the sentinel-2 image were significantly(ps related to the gypsum content and the slope of the *** FI was related to the carbon and water *** NDVI derived from the satellite image identified the influence of soil degradation on the trees and the carbon *** spatialtemporal changes of the indices might help in tracking soil changes over time.

Acetone–butanol–ethanol(ABE)fermentation process can be exploited for the generation of butanol as biofuel,however it does need to overcome its low volumetric solvent productivity before it can commercially compete with fossil fuel *** this regard,mathematical modelling and simulation analysis are tools that can serve as the base for process engineering development of biological *** this work,a novel phenomenological kinetic model of Clostridium acetobutylicum ATCC 824 was considered as a benchmark system to evaluate the behaviour of an ABE fermentation under different process configurations using both free and immobilized cells:single stage batch operation,fed-batch,single stage Continuous stirred Tank Reactor(CsTR)and multistage CsTRs with and without biomass *** proposed model achieved a linear correlation index r^2=0.9952 and r^2=0.9710 over experimental data for free and immobilized cells *** predicted maximum butanol concentration and productivity obtained were 13.08 g·L^(-1)and 1.9620 g·L^(-1)·h^(-1)respectively,which represents an increase of 1.01%and 990%versus the currently developed industrial scale process reported currently into the *** results provide a reliable platform for the design and optimization of the ABE fermentation system and showcase the adequate predictive nature of the proposed model.

AMP-activated kinase （AMPK）, a protein that regulates energy balance and metabolism, has recently been identified in boar spermatozoa where regulates key functional sperm processes essential for fertilization. This work＇s aims are AMPK identification, intracellular localization, and their role in human spermatozoa function. semen was obtained from healthy human donors. sperm AMPK and phospho-Thr172-AMPK were analyzed by Western blotting and indirect immunofluorescence. High- and low-quality sperm populations were separated by a 40%-80% density gradient. Human spermatozoa motility was evaluated by an Integrated semen Analysis system （IsAs） in the presence or absence of the AMPK inhibitor compound C （CC）. AMPK is localized along the human spermatozoa, at the entire acrosome, midpiece and tail with variable intensity, whereas its active form, phospho-Thr172-AMPK, shows a prominent staining at the acrosome and sperm tail with a weaker staining in the midpiece and the postacrosomal region. Interestingly, spermatozoa bearing an excess residual cytoplasm show strong AMPK staining in this subcellular compartment. Both AMPK and phospho-Thr172-AMPK human spermatozoa contents exhibit important individual variations. Moreover, active AMPK is predominant in the high motility sperm population, where shows a stronger intensity compared with the low motility sperm population. Inhibition of AMPK activity in human spermatozoa by CC treatment leads to a significant reduction in any sperm motility parameter analyzed： percent of motile sperm, sperm velocities, progressivity, and other motility coefficients. This work identifies and points out AMPK as a new molecular mechanism involved in human spermatozoa motility. Further AMPK implications in the clinical efficiency of assisted reproduction and in other reproductive areas need to be studied.

Mos_(2) is a highly promising material for application in lithium-ion battery anodes due to its high theoretical capacity and low ***,problems with a fast capacity decay over cycling,especially at the first cycles,and poor rate performance have deterred its practical ***,electrodes comprised solely of few-layers 2D Mos_(2) nanosheets have been manufactured by scalable liquid-phase exfoliation and spray deposition *** long-standing controversy questioning the reversibility of conversion processes of Mos_(2)-based electrodes was *** studies revealed that,in 2D Mos_(2) electrodes,conversion processes are indeed reversible,where nanostructure played a key *** of the electrodes at high current rates revealed an intriguing phenomenon consisting of a continuously increasing capacity after ca.100-200 *** phenomenon was comprehensively addressed by a variety of electrochemical and microscopy methods that revealed underlying physical activation mechanisms that involved a range of profound electrode structural *** mechanisms delivered a capacitive electrode of a superior rate performance and cycling stability,as compared to the corresponding pristine electrodes,and to Mos_(2) electrodes previously ***,we have devised a methodology to overcome the problem of cycling stability of 2D Mos_(2) ***,activation of electrodes constitutes a methodology that could be applied to enhance the energy storage performance of electrodes based on other 2D nanomaterials,or combinations thereof,strategically combining chemistries to engineer electrodes of superior energy storage properties.

Current approaches for simulating the energy performance of buildings on a large scale are limited by numerous assumptions and simplifications,which can lead to inaccurate *** new tools and procedures are emerging to improve accuracy,there remains a need for more user-friendly *** study proposes a new tool based on online maps to create the geometry of districts in a simple *** tool also enables an automatic evaluation of all buildings through dynamic hourly simulations,using a building simulation software and allowing to consider different weather *** illustrate the procedure,a district at risk of energy poverty in seville(spain)is modeled,where hourly temperature data for a whole year are available to demonstrate the need for building *** tool is used to evaluate the energy demands of the district under several retrofitting alternatives,and free-floating simulations are also performed to evaluate the improvement of thermal comfort without air-conditioning *** aim is not to discuss the actual values for this particular case,but rather to identify the correct direction for large-scale studies,so as to make them more easily ***,it may be concluded that the results provided by comprehensive tools,such as the one proposed in this study,enable easy yet accurate evaluations of buildings on a large scale with significant time savings,as well as the identification of locations where retrofitting interventions would have the greatest impact.

Continuous hydrothermal synthesis was highlighted in a recent review as an enabling technology for the production of nanoparticles. In recent years, it has been shown to be a suitable reaction medium for the synthesis of a wide range of nanomaterials. Many single and complex nanomaterials such as metals, metal oxides, doped oxides, carbonates, sulfides, hydroxides, phosphates, and metal organic frameworks can be formed using continuous hydrothermal synthesis techniques. This work presents a methodology to characterize continuous hydrothermal flow systems both experimentally and numerically, and to determine the scalability of a counter current supercritical water reactor for the large scale production （〉1,000 T-year-1） of nanomaterials. Experiments were performed using a purpose-built continuous flow rig, featuring an injection loop on a metal salt feed line, which allowed the injection of a chromophoric tracer. At the system outlet, the tracer was detected using UV/Vis absorption, which could be used to measure the residence time distribution within the reactor volume. Computational fluid dynamics （CFD） calculations were also conducted using a modeled geometry to represent the experimental apparatus. The performance of the CFD model was tested against experimental data, verifying that the CFD model accurately predicted the nucleation and growth of the nanomaterials inside the reactor.
