Portable dielectrophoresis for biology:AdePT facilitates cell trapping,separation,and interactions

Dielectrophoresis is a powerful and well-established technique that allows label-free,non-invasive manipulation of cells and particles by leveraging their electrical properties.The practical implementation of the associated electronics and user interface in a biology laboratory,however,requires an engineering background,thus hindering the broader adoption of the technique.In order to address these challenges and to bridge the gap between biologists and the engineering skills required for the implementation of deP platforms,we report here a custom-built,compact,universal electronic platform termed AdePT(adaptable dielectrophoresis embedded platform tool)for use with a simple microfluidic chip containing six microelectrodes.The versatility of the open-source platform is ensured by a custom-developed graphical user interface that permits simple reconfiguration of the control signals to address a wide-range of specific applications:(i)precision positioning of the single bacterium/cell/particle in the micrometer range;(ii)viability-based separation by achieving a 94%efficiency in separating live and dead yeast;(iii)phenotype-based separation by achieving a 96%efficiency in separating yeast and Bacillus subtilis;(iv)cell–cell interactions by steering a phagocytosis process where a granulocyte engulfs E.coli RGB-S bacterium.Together,the set of experiments and the platform form a complete basis for a wide range of possible applications addressing various biological questions exploiting the plug-and-play design and the intuitive GUI of AdePT.

Seed Dispersal of Citharexylum tetramerum and Ziziphus penduculata by Carnivorous in a Xerophilous Scrub at Tehuacan, Puebla, Mexico

Seed dispersion reduces the depredation rate and increases genetic flow. Some species of Carnivora consume fruits as a standard component of their diet, so they become potential seed dispersers. In Mexico, a few studies evaluated carnivorous as seed dispersers, especially in dry ecosystems. Citharexylum tetramerum and Ziziphus pedunculata are endemic plants from the Tehuacán-Cuicatlán’s Biosphere Reserve (TCBR);however, there are very little data about dispersal seeds for both species. We assessed the germination of seeds ingested by carnivores obtained from feces. We compared them with seeds from fruits as a control group. At the Metropolitan University laboratory, we germinated seeds, from: plants, scats, and fruits, and we used Kruskall Wallis Test to compare percentage and germination rate. C. tetramerum seeds ingested by a carnivorous decreased in germination percentage and rate. In contrast, the percentage and rate of germination of Z. pedunculata seeds from carnivorous feces were higher than the control group. However, Z. pedunculata control seeds did not germinate during tests, but at the end of the tests, we found that 90% of control seeds were not dead, so they were dormant seeds. Carnivores interact as legitime dispersers for both plants because they are keeping the viability of seeds. Still, we only showed that Z. pedunculata gut carnivores broke with the dormancy of seeds. We consider that in future research, it could be essential to identify other animals that consume fruits of those plants and if it is possible to follow seeds after they have been removed. Those points could help to get better understanding of those endemic plants from Tehuacán.

Identification of Rhizobia Isolated from Nodules of Mexican Commercial Soybean Varieties

Rhizobia, crucial for nitrogen fixation in leguminous plants, play a vital role in soybean cultivation. This study, conducted in Mexico, a major soybean importer, aimed to identify bacteria from nodules of five soybean varieties in high-production regions. Multilocus sequence analysis (MlSA) was employed for enhanced species resolution. The study identified six Bradyrhizobium species: Bradyrhizobium japonicum USDA 110, Bradyrhizobium japonicum USDA 6, Bradyrhizobium elkanii USDA 76, Bradyrhizobium neotropicale, Bradyrhizobium lablabi, and Bradyrhizobium icense. Bradyrhizobium japonicum USDA 110 predominated in the soils, displaying symbiotic preference for the Huasteca 400 variety. However, phylogenetic analysis didn't reveal a clear association between strains, soil, and soybean variety. This research sheds light on the diversity of rhizobia in Mexican soybean cultivation, contributing to the understanding of symbiotic relationships in soybean production systems.

The dorsal root ganglion as a target for neurorestoration in neuropathic pain

Neuropathic pain is a severe and chronic condition widely found in the general population.The reason for this is the extensive variety of damage or diseases that can spark this unpleasant constant feeling in patients.During the processing of pain,the dorsal root ganglia constitute an important region where dorsal root ganglion neurons play a crucial role in the transmission and propagation of sensory electrical stimulation.Furthermore,the dorsal root ganglia have recently exhibited a regenerative capacity that should not be neglected in the understanding of the development and resolution of neuropathic pain and in the elucidation of innovative therapies.Here,we will review the complex interplay between cells(satellite glial cells and inflammatory cells)and factors(cytokines,neurotrophic factors and genetic factors)that takes place within the dorsal root ganglia and accounts for the generation of the aberrant excitation of primary sensory neurons occurring in neuropathic pain.More importantly,we will summarize an updated view of the current pharmacologic and nonpharmacologic therapies targeting the dorsal root ganglia for the treatment of neuropathic pain.

Parkinsonism by unilateral,intranigralβ-sitosterolβ-D-glucoside administration in rats is distinguished in that theα-synuclein insult begins unilaterally but spreads bilaterally and increases in severity over time,thus replicating several clinical features of Parkinson’s disease,a typicalα-synucleinopathy.As Nurr1 repressesα-synuclein,we evaluated whether unilateral transfected of rNurr1-V5 transgene via neurotensin-polyplex to the substantia nigra on day 30 after unilateralβ-sitosterolβ-D-glucoside lesion could affect bilateral neuropathology and sensorimotor deficits on day 30 post-transfection.This study found that rNurr1-V5 expression but not that of the green fluorescent protein(the negative control)reducedβ-sitosterolβ-D-glucoside-induced neuropathology.Accordingly,a bilateral increase in tyrosine hydroxylase-positive cells and arborization occurred in the substantia nigra and increased tyrosine hydroxylase-positive ramifications in the striatum.In addition,tyrosine hydroxylase-positive cells displayed less senescence markerβ-galactosidase and more neuron-cytoskeleton markerβIII-tubulin and brain-derived neurotrophic factor.A significant decrease in activated microglia(positive to ionized calcium-binding adaptor molecule 1)and neurotoxic astrocytes(positive to glial fibrillary acidic protein and complement component 3)and increased neurotrophic astrocytes(positive to glial fibrillary acidic protein and S100 calcium-binding protein A10)also occurred in the substantia nigra.These effects followed the bilateral reduction inα-synuclein aggregates in the nigrostriatal system,improving sensorimotor behavior.Our results show that unilateral rNurr1-V5 transgene expression in nigral dopaminergic neurons mitigates bilateral neurodegeneration(senescence and loss of neuron-cytoskeleton and tyrosine hydroxylase-positive cells),neuroinflammation(activated microglia,neurotoxic astrocytes),α-synuclein aggregation,and sensorimotor deficits.Increased neurotrophic astrocytes and brain-derived neurotrophic factor can mediate t

Origin of Traditional Chinese Medicine in Cuba in the 19th Century from Its Main Exponents and Some Notable Medical descendants in the 20th Century

The Cuban people are made up of three major migratory currents, the Chinese are one of them. They brought their culture, the methods, and procedures of traditional Chinese medicine(TCM) in the 19th century. Few were able to return and so they created families in Cuba;some of their descendants dedicated themselves to medicine. In order to investigate the practices that were predecessors of TCM in Cuba in the 19th century, a qualitative phenomenological research was carried out, reviewing what was published by various sources, applying documentary analysis, logical historical analysis, abstraction, synthesis, and systematization of the results on the regularities of the work and human behavior of Chinese doctors in the Cuban 19th and 20th centuries. This made it possible to identify six Chinese doctors in the 19th century in Cuba who gave rise to the beginning of some practices of TCM in Cuba, and five from the 20th century, descendants of coolies who dedicated themselves to other specialties of medicine. It was found that despite their geographical and time disperse, they were all notorious for their outstanding professional and human behavior, with a trail of accumulated successes in achieving “almost the impossible” with the patient. They have left their mark on Cuban culture.

BACKGROUND Dietary methyl donors might influence DNA methylation during carcinogenesis of colorectal cancer(CRC).However,whether the influence of methyl donor intake is modified by polymorphisms in such epigenetic regulators is still unclear.AIM To improve the current understanding of the molecular basis of CRC.METHODS A literature search in the Medline database,Reference Citation Analysis(https://***/),and manual reference screening were performed to identify observational studies published from inception to May 2022.RESUlTS A total of fourteen case-control studies and five cohort studies were identified.These studies included information on dietary methyl donors,dietary components that potentially modulate the bioavailability of methyl groups,genetic variants of methyl metabolizing enzymes,and/or markers of CpG island methylator phenotype and/or microsatellite instability,and their possible interactions on CRC risk.CONClUSION Several studies have suggested interactions between methylenetetrahydrofolate reductase polymorphisms,methyl donor nutrients(such as folate)and alcohol on CRC risk.Moreover,vitamin B6,niacin,and alcohol may affect CRC risk through not only genetic but also epigenetic regulation.Identification of specific mechanisms in these interactions associated with CRC may assist in developing targeted prevention strategies for individuals at the highest risk of developing CRC.

The Sergipano Orogenic System(SOS)in southern Borborema Province(NE Brazil)hosts voluminous Neoproterozoic plutonism related to the Brasiliano/Pan-African Orogeny.This paper presents comprehensive whole-rock geochemical data,titanite U-Pb ages,and the first combined zircon U-Pb and lu-Hf isotope results for plutonic rocks from the MacururéDomain to constrain their sources,petrogenesis,and tectonic setting.Three magmatic episodes are recognized and record the evolutionary stages of the orogen.(i)Early-collisional magmatism(643–628 Ma)comprises gabbros and diorites with minor tonalites characterized by well-developed tectonic foliation and evidence of solid-state deformation.These rocks are magnesian,high-K calc-alkaline,lIlE-and lREE-enriched and provide subchondriticε_(Hf)(t)values(6.5 to4.7)and Orosirian Hf-T^(C)_(DM)model ages(1.83–1.94 Ga).Such features indicate derivation from a lithospheric mantle source metasomatized by incorporating crustal components through subduction processes prior to magma generation,possibly related to the Rhyacian Orogeny(2.20–1.96 Ga).Extensive mixing/mingling between basaltic and crust-derived magmas took place at lower crustal depths,producing coeval hybrid diorites and quartz-diorites.(ii)Syn-collisional magmatism(630–624 Ma)encompasses biotite-and muscovite-bearing granodiorites and monzogranites,preserving their structures parallel to the schistosity of the country rocks.These rocks are leucocratic,weakly metaluminous to peraluminous,and contain abundant surmicaceous enclaves.Petrographic features and geochemical composition suggest an origin related to the partial melting of graywacke protoliths with a subordinate igneous component.(iii)late-collisional magmatism(625–600 Ma)includes undeformed and isotropic monzonites and granodiorites,which truncate the regional foliation.These rocks are consistently metaluminous and magnesian,showing affinities with the high-K calc-alkaline to shoshonite series.Trace element modeling with subchondriticε_(Hf)(t)values(8.3 to4.1)and Paleo

Effect of lead chloride on spermatogenesis and sperm parameters in mice

Aim: To evaluate the effect of acute lead chloride exposure on testis and sperm parameters in mice. Methods: PbCl2, 74 mg/kg, was daily administered to sexually mature male mice for 3 days and the effects on the testicular histology and ultrastructure as well as the motility and density of spermatozoa in cauda epididymis were observed. An additional group of mice were treated for 1-3 days and were allowed to recover for 32 days to determine the reversibility of lead-induced changes. Results: The testicular weight, seminiferous tubular diameter and sperm counts were significantly decreased following 3 days of PbCl2 treatment, but were unaffected by shorter-term exposures. The changes caused by lead are mostly reversible. Conclusion: Acute lead chloride exposure injures the fertility parameters of male mice and the effects are partially reversible.

Absence of Fruit Mexican Fly (<i>Anastrepha ludens</i>) in Pitaya de Mayo (<i>Stenocereus pruinosus</i>)

Pitaya de mayo (Stenocereus pruinosus) exportation is considered feasible especially to the United States of America (USA) using the adequate preservation and transportation techniques. One of the exportation requirements is that pitayas, as well as any other tropical fruits, have to be free from Mexican fruit fly contamination and certified by the Food National Sanitary, Iniquity, and Quality Service. This review proved that this fruit is not attacked by the Mexican fruit fly A. ludens or any other Anastrepha species. The fruit fly may be found in some States of Mexican Republic such as some tropical regions but not in the semi-arid regions where cacti fruits are grown.
