Extent of liver resection modulates the activation of transcription factors and the production of cytokines involved in liver regeneration

AIM： To investigate the molecular events involved in liver regeneration following subtotal hepatectomy （SH） as previous studies have largely focused on partial hepatectomy （PH）. METHODS： Male Wistar rats were subjected to 70% PH or 90% SH, respectively, and sacrificed at different times after surgery. untreated and sham-operated animals served as controls. Serum and liver samples were obtained to investigate liver function, apoptosis （TuNEL assay） and transcription factors （NF-KB, Stat3; ELISA） or cytokines （HGF, TNF-α, IL-6, TGF-α, TGF-β; quantitative RT-PCR） involved in liver regeneration. RESuLTS： Serum levels of ALT and AST in animals with 70% PH differed significantly from sham-operated and control animals. We found that the peak concentration 12 h after surgery returned to control levels 7 d after surgery. LDH was increased only at 12 h after 70% PH compared to sham. Bilirubin showed no differences between the sham and 70% resection. After PH, early NF-kB activation was detected 12 h after surgery （313.21 ± 17.22 ng/mL）, while there was no activation after SH （125.22 ± 44.36 ng/mL） compared to controls （111.43 ± 32.68 ng/mL） at this time point. In SH, however, NF-kB activation was delayed until 24 h （475.56 ± 144.29 ng/mL）. Stat3 activation was similar in both groups. These findings correlated with suppressed and delayed induction of regenerative genes after SH （i.e. TNF-α 24 h postoperatively： 2375 ± 1220 in 70% and 88 ± 31 in 90%; IL-6 12 h postoperatively： 2547 ± 441 in 70% and 173 ± 82 in 90%）. TuNEL staining revealed elevated apoptosis rates in SH （0.44% at 24 h; 0.63% at 7 d） compared to PH （0.27% at 24 h; 0.15% at 7 d）. CONCLuSION： The molecular events involved in liver regeneration are significantly influenced by the extent of resection as SH leads to suppression and delay of liver regeneration compared to PH, which is associated with delayed activation of NF-kB and suppression of proregenerative cytokines.

Hispanic Women＇s Presence in the Actual u.S. Congress

The Hispanic population of the united States has become, in recent times, into a powerful electoral force whose vote is historically oriented towards the Democratic Party of the incumbent uS President Barack H. Obama. Obviously, this growing electoral strength is reflected into its political presence in the American life that has been accomplished with their numeric inclusion in the uS Congress. Although in the recent decades several studies have arisen on the status of the Hispanic community in the united States from both sides of the Atlantic, these have been primarily focused on casuistic, demographic, developmental or educational aspects. But only a few of them, if any, has focused exclusively on the numerical and symbolical representation of the community in the political arena, and more specifically into the main legislative body of the country, the uS Congress. And therein it lays the purpose of this article. We intend to investigate not only the historical presence of Hispanic legislators in Congress but also and especially their political influence in the current u.S. Congress. Finally, and prior to the conclusions drawn throughout the paper, we will analyze the political influence of the Hispanic congresswomen in the current 114th Congress. As we will be breaking down, this group has been changing gradually from a poor oppressed minority into one of the most dynamic groups of the country, introduced at all levels of government.

Eutrophication Levels through San Pedro-Mezquital River Basin

This paper presents an eutrophication diagnosis for the San Pedro-Mezquital basin at Mexico;procedure accounted to set up 10 sampling points distributed along the river pathway, sample collection was done at two extreme conditions: rainy season and dry season. Five parameters were analyzed in this work: pH, % Dissolved Oxygen, reactive phosphorus, and nitrogen species as nitrate and ammonia compounds. Obtained concentrations were statistically analyzed to establish if there were significant differences among sites and between seasons;being the dry season the one with higher nutrient content, ammonia can be considered statistical significance due to the total values found for each season. Calculation of the Karydis eutrophication index for each parameter showed that more damaged sites were those nearby the cities, and most of the sampled sites belonged to the eutrophic category.

The opening of maitotoxin-sensitive calcium channels induces the acrosome reaction in human spermatozoa： differences from the zona pellucida

The acrosome reaction （AR）, an absolute requirement for spermatozoa and egg fusion, requires the influx of Ca2＋ into the spermatozoa through voltage-dependent Ca2＋ channels and store-operated channels. Maitotoxin （MTx）, a Ca2＋-mobilizing agent, has been shown to be a potent inducer of the mouse sperm AR, with a pharmacology similar to that of the zona pellucida （ZP）, possibly suggesting a common pathway for both inducers. using recombinant human ZP3 （rhZP3）, mouse ZP and two MTx channel blockers （u73122 and u73343）, we investigated and compared the MTx- and ZP-induced ARs in human and mouse spermatozoa. Herein, we report that MTx induced AR and elevated intracellular Ca2＋（[Ca2＋]~） in human spermatozoa, both of which were blocked by u73122 and u73343. These two compounds also inhibited the MTx-induced AR in mouse spermatozoa. In disagreement with our previous proposal, the AR triggered by rhZP3 or mouse ZP was not blocked by u73343, indicating that in human and mouse spermatozoa, the AR induction by the physiological ligands or by MTx occurred through distinct pathways. u73122, but not u73343 （inactive analogue）, can block phospholipase C （PLC）. Another PLC inhibitor, edelfosine, also blocked the rhZP3- and ZP-induced ARs. These findings confirmed the participation of a PLC-dependent signalling pathway in human and mouse zona protein-induced AR. Notably, edelfosine also inhibited the MTx-induced mouse sperm AR but not that of the human, suggesting that toxin-induced AR is PLC-dependent in mice and PLC-independent in humans.

Diversity of Filamentous Fungi of Area from Brazilian Caatinga and High-Level Tannase Production using Mango (<i>Mangifera indica</i>L.) and Surinam Cherry (<i>Eugenia uniflora</i>L.) Leaves under SSF

Tannase is a biotechnologically important enzyme that can be produced during fungal fermentation of organic matter. The Caatinga is an exclusive Brazilian ecosystem that has been largely unexplored by science, particularly its filamentous fungal diversity. This study evaluated the diversity of filamentous fungi in the Caatinga soils of Pernambuco, Brazil, and their potential for tannase production by solid-state fermentation (SSF) of mango (Mangifera indica L.) and Surinam cherry (Eugenia uniflora L.) leaves. A total of 4711 isolates were obtained, 2090 during the rainy seasonand 2621 during the dry season. The isolates belonged to 18 genera and 66 species, with Aspergillus and Penicillium having the highest species richness. The dry season had a higher diversity index. Aspergillus was the dominant genus, and A. flavus, A. sclerotiorum, and A. ochraceus the most abundant species. A representative of each species was tested for tannase production using dried mango and Surinam cherry leaves as substrates;the leaves contained 14.28 and 7.0 g/L tannin, respectively. Most fungal species produced tannase, but the highest yields were obtained when mango leaves were used as substrate for Penicillium restrictum (accession uRM 6044), Aspergillus flavofurcatus (uRM 6142), and A. stromatoides (uRM 6609), which produced 104.16, 87.51, and 81.83 u/mL tannase, respectively. These yields exceeded previously published reports. Filamentous fungi from Caatinga soils have great potential for producing tannase by SSF, and low-cost mango leaves make excellent substrate.
