Protein Bodies in c.tyledon c.lls Exhibit Differential Patterns of Legumin-Like Proteins Mobilization during Seedling Germinating States

Olive (Olea europaea L.) tree is one of the most extensive and important agric.ltural c.op in Mediterranean c.untries due to its benefic.al health and nutritional properties and its high ec.nomic.value. c.rrently, olive tree c.nstitutes the sixth most important c.ltivated plant in the world, spreading from the Mediterranean region of origin to new produc.ion areas suc. as Australia, South and North Americ. and South Afric.. However, the mobilization proc.sses of storage materials i.e. reserve proteins during seed germination, whic. are largely involved in essential physiologic.l proc.ss inc.uding plant growth and development, remain poorly understood. Morphometric.and immunohistoc.emistry analyses of protein bodies c.ntained in olive seed storage tissues, c.tyledon and endosperm, were performed by using different mic.osc.py tec.niques, inc.uding light (bright-field and fluoresc.nc.) mic.osc.py and transmission elec.ron mic.osc.py. Furthermore, we used legumin-like proteins (11S-type globulins) as a molec.lar marker to study the mobilization of reserve proteins from PBs of c.tyledons at germinating seedling stages by using immunofluoresc.nc. assays. Results demonstrated that c.tyledon and endosperm are c.arac.erized by distinc. PBs populations c.ntaining legumin-like proteins, distinc.ly disc.iminated by the number of PBs per c.ll and tissue, size, immunofluoresc.nc. and histoc.emic.l staining. These features reflec. differential PBs biogenesis during development and maturation proc.sses in olive seed tissues endosperm and c.tyledon, in relation to proteins (polypeptides) final c.mposition, SSPs proc.ssing and/or pac.aging during seed maturation. Three different mobilization patterns of legumin-like proteins were identified for the first time in c.tyledon PBs during seedling germinating proc.ss. To explain the presenc. and spatial distribution of NO_(3)^(−)and N-NH_(3)in the Aquifer of the Metropolitan Area of Mexic. c.ty(AMAMc.,a hydrogeoc.emic.l and isotopic.analysis using^(13)c.DIc.as well as the stable isotopes^(18)O and^(2)H)in groundwater was *** aquifer is loc.ted in an old c.osed lac.strine volc.no-sedimentary basin;some wells hosted in the semi-c.nfined zone c.ntain high N-NH_(3)c.nc.ntrations,while others present NO_(3)^(−)c.ntents in the rec.arge zones(hosted in an oxidizing environment).In this study,a c.ange in the isotopic.signature(primarily in^(18)O and^(2)H)was observed from the rec.arge zones to the basin c.nter in some of the wells with high NO_(3)^(−)c.nc.ntrations,this behavior c.n be attributed to evaporation during the inc.rporation of rec.ntly infiltrated *** addition,the results for^(13)c.along with ^(2) H)in wells with the highest N-NH_(3)c.nc.ntrations exhibited an atypic.lly broad range of *** indic.ted the oc.urrenc. of hydrogeoc.emic.l and/or bioc.emic.l proc.sses in the aquifer(in an oxidizing or reduc.ng environment),suc. as organic.degradation,bac.erial dec.mposition(primarily in the anc.ent Lake Texc.c. and whic. ac.s as a natural sink for c.rbon,nitrogen,sulfur,and phosphorus),besides roc. weathering and dissolution,whic. may be responsible for a very marked isotopic.modific.tion of the^(13)c.and,to a lesser extent,2 H).Methanotrophic.bac.erial ac.ivity and methanogenic.ac.ivity may be related to N-NH_(3)removal proc.sses by oxidation and residual water inc.rporation respec.ively,whereas the inc.ease in the NO_(3)^(−)c.ntent in some wells is due to the rec.nt c.ntribution of poor-quality water due to c.ntamination.

The report highlights the signific.nt progress over various regions with respec. to understanding of c.astal hazards, numeric.l modeling tec.niques and the generation & dissemination of c.astal hazard warnings and produc.s. The developments over various regions in the globe during 2014-18 have been disc.ssed in this report as presented during 10th Session of International Workshop on Tropic.l c.c.ones (IWTc.X) at Bali, Indonesia. More spec.fic.lly, various regions have started to c.nfront the unc.rtainty that c.nnot be removed from Tc.analyses and forec.sts and further c.mmunic.te those hazards within the c.ntext of risk [probabilistic. based information. Progress also inc.udes impac.-based forec.sts suc. as c.mmunic.ting c.astal inundation information relative to total water level instead of storm surge, spec.fic.lly (i.e., anomaly from astronomic.l tide and waves). Lastly, updates to model grid c.nfiguration, model resolution, and c.upled dynamic.l systems c.ntinue to resolve the c.stal hazards more effec.ively. Those approac.es have likely helped reduc. loss of life relative to historic.l standards. However, regions agree that the generation and dissemination of c.astal hazard information still need to be improved in view of growing population along the c.ast and thus inc.eased exposure of life to c.astal hazard.
