Impacts of land use and land cover changes on hydrological processes and sediment yield determined using the SWAT model

Land use and land cover (LULC) changes strongly affect local hydrology and sediment *** current study focused on a basin in the Brazilian Amazon and had the following three objectives:(1) to perform an effective diagnosis of flow and sediment yield,(2) to evaluate the impacts of LULC changes over the last 40 years on the hydro-sedimentological variables,and (3) to investigate the impacts of the possible trends or breaking points in the flow,surface runoff,and sediment yield *** Soil and Water Assessment Tool (SWAT) model validation showed that the simulated results were consistent with the data measured in the dynamic reproduction of flow ***,changes in LULC altered surface runoff,sediment yield,and flow according to the Mann-Kendall and Pettitt non-parametric *** was also observed that the sub-basins in which pastureland is predominant are more susceptible to increased surface runoff and sediment *** to trend and homogeneity tests,these sub-basins had break points in the time series of these *** the other hand,in the sub-basins whose land cover is predominantly forest,the time series is homogeneous and *** current study highlighted the impacts that 40 years of LULC changes in an Amazonian basin had on hydro-sedimentological variables,which,in turn,can play an important role in changing the hydrological cycle,consequently these changes can also impact diverse human activities in the region,such as agribusiness,livestock,energy production,food security,and public water ***,it is concluded that the SWAT model is a good estimator of hydro-sedimentological processes in Amazonian basins and can be used by decision makers in the management of water and environmental resources.

Valproic Acid,a Drug with Multiple Molecular Targets Related to Its Potential Neuroprotective Action

Valproic acid (VA) is used worldwide as an antiepileptic drug and a mood stabilizer. Recently, VA was shown to act on cell growth, differentiation and apoptosis, by regulating gene expression at the molecular level, through epigenetic mechanisms. Thus, VA was demonstrated to act on the chromatin remodeling what is a consequence of the drug inhibition of histone deacetylases (HDACs) activity. Other studies uncovered the potential of VA to interfere with multiple regulatory mechanisms besides HDACs, as the GSK3 alpha and beta, Akt, ERK and phosphoinositol pathways, tricarboxylic acid cycle, GABA and OXPHOS system. The review focuses on the mechanisms of action of VA, showing that HDAC inhibitors, as VA, can be successfully used in the treatment of neurodegenerative disorders. This molecule, whose biological activities range from interactions with receptors and ion channels to the regulation of many catalytic reactions, has a central role in cellular cascades that regulate gene expression. Thus, inhibitors of HDACs, by positively affecting both neuronal degeneration and cognitive deficits, appear as promising drugs against various pathological conditions and neurodegenerative diseases. VA is known to present anti-inflammatory and antioxidative properties. And, since inflammation and oxidative stress are common links in neurodegeneration, VA is a drug that, from a clinical point of view, shows a great potential as a candidate for the treatment of neurodegenerative diseases related to excitotoxic events.

Endothelin-2 Differential Expression in Normal and Early-Stages of Colon Cancer Development

The endothelin family has been related with several pathological diseases including cardiovascular disorders, hypertension and cancer. However, little is known about endothelin system in early stages of colorectal cancer and there are no studies evaluating differences in proximal and distal colon segments. To deepen in this issue, we have studied the endothelin’s family gene and protein expression in normal mice and early stage of a mice model of Azoxymethane (AOM) and Dextran Sodium Sulphate (DSS) induced colorectal cancer in proximal and distal segments. Additionally, using nonlinear microscopy (NLM) techniques, we have characterized collagen changes in early stages of cancer disease development. In the present study, we have found significant differential gene expression and protein localization between these colon regions, which allow us to hypothesize a new role for the ET-2 as an early marker of colon cancer development.

Pompe disease (PD) is a rare inborn error of metabolism due to an abnormal acid alpha-glucosidase (GAA) activity that comprises glycogen breakdown mainly in the lysosomes. Since the introduction of enzyme replacement therapy (ERT), with recombinant human GAA for the early onset PD patient, a relevant field of clinical research due to the benefits regarding survival rate has been widely documented worldwide. Objective: To describe the clinical characteristics and the ERT effects in a series of Brazilian patients with infantile onset PD (IOPD) under ERT. Methods: Brazilian patients diagnosed with IOPD under ERT were recruited through their physicians participating in the International Pompe Disease Registry from 2009 to 2017. Data were collected by an online survey. Results: 10 IOPD patients were identified through the survey with a death rate of 30% and technology dependency rate reported as 80% (motor, respiratory or nutritional fields) of the patients. After the third year of ERT, motor disabilities were lost in 50% of ambulated patients. The overall characteristics were similar to international studies. Conclusion: Despite ERT benefits in cardiac involvement, motor disabilities seem to be much more compromised in IOPD patients, with high technology dependence, especially after three years of age.

Estimation of visceral fat is useful for the diagnosis of significant fibrosis in patients with non-alcoholic fatty liver disease

Nutrient Analysis Critical Control Point (NACCP): Hazelnut as a Prototype of Nutrigenomic Study

The aim of this study is to apply the Nutrient Analysis Critical Control Point (NACCP) process to ensure that the highest nutrient levels in food can determine a beneficial effect on the health of the consumer. The NACCP process involves a sequence of analysis and controls that depart from raw material production to the evaluation of the effect of nutrition on health. It is articulated through the following points: 1) identification of nutrient level in the food;2) identification of critical control points (environmental, genetic data, chemical and physical data, production technology, distribution and administration);3) establishing critical limits that can impoverish and damage the nutrient;4) establishing measures to monitor;5) establishing corrective actions. We selected as biomarkers the total phenolic content (TPC) and total antioxidant capacity (TAC) of a genotyped Italian hazelnut cultivars (Corylus e avellana L.). We performed a clinical study evaluating: a) nutritional status;b) clinical-biochemical parameters;c) low density lipoprotein oxidation (LDL-ox);d) the expression level changes of oxidative stress pathway genes in the blood cell at baseline and after 40 g/die of hazelnut consumption. In this study, we found a significant lowering (p ≤ 0.005) of LDL oxidized proteins, in association with the consumption of 40 g/d of hazelnuts. Also, we found a significant variation (p ≤ 0.005) of gene expression of antioxidant and pro-oxidant genes, between the intake of dietary with and without hazelnuts. This results support the hypothesis that the NACCP process could be applied to obtain significant benefits in terms of primary prevention and for contributing to the amelioration of food management at the consumer level.

Modeling of the Primary Acts of the Interaction between a Cell and an External Mechanical Field

The mechanism of interaction between a cell and an external mechanical field is still poorly understood, and the accumulated diverse experimental data are often scattered. Therefore, the aim of this work was to systematize the experimental data in a mathematical model of the interaction between a cell and an external mechanical field based on standard kinetic equations and Fick’s diffusion equation. Assuming that the cortical cytoskeleton proteins play a key role in cell mechanosensitivity, we compared the results of mathematical modeling and experimental data concerning the content of cytoskeletal proteins at the early stages of a mechanical field change. In addition, the proposed mathematical model suggests the dynamics of changes of a key transcription factor, which is necessary for the expression of certain genes encoding cytoskeletal proteins.

Next-generation resorbable polymer scaffolds with surface-precipitated calcium phosphate coatings

Next-generation synthetic bone graft therapies will most likely be composed of resorbable polymers in combination with bioactive *** this article,we continue our exploration of E1001(1k),a tyrosine-derived polycarbonate,as an orthopedic implant ***,we use E1001(1k),which is degradable,nontoxic,and osteoconductive,to fabricate porous bone regeneration scaffolds that were enhanced by two different types of calcium phosphate(CP)coatings:in one case,pure dicalcium phosphate dihydrate was precipitated on the scaffold surface and throughout its porous structure(E1001(1k)+CP).In the other case,bone matrix minerals(BMM)such as zinc,manganese and fluoride were co-precipitated within the dicalcium phosphate dihydrate coating(E1001(1k)+BMM).These scaffold compositions were compared against each other and against ChronOS(Synthes USA,West Chester,PA,USA),a clinically used bone graft substitute(BGS),which served as the positive control in our experimental *** BGS is composed of poly(lactide co-e-caprolactone)and beta-tricalcium *** used the established rabbit calvaria critical-sized defect model to determine bone regeneration within the defect for each of the three scaffold *** bone formation was determined after 2,4,6,8 and 12 weeks by micro-computerized tomography(mCT)and *** experimental tyrosine-derived polycarbonate,enhanced with dicalcium phosphate dihydrate,E1001(1k)+CP,supported significant bone formation within the defects and was superior to the same scaffold containing a mix of BMM,E1001(1k)+*** comparison with the commercially available BGS was complicated by the large variability in bone formation observed for the laboratory preparations of E1001(1k)*** all time points,there was a trend for E1001(1k)+CP to be superior to the commercial ***,only at the 6-week time point did this trend reach statistical *** analysis of the μCT data suggested an increase in bone fo

Presence of Illicit Drugs in the Sarno River (Campania Region, Italy)

The presence of illicit drugs and their metabolites in surface waters has to be considered a new type of hazard, still unknown, for both the human health and the aquatic ecosystem, due to the potent pharmacological activities of all the illicit drugs. Our research was aimed at evaluating the presence of illicit drugs in the Sarno River (Campania region, Italy), crossing a densely populated area, the basin of the Sarno River, one of the largest and most important economic areas in Campania region, famous for the presence of zones with high landscape-environmental value. The drugs selected for this study were the most used in Campania region. The presence of illicit drugs in surface water was analyzed by a selective multi-residue assay based on liquid chromatographytandem mass spectrometry. The analysis showed the presence of all the illicit drugs investigated: cocaine and its main metabolites (benzoylecgonine, nor-benzoylecgonine), morphine, THC-COOH and codeine;cocaine was the most abundant illicit drug. The presence of illicit drugs and their metabolites in the Sarno River suggests new potential risk for the basin’s inhabitants, using water in the food chain, via field irrigation and animal feed, and for the health of the aquatic fauna.
