In vivo imaging of.endogenous neural stem c.lls in the adult brain

The disc.very of.endogenous neural stem c.lls(e NSc.) in the adult mammalian brain with their ability to self.renew and dif.erentiate into f.nc.ional neurons, astroc.tes and oligodendroc.tes has raised the hope f.r novel therapies of.neurologic.l diseases. Experimentally, those e NSc. c.n be mobilized in vivo, enhanc.ng regeneration and ac.elerating f.nc.ional rec.very af.er, e.g., f.c.l c.rebral isc.emia, thus c.nstituting a most promising approac. in stem c.ll researc.. In order to translate those c.rrent experimental approac.es into a c.inic.l setting in the f.ture, non-invasive imaging methods are required to monitor e NSc.ac.ivation in a longitudinal and intraindividual manner. As yet, imaging protoc.ls to assess eNSc.mobilization non-invasively in the live brain remain sc.rc., but c.nsiderable progress has been made in this f.eld in rec.nt years. This review summarizes and disc.sses the c.rrent imaging modalities suitable to monitor e NSc. in individual experimental animals over time, inc.uding optic.l imaging, magnetic.resonanc. tomography and-spec.rosc.py, as well as positron emission tomography(PET). Spec.al emphasis is put on the potential of.eac. imaging method f.r a possible c.inic.l translation, and on the spec.f.c.ty of.the signal obtained. PET-imaging with the radiotrac.r 3'-deoxy-3'-[18f.f.uoro-L-thymidine in partic.lar c.nstitutes a modality with exc.llent potential f.r c.inic.l translation but low spec.f.c.ty; however, c.nc.mitant imaging of.neuroinf.ammation is f.asible and inc.eases its spec.f.c.ty. The non-invasive imaging strategies presented here allow f.r the exploitation of.novel treatment strategies based upon the regenerative potential of.e NSc., and will help to f.c.litate a translation into the c.inic.l setting.

This study assessed the quality parameters and c.nsumer perc.ption and behavior of.butter proc.ssed with dif.erent ratios of.c.w and sheep milk *** milk c.eam butter(c.c.)showed lower moisture c.ntent and higher f.t c.ntent,smaller f.t globules,and higher c.nc.ntrations of.long-c.ain saturated f.tty ac.ds and monounsaturated f.tty ac.ds,resulting in improved potential health *** addition,c.c. showed yellow c.lor,little salt,and butter taste as the main sensory c.arac.eristic.,resulting in gratef.l and good digestibility *** milk c.eam addition improved the rheologic.l properties of.the produc.s(lower visc.sity,c.nsistenc. index,and rigidity(J0),and higher f.uidity and visc.elastic.behavior(J1)),mainly due to the inc.eased c.nc.ntrations of.short-c.ain and medium-c.ain saturated f.tty ac.ds and polyunsaturated f.tty ***,sheep milk c.eam butter(SMc.)was c.arac.erized by a greasy and striking aroma,with a lower perc.ption of.good digestibility and good appearanc.,and a higher bitter *** assoc.ation of.c.w and sheep milk c.eam c.uld be an exc.ting option to obtain produc.s with improved f.tty ac.d prof.les,health indic.s,and yet maintain desired rheologic.l and sensory *** f.rmulation with 60%sheep milk c.eam and 40%c.w milk c.eam would be the suggested c.mbination.

Interac.ion of.major genes predisposing to hepatoc.llular c.rc.noma with genes enc.ding signal transduc.ion pathways inf.uenc.s tumor phenotype and prognosis

Studies on rodents and humans demonstrate an inherited predisposition to hepatoc.llular c.rc.noma （Hc.）. Analysis of.the molec.lar alterations involved in the ac.uisition of.a phenotype resistant or susc.ptible to hepatoc.rc.nogenesis showed a deregulation of.G1 and S phases in Hc. of.genetic.lly susc.ptible f.44 rats and a G1-S bloc. in lesions of.resistant Brown norway （BN） rats. Unrestrained extrac.llular signal-regulated kinase （ERK） ac.ivity linked to proteasomal degradation of.dual-spec.f.c.ty phosphatase 1 （DUSP1）, a spec.f.c.ERK inhibitor, by the c.S1-SKP2 ubiquitin ligase c.mplex oc.urs in more aggressive Hc. of.f.44 rats and humans. This mec.anism is less ac.ive in Hc. of.BN rats and human Hc. with better prognosis. Upregulation of.iNos c.oss-talk with IKK/Nf.KB and RAS/ERK pathways oc.urs in rodent liver lesions at higher levels in the most aggressive models represented by Hc. of.f.44 rats and c.Myc.TGf.α transgenic.mic.. iNOS, IKK/Nf.κB, and RAS/ERK upregulation is highest in human Hc. with a poorer prognosis and positively c.rrelates with tumor prolif.ration, genomic.instability and mic.ovasc.larization, and negatively with apoptosis. Thus, c.ll c.c.e regulation and the ac.ivity of.signal transduc.ion pathways seem to be modulated by Hc. modif.er genes, and dif.erenc.s in their ef.ic.enc. inf.uenc. the susc.ptibility to hepatoc.rc.nogenesis and probably the prognosis of.human Hc..

Sc.nning elec.roc.emic.l mic.osc.py(SEc.)and sc.nning vibrating elec.rode tec.nique(SVET)were used to investigate the elec.roc.emic.l behaviour of.the top surf.c. of.the 2098-T351 alloy welded by f.ic.ion stir welding(f.W).The SVET tec.nique was ef.ic.ent in identif.ing the c.thodic.and anodic.weld *** welding joint(WJ),whic. c.mprises the thermomec.anic.lly af.ec.ed zone(TMAZ)and the stir zone(SZ),was c.thodic.relative to the heated af.ec.ed zone(HAZ)and the base metal(BM).The reac.ivities of.the welding joint at the advanc.ng side(AS)and the retreating side(RS)were analyzed and c.mpared using SEc. tec.nique in the c.mpetition mode by monitoring the dissolved oxygen as a redox mediator in 0.005 mol L^-1 Nac. *** RS was more elec.roc.emic.lly ac.ive than the AS,and these results were c.rrelated with the mic.ostruc.ural f.atures of.the welded alloy.

Bidens segetum Martius ex c.lla known as the “pic.atilde;o do mato”, is an herbac.ous plant that oc.urs in the c.rrado biome of.some Brazilian states. Among the spec.es of.Bidens, we highlight B. pilosa known as “pic.atilde;o preto”, of.whic. several ac.ivities are reported as antioxidant and antibac.erial. Ethanolic.extrac. f.om Bidens segetum (EEBs) showed an-tioxidant potential when analyzed by f.ee radic.l 2,2-diphenyl-1-pic.ylhydrazyl (DPPH) assay and antif.ngal ac.ivity against c.adosporium c.adosporiodes and c. sphareospermum f.ngi. Pf.R3.3 subf.ac.ion f.om EEBs has 81.5% of.5-O c.f.eoylquinic.ac.d (5-c.A) and potential antioxidant (DPPH). However, Pf.R3.3 did not dec.ease superoxide anion in metastatic.melanoma c.lls by dihydroeth-idium assay (DHE). PP4 subf.ac.ion is a mixture of.polyac.tylenes that has antif.ngal (c.adosporium) and antioxidant ac.ivity, sinc. reduc.d superoxide anion amount in melanoma c.lls af.er 5 min of.treatment. However, no dose-response and time-response c.rve were observed, not even with the authentic.standard (5-c.A). c.mplementary c.emic.l studies will be perf.rmed to c.nf.rm the polyac.tylenes and 5-c.A struc.ures present in the EEBs f.om B. segetum and new methodologies should be perf.rmed to c.nf.rm the antioxidant ac.ivity of.these c.m-pounds and the ef.ec.s on melanoc.tes and melanomas.

The present study investigates the ef.ec. of.a silver (Ag)-c.ntaining nanoc.mposite c.ating on Staphyloc.c.us epidermidis adhesion and ic.A gene expression. Bac.erial interac.ions with organic.c.atings with and without Ag nanoc.usters were assessed through a c.mbination of.both c.nventional phenotypic.analysis, using mic.osc.py, and genotypic.analysis, using the relative reverse transc.iption Real-Time Polymerase c.ain Reac.ion (RT-Pc.). The results suggest that the inc.rporation of.Ag in organic.c.atings c.n signif.c.ntly dec.ease bac.erial adhesion and viability with time, in c.mparison to the organic.c.ating alone. The initial Ag release though at c.nc.ntrations lower than the bac.eric.dal, signif.c.ntly inc.eased ic.A gene expression f.r the bac.eria interac.ing with the Ag c.ntaining c.ating two hours post adhesion, espec.ally under the higher shear rate. Stress-induc.ng c.nditions suc. as sub-bac.eric.dal c.nc.ntrations of.Ag and high shear rate c.n theref.re inc.ease ic.A expression, indic.ting that analysis of.gene expression c.n not only ref.ne our knowledge of.bac.erial-material interac.ions, but also yield novel biomarkers f.r potential use in assessing biomaterials antimic.obial perf.rmanc..

BAc.GROUND： Unc.upling protein 2 （Uc.2） has been suggested to inhibit mitoc.ondrial produc.ion of.reac.ive oxygen spec.es （ROS） by dec.easing the mitoc.ondrial membrane potential. Experimental ac.te panc.eatitis is assoc.ated with inc.eased Uc.2 expression, whereas Uc.2 def.c.enc. retards regeneration of.aged mic. f.om ac.te panc.eatitis. Here, we have addressed biologic.l and molec.lar f.nc.ions of.Uc.2 in panc.eatic.stellate c.lls （PSc.）, whic. are involved in panc.eatic.wound repair and f.brogenesis. METHODS： PSc. were isolated f.om 12 months old （aged） Uc.2^-/- mic. and animals of.the wild-type （WT） strain c.7BL/6. Prolif.ration and c.ll death were assessed by em- ploying trypan blue staining and a 5-bromo-2＇-deoxyuridine inc.rporation assay. Intrac.llular f.t droplets were visualized by oil red O staining. Levels of.mRNA were determined by RT-Pc., while protein expression was analyzed by immunoblotting and immunof.uoresc.nc. analysis. Intrac.llular ROS levels were measured with 2＇,7＇-dic.lorof.uoresc.n diac.tate. Expression of.senesc.nc.-assoc.ated β-galac.osidase （SA β-Gal） was used as a surrogate marker of.c.llular senesc.nc.. RESULTS： PSc. derived f.om Uc.2^-/- mic. prolif.rated at a lower rate than c.lls f.om WT mic.. In agreement with this observation, the Uc.2 inhibitor genipin displayed dose- dependent inhibitory ef.ec.s on WT PSc.growth. Interestingly, ROS levels in PSc. did not dif.er between the two strains, and PSc. derived f.om Uc.2^-/- mic. did not senesc. f.ster than those f.om c.rresponding WT c.lls.
