Petiveria alliacea Suppresses Airway Inflammation and Allergen-Specific Th2 Responses in Ovalbumin-Sensitized Murine Model of Asthma

Objective： To examine the effect of metanol extract of Petiveria alliacea （PM） on airway inflammation in a murine model of chronic asthma. Methods： Two-month-old male BALB/c mice （n=6-8/group） were sensitized on days 0 and 14 by intraperitoneal injection of 20μg ovalbumin （OVA）. On day 25, the mice received an airway challenge with OVA （3%, w/v, in phosphate buffered saline）. PM was administered orally by oral gavage to mice at doses of 100, 200 and 400 mg/kg body weight once daily from days 18 to 23. Control mice were orally administered phosphate buffered saline （PBS） to induce a model of asthma. At the end of the test, respiratory reactivity was assayed, the total cell number, interleukin-4 （IL-4）, IL-5, IL-13, tumor necrosis factor-alpha （TNF-α） and reactive oxygen species （ROS） in the bronchoalveolar lavage fluid （BALF） were determined and the levels of serum IgE, intercellular cell adhesion molecule 1 （ICAM-1） and eotoxin were measured. In addition, lung tissue was used to qualify the IL-4, IL-5, IL-13, TNF-α and transforming growth factor beta 1 （TGF-β1）. Histologic examination was performed to observe inflammatory cellular infiltration. Results： The administration of PM in comparison with the OVA-only treated group significantly attenuated the infiltration of eosinophUs and other inflammatory cells （P〈0.01）. Airway resistance （RI） in the OVA-only induced group was significantly higher than that of the PBS control group （P〈0.01） when methacholine was added. TNF-α, IgE, TGF-β1 and cytokine levels IL-4, IL-5, IL-β in the BALF decreased compared to control mice （P〈0.01 or P〈0.05）. PM treatment also inhibited the production of chemokines, eotaxin and ICAM-1 in BALF （P〈0.01）, which improved lung function. Histopathological examination revealed that the sensitized treated PM groups had significant lower in inflammatory scores similar to dexamethasone treatments and the untreated group. Segmentation of thermal infrared images of cucumber leaves using K-means clustering for estimating leaf wetness duration

Leaf wetness duration(LWD)is a critical parameter used to predict plant disease,but its determination under actual field conditions is a major *** this study,a method for determining LWD using thermal infrared imaging was developed and applied to cucumber plants grown in a solar *** images of the plant leaves were captured using an infrared scanning camera,and a leaf wetness area segmentation method consisting of two procedures was ***,a color space conversion was performed automatically by an image-processing ***,the K-means clustering algorithm was applied to enable the segmentation of the wetness area on the thermal ***,to enable overall thermal image analysis,an initial leaf wetness threshold(LWT)of 5%was defined(where wetness values higher than 5%indicated that the leaf was in a wet state).The results of comparative experiments conducted using thermal images of plant leaves captured using an infrared scanning camera and human visual observation indicated that the estimated LWD values were generally higher than the observed LWD values,because slight leaf wetness condensations were overlooked by the human eye but detected by the infrared scanning *** these differences were not found to be statistically significant in this study,the proposed method for determining LWD using thermal infrared imaging may provide a new LWD detection method for cucumber and other plants grown in solar greenhouses.

Therapeutic strategies for post-transplant recurrence of hepatocellular carcinoma

Function, kinetic properties, crystallization, and regulation of microbial malate dehydrogenase

Malate dehydrogenase （MDH） is an enzyme widely distributed among living organisms and is a key protein in the central oxidative pathway. It catalyzes the interconversion between malate and oxaloacetate using NAD＋ or NADP＊ as a cofactor. Surprisingly, this enzyme has been extensively studied in eukaryotes but there are few reports about this enzyme in prokaryotes. It is necessary to review the relevant information to gain a better understanding of the function of this enzyme. Our review of the data generated from studies in bacteria shows much diversity in their molecular properties, including weight, oligomeric states, cofactor and substrate binding affinities, as well as differences in the direction of the enzymatic reaction. Furthermore, due to the importance of its function, the transcription and activity of this enzyme are rigorously regulated. Crystal structures of MDH from different bacterial sources led to the identification of the regions involved in substrate and cofactor binding and the residues important for the dimer-dimer interface. This structural information allows one to make direct modifications to improve the enzyme catalysis by increasing its activity, cofactor binding capacity, substrate specificity, and thermostability. A comparative analysis of the phylogenetic reconstruction of MDH reveals interesting facts about its evolutionary history, dividing this superfamily of proteins into two principle clades and establishing relationships between MDHs from different cellular compartments from archaea, bacteria, and eukaryotes.

Mechanical Properties of Composite Mortars Using Flurogypsum and PVC Particles

The present work describes the viability of a mortar binder based on two industrial by-products: poly(vinyl chloride) (PVC) particles from scrap and anhydrite (CaSO4) from fluorgypsum. Mortar composites were made incorporating different amounts of PVC particles and cured at constant room temperature during various periods of time. From X-ray diffraction, it was possible to follow the hydration process and to estimate the effect of the PVC particles on anhydrite transformation to gypsum (CaSO4·2H2O). Compressive strength from uniaxial testing was measured from stress-strain curves carried out at room temperature. According to these results, the hydration rates of the composites depend on the concentration of PVC particles and there is an enhancement in their compressive strength as particle content increases, reaching values of 36 MPa after 28 days.

Cryopreservation by Vitrification of Vitis vinifera cv. ＂Red Globe＂ Zygotic Embryos and Effect on the Expression of DNA Methyltransferase Genes

Mexico is a large producer of table grape （Vitis vinifera L.） and therefore it is important to develop protocols to store the grape varieties germplasm. The objective of the present work was to design a protocol for the cryopreservation by vitrification of zygotic embryos of V. vinifera cv. ＂Red Globe＂ and evaluate possible epigenetics changes. The plant vitrification solution 2 （PVS2） was utilized before the utilization of liquid nitrogen （LN）. The effect of this protocol on embryo viability was tested by the triphenyl-tetrazolium chloride solution, as well as by the in vitro development of grape embryos into plantlet. A cDNA expression library of grape zygotic embryos was created to isolate expressed sequence tags of several DNA methyltrasferases. Gene expression of domains rearranged methyltransferase type 1 （DMR1） and DNA （cytosine-5）-methyltransferase 1 （MET1-2） isozymes was analyzed by quantitative reverse transcriptase PCR. The optimal conditions for vitrification were 10 min in 50% PVS2, followed by I0 min in 100% PVS2. Under these conditions, about 30% of plantlet was obtained from embryos after cryopreservation. It was recorded a reduction in the MET1-2 gene expression, which plays a role in the maintenance of DNA methylation. It is possible to cryopreserve viable grape zygotic embryos, although the treatment seems to induce alterations in the normal DNA methylation pattern of the zygotic embryo genome.

Role of Transparent Exopolymer Particles on Phytoplankton Dynamics in a Subtropical Estuary,Cananeia-Iguape(Sp,Brazil)

Transparent Exopolymer Particles (TEP) has a known crucial role in vertical fluxes of carbon in the ocean and has been intensively studied in the last decade. Nevertheless, few studies have considered horizontal fluxes of TEP. These fluxes arise prominently between estuaries and the coast, where its importance is associated to the predominant horizontal transport. This is due both to the low density of the TEP agglomerate generated in low density waters of continental origin, and to the fact that the main component of the advective transport at estuarine regions is the horizontal one. In this study, the significance of TEP in the particulate matter exchange between estuary and coast was analyzed in the estuarine lagoon system of Cananéia-Iguape (southeast coast of Brazil). TEP, total seston (TS), organic seston (OS) and chlorophyll a (Chl.a) were analyzed over complete tidal cycles, during the dry and rainy seasons both at spring and neap tides. Horizontal fluxes and net transport rates of these variables were also calculated. TEP concentrations (max. 4991 μg Xeq/L) were almost one order of magnitude higher during the rainy season. TEP horizontal transport rates as high as 1.8 g Xeq. m/s were observed at the northern inlet of the system. In terms of particulate carbon, it represents 32.7 g TEP-C m/s or 4% of the OS transport rates. Our study quantifies the advective transport of TEP and their importance in particulate matter exchange between a subtropical estuarine system and the adjacent coastal region. Our results contribute to the knowledge of exopolimer particles dynamics in subtropical estuarine systems, and their relationship to phyto-plankton biomass and particulate matter.
