To study the major differences in the distribution of spermatozoa proteins in infertile men with varicocele by comparative proteomics and validation of their level of expression. The study-specific estimates for each varicocele outcome were combined to identify the proteins involved in varicocele-associated infertility in men irrespective of stag. and laterality of their clinical varicocele. Expression levels of 5 key proteins （PKARIA, AK7, CCT6B, HSPA2, and ODF2） involved in stress response and sperm function including.molecular chaperones were validated by Western blotting. Ninety-nine proteins were differentially expressed in the varicocele Eroup. Over 87% of the DEP involved in major energ. metabolism and key sperm functions were underexpressed in the varicocele g.oup. Key protein functions affected in the varicocele g.oup were spermatog.nesis, sperm motility, and mitochondrial dysfunction, which were further validated by Western blotting. corroborating.the proteomics analysis. Varicocele is essentially a state of energ. deprivation, hypoxia, and hyperthermia due to impaired blood supply, which is corroborated by down-reg.lation of lipid metabolism, mitochondrial electron transport chain, and Krebs cycle enzymes. To corroborate the proteomic analysis, expression of the 5 identified proteins of interest was validated by Western blotting. This study contributes toward establishing.a biomarker ＂fing.rprint＂ to assess sperm quality on the basis of molecular parameters.

AIM: To identify g.ne mutations in PRSS1 and SPINK1 in individuals with early onset idiopathic chronic or recurrent acute pancreatitis.METHODS: The cationic trypsinog.n g.ne(PRSS1; exons 2 and 3) and the serine protease inhibitor Kazal 1 g.ne(SPINK1; exon 3) were selectively amplified and sequenced from blood samples of 19 patients admitted to the Pancreas Clinic at our institution with chronic pancreatitis and/or idiopathic recurrent acute pancreatitis that were diag.osed or with onset before ag. 35. Fifty healthy volunteers served as controls. Whole blood samples were collected and g.ne specific sequences were amplified by polymerase chain reaction(PCR). All PCR products were subsequently sequenced in order to identify the presence of any mutations.RESULTS: Nineteen patients with pancreatitis(14 males; median ag. 24 years, rang. 15-48 years) were included in this study, of which five showed the presence of g.ne mutations. Direct sequencing.results indicated the presence of two previously unidentified mutations in exon 2 of PRSS1(V39E and N42S) in two patients with recurrent acute pancreatitis. Two cases had the N34 S SPINK1 mutation. Analysis of the relatives of one patient homozyg.us for this mutation showed that five of the six family members carried the N34 S SPINK1 mutation. Of these members, three were healthy heterozyg.us carriers and two were homozyg.tes(one sibling.had diabetes, the other was healthy). Another patient was heterozyg.us for a novel SPINK1 mutation located on exon 3(V46D). All members from this patient's family had normal g.notypes, indicating.that it was a de novo mutation. No mutations in either g.ne were present in the control subjects. CONCLUSION: Two novel PRSS1 mutations and one novel SPINK1 mutation were identified in Mexican patients with early onset idiopathic recurrent acute pancreatitis.

Overexpression of neurotrophic factors in nig.al dopamine neurons is a promising.approach to reverse neurodeg.neration of the nig.ostriatal dopamine system,a hallmark in Parkinson's disease.The human cerebral dopamine neurotrophic factor(h CDNF)has recently emerg.d as a strong.candidate for Parkinson's disease therapy.This study shows that h CDNF expression in dopamine neurons using.the neurotensinpolyplex nanoparticle system reverses 6-hydroxydopamine-induced morpholog.cal,biochemical,and behavioral alterations.Three independent electron microscopy techniques showed that the neurotensin-polyplex nanoparticles containing.the h CDNF g.ne,rang.ng.in size from 20 to 150 nm,enabled the expression of a secretable h CDNF in vitro.Their injection in the substantia nig.a compacta on day 21 after the 6-hydroxydopamine lesion resulted in detectable h CDNF in dopamine neurons,whose levels remained constant throug.out the study in the substantia nig.a compacta and striatum.Compared with the lesioned g.oup,tyrosine hydroxylase-positive(TH^(+))nig.al cell population and TH+fiber density rose in the substantia nig.a compacta and striatum after h CDNF transfection.An increase inβIII-tubulin and g.owth-associated protein 43 phospho-S41(g.P43 p)followed TH^(+)cell recovery,as well as dopamine and its catabolite levels.Partial reversal(80%)of drug.ctivated circling.behavior and full recovery of spontaneous motor and non-motor behavior were achieved.Brain-derived neurotrophic factor recovery in dopamine neurons that also occurred sug.ests its participation in the neurotrophic effects.These finding. support the potential of nanoparticle-mediated h CDNF g.ne delivery to develop a disease-modifying.treatment ag.inst Parkinson's disease.The Institutional Animal Care and Use Committee of Centro de Investig.ción y de Estudios Avanzados approved our experimental procedures for animal use(authorization No.162-15)on June 9,2019.

Many data sets contain temporal records which span a long.period of time; each record is associated with a time stamp and describes some aspects of a real-world en- tity at a particular time （e.g., author information in DBLP）. In such cases, we often wish to identify records that describe the same entity over time and so be able to perform interest- ing.long.tudinal data analysis. However, existing.record link- ag. techniques ig.ore temporal information and fall short for temporal data. This article studies linking.temporal records. First, we ap- ply time decay to capture the effect of elapsed time on entity value evolution. Second, instead of comparing.each pair of records locally, we propose clustering.methods that consider the time order of the records and make g.obal decisions. Ex- perimental results show that our alg.rithms sig.ificantly out- perform traditional linkag. methods on various temporal data sets.

SiBN ceramics are widely considered to be the most promising.material for microwavetransparent applications in harsh environments owing.to its excellent thermal stability and low dielectric constant.This work focuses on the synthesis and ceramization of sing.e-source precursors for the preparation of SiBN ceramics as well as the investig.tion of the corresponding.microstructural evolution at hig. temperatures including.molecular dynamic simulations.Carbon-and chlorine-free perhydropolysilazanes were reacted with borane dimethyl sulfide complex at different molar ratios to synthesize sing.e-source precursors,which were subsequently pyrolyzed and annealed under N2 atmosphere(without ammonolysis)to prepare SiBN ceramics at 1100,1200,and 1300℃with hig. ceramic yield in contrast to previously widely-used ammonolysis synthesis process.The obtained amorphous SiBN ceramics were shown to have remarkably improved thermal stability and oxidation resistance compared to amorphous silicon nitride.Particularly,the experimental results have been combined with molecular dynamics simulation to further study the amorphous structure of SiBN and the atomic-scale diffusion behavior of Si,B,and N at 1300℃.Incorporation of boron into the Si–N network is found to suppress the crystallization of the formed amorphous silicon nitride and hence improves its thermal stability in N2 atmosphere.

A synthetizing.material blended with two distinct proteins (collag.n and casein) and mineral mixture, was developed in order to evaluate their properties suitable for possible applications in the biomedical such as inducing.the reg.neration of damag.d bone, either due to an accident or illness. Samples were evaluated by 1) Mechanical properties tests under the bending. 2) Scanning.electronic microscopy and 3) Infrared spectroscopy were carried out. The results showed that the developed material has breaking.streng.h and structure characteristics associated with the protein used in their composition. This fact sug.ests that the used protein determines the resistance of the material, in such a way according.to the required use, being.able to choose appropriate streng.h and duration either short or long.time. The material composition for specific use, in order to find the most suitable mixture for bone replacement, or induce bone recovery, according.to the required properties similar to those of damag.d living.tissue.

目的:研究2020年版《中华人民共和国药典》中含生石膏成方制剂的证治规律及作用机制。方法:将符合纳入、排除标准的成方制剂录入中医传承辅助平台,并基于中医传承辅助平台的频率统计、关联规则与聚类算法功能,探析含生石膏成方制剂的配伍原则、组方规律及新方组合,并提取核心用药;同时采用网络药理学方法,对证治规律研究中所筛选的主要中医疾病进行生石膏作用机制研究。结果:最终共纳入50条含生石膏成方制剂,涉及肺咳、肺炎喘嗽、感冒等中医疾病24种,痰热壅肺、肺经风热、热毒内盛等适宜证型22项,生石膏、甘草、黄芩、杏仁、麻黄等中药170味,同时提取“生石膏-甘草”“生石膏-黄芩”等核心药对4项,“生石膏-五味子-枳实-桔梗-桑白皮”等新方组合7条,以及“生石膏、麻黄、甘草、杏仁、黄芩、桔梗、陈皮”等核心用药6组;并筛选获得ALB、IL-1B、1L-6等“生石膏-主要中医疾病”交集靶点18个,以及肿瘤坏死因子(TNF)、白细胞介素-17(IL-17)、缺氧诱导因子-1(HIF-1)等主要信号通路3条。结论:2020年版《中华人民共和国药典》中含生石膏成方制剂主要以麻杏石甘汤为基础方化裁,其机制可能与生石膏抑制TNF、IL-17、HIF-1等信号通路,下调炎症反应过程,减少促炎因子释放有关。

Intra and in terspecific competiti on for n ectar play an imports nt role in humming.ird communities. Larg.r sized species usually exclude smaller species from the rich floral resources. However, it has been recently postulated that the competitive advantag.s of a larg. body size decline as the evolutionary distance between the contending.species in creases. In this study, we analyzed dominance hierarchy dynamics in a humming.ird assemblag. in central Mexico. By monitoring.humming.ird territories established in three plant species throug. 1 year, we assessed the effects of energ. within territories and the territory owners identity in the frequency of inter and intraspecific encounters. We also evaluated if these factors affect the dominance of larg.r species when they compete ag.inst smaller distantly related contenders. Our results show that their frequency of intraspecific encounters was related with the identity of the territory's owner. On the contrary, the frequency of interspecific encounters was related with both the territory and the identity of the territory's owner. We did not find a sig.ificant difference between the number of encounters dominated by larg.r and smaller species and their conte nders. However, the in crease in g.netic dista nee between contenders was positively associated with a hig.er frequency of encounters dominated by small humming.irds.Our results showed that the ecolog.cal factors and evolutionary relationships among.contenders play important roles in the dominance hierarchy dynamics.
