T follicular helper expansion and humoral-mediated rejection are independent of the HVEM/BTLA pathway

The molecular pathways contributing to humoral-mediated allograft rejection are poorly defined. In this study, we assessed the role of the herpesvirus entry mediator/B- and T-lymphocyte attenuator （HVEM/BTLA） signalling pathway in the context of antibody-mediated allograft rejection. An experimental setting was designed to elucidate whether the blockade of HVEM/BTLA interactions could modulate de novo induction of host antidonor-specific antibodies during the course of graft rejection. To test this hypothesis, fully allogeneic major histocompatibility complex-mismatched skin grafts were transplanted onto the right flank of recipient mice that were treated with isotype control, anti-CD40L or modulatory antibodies of the HVEM/BTLA signalling pathway. The frequencies of CD4 T follicular helper （Tfh） cells （B220-, CD4＋ CXCR5＋ PD-lhigh）, extrafollicular helper cells （B220-, CD4＋ CXCR5- PD-1＋ and PD-1-） and germinal centre （GC） B cells （B220＋Fas＋ GL7＋） were analysed by flow cytometry in draining and non-draining lymph nodes at day 10 post transplantation during the acute phase of graft rejection. The host antidonor isotype-specific humoral immune response was also assessed. Whereas blockade of the CD40/CD40L pathway was highly effective in preventing the allogeneic humoral immune response, antibody-mediated blockade of the HVEM/BTLA-interacting pathway affected neither the expansion of Tfh cells nor the expansion of GC B cells. Consequently, the course of the host antidonor antibody-mediated response proceeded normally, without detectable evidence of impaired development. In summary, these data indicate that HVEM/BTLA interactions are dispensable for the formation of de novo host antidonor isotype-specific antibodies in transplantation.

Autophagy in eukaryotic cells is a constitutive process and functions as a homeostatic mechanism; it is up-regulated in response to specific stress stimuli such as starvation, hypoxia and as oxidative stress. In addition to playing a crucial role in adaptive responses to different stimuli, autophagy is also required for intracellular quality control. This second aspect is important to prevent the activation of pathological processes. Autophagy also plays a central role in cellular development and differentiation because it is involved in the regulation of energetic balance. This final aspect is critical for maintaining proper bone and muscle function as well as to prevent any pathological changes. Therefore, identifying new molecular targets involved in autophagy is critical to assure a good quality of life.

Since the advent of sequencing technologies,the determination of microbial diversity to predict microbial functions,which are the major determinants of soil functions,has become a major topic of interest,as evidenced by the 900 publications dealing with soil metagenome published up to ***,the detection of a gene in soil does not mean that the relative function is expressed,and the presence of a particular taxon does not mean that the relative functions determined in pure culture also occur in the studied *** critical step is to link microbial community composition or function to the product analyzed to determine flux ***,flux rates might not only be highly dynamic,but several metabolites can depend on different reactions,which makes the link to one process of interest difficult or even *** review also discusses biases caused by sampling,storage of samples,DNA extraction and purification,sequencing(*** sequencing),and bioinformatic data *** and the limits of predicting microbial interactions by network inference methods are critically discussed,and finally,future directions for a better understanding of soil functions by using measurements of microbial diversity are presented.

Contribution of the ear and the flag leaf to grainfilling in durum wheat inferred from the carbon isotope signature: Genotypic and growing conditions effects

The ear, together with the flag leaf, is believed to play a major role as a source of assimilates during grain filling in C3cereals. However, the intrusive nature of most of the available methodologies prevents reaching conclusive results in this regard. This study compares the carbon isotope composition（d13C） in its natural abundance in the water‐soluble fractions of the flag leaf blade and the ear with the d13C of mature kernels to assess the relative contribution of both organs to grain filling in durum wheat（Triticum turgidum L. ***）. The relative contribution of the ear was higher in landraces compared to modern cultivars, as well as in response to nitrogen fertilization and water stress. Such genotypic and environmentally driven differences were associated with changes in harvest index（HI）, with the relative contribution of the ear being negatively associated with HI. In the case of the genotypic differences, the lower relative contribution of the ear in modern cultivars compared with landraces is probably associated with the appearance in the former of a certain amount of source limitation driven by a higher HI. In fact, the relative contribution of the ear was far more responsive to changes in HI in modern cultivars compared with landraces.

Plasma betatrophin levels in patients with liver cirrhosis

AIM: To investigate the plasma levels of betatrophin in patients with ***: Forty patients diagnosed at the clinic with liver cirrhosis according to biological, ultrasonographic,or histological criteria were *** severity of cirrhosis was classified according to Pugh's modification of Child's classification and MELD score. Insulin resistance(IR) was assessed by the Homeostasis Model Assessment. A total of 20 patients showed a MELD score higher than 14. The control group consisted in 15 sex-and aged-matched *** blood samples were obtained for subsequent analysis. Serum insulin was determined by Liaison automated immune chemiluminiscence assay(diaSorin S.p.A.) using a sandwich assay. The sensitivity of the assay was 0.2 μU/mL. The intra and interassay variation coefficients were disease severity, being higher in Child-Pugh C patients compared to Child-Pugh B(Pdisplayed a MELD score higher than 14 points compared to patients with lower punctuation(P = 0.01). In addition, we found a positive correlation between plasma betatrophin levels and the severity of cirrhosis according to Child-Pugh classification(r = 0.53; P < 0.01) or MELD score(r = 0.45; P <0.01). In the overall cohort, a moderate correlation between serum betatrophin and plasmatic bilirrubin(r= 0.39; P < 0.01) has been observed, as well as an inverse correlation between betatrophin and albumin(r =-0.41; P < 0.01) or prothrombin time(r =-0.44;P <0.01). Morphometry and subpopulation structure of Holstein bull spermatozoa： variations in ejaculates and cryopreservation straws

Sperm quality is evaluated for the calculation of sperm dosage in artificial reproductive programs. The most common parameter used is motility, but morphology has a higher potential as a predictor of genetic quality. Morphometry calculations from CASA-Morph technology improve morphological evaluation and allow mathematical approaches to the problem. Semen from 28 Holstein bulls was collected by artificial vagina, and several ejaculates were studied. After general evaluation, samples were diluted, packaged in 0.25 ml straws, and stored in liquid nitrogen. Two straws per sample were thawed, and slides were processed and stained with diff-Quik. Samples were analyzed by a CASA-Morph system for eight morphometric parameters. In addition to the ＂classical＂ statistical approach, based on variance analysis （revealing differences between animals, ejaculates, and straws）, principal component （PC） analysis showed that the variables were grouped into PC1, related to size, and PC2 to shape. Subpopulation structure analysis showed four groups, namely, big, small, short, and narrow from their dominant characteristics, representing 31.0%, 27.3%, 24.1%, and 17.7% of the total population, respectively. The distributions varied between animals and ejaculates, but between straws, there were no differences in only four animals. This modern approach of considering an ejaculate sperm population as divided into subpopulations reflecting quantifiable parameters generated by CASA-Morph systems technology opens a new view on sperm function. This is the first study applying this approach to evaluate different ejaculates and straws from the same individual. More work must be done to improve seminal dose calculations in assisted reproductive programs.

Functionalization of micropipette tips with hydrophobin-laccase chimera and application to the electrochemical determination of caffeic acid in tea samples

Performing on-site measurements is one of the main challenges of current(bio)chemical *** tips,which are among the most common utilities in laboratories,can be useful and cheap elements for designing out-of-the-box innovative analytical ***,most of the analytical properties of these bioplatforms depend on the immobilization of the recognition element(e.g.,enzymes).In this context,we have recently designed a chimeric protein in which laccase PoxA1b from the fungus Pleurotus ostreatus has been genetically fused to the class I hydrophobin *** ability to self-assemble at hydrophobic-hydrophilic interfaces of hydrophobin,allows an easy laccase *** this work,the Vmh2-PoxA1b fusion protein was immobilized into common polypropylene micropipette tips to develop an innovative enzymatic bioplatform that can produce fast results in a simple *** proof of applicability,the platform was applied to the determination of caffeic acid by enzymatic oxidation and further electrochemical reduction of the product on a screen-printed electrode,achieving a limit of detection of 1.4-10^(-6) *** was then used for determining caffeic acid in tea samples.
