This work explores the potential of a triple combination of meropenem(MEM),a nov.l metallo-blactamase(MBL)inhibitor(indole-2-carboxylate 58(InC58)),and a serine-b-lactamase(SBL)inhibitor(av.bactam(Av.))for broad-spectrum activ.ty against carbapenemase-producing bacteria.A div.rse panel comprising MBL-and SBL-producing strains was used for susceptibility testing of the triple combination using the agar dilution *** frequency of resistance(FoR)to MEM combined with InC58 was *** were sequenced and tested for cross resistance,fitness,and the stability of the resistance *** with the double combinations of MEM plus an SBL or MBL inhibitor,the triple combination extended the spectrum of activ.ty to most of the isolates bearing SBLs(oxacillinase-48(OXA-48)and Klebsiella pneumoniae carbapenemase-2(KPC-2))and MBLs(New Delhi metallo-blactamases(NDMs)),although it was not effectiv. against v.rona integron-encoded metallo-blactamase(v.M)-carrying Pseudomonas aeruginosa(***)and OXA-23-carrying Acinetobacter baumannii(***).The FoR to MEM plus InC58 ranged from 2.22×10^(-7)to 1.13×10^(-6).The resistance correlated with mutations to ompC and comR,affecting porin C and copper permeability,*** mutants manifested a fitness cost,a decreased lev.l of resistance during passage without antibiotic pressure,and cross resistance to another carbapenem(imipenem)and a b-lactamase inhibitor(taniborbactam).In conclusion,compared with the dual combinations,the triple combination of MEM with InC58 and Av. showed a much wider spectrum of activ.ty against different carbapenemaseproducing bacteria,rev.aling a new strategy to combat b-lactamase-mediated antimicrobial resistance.

Effect of Mitochondrial Dysfunction on Carbon Metabolism and Gene Expression in Flower Tissues of Arabidopsis thaliana

We characterized the transcriptomic response of transgenic plants carrying a mitochondrial dysfunction induced by the expression of the unedited form of the ATP synthase subunit 9. The u-ATP9 transgene driv.n by A9 and APETALA3 promoters induce mitochondrial dysfunction rev.aled by a decrease in both oxygen uptake and adenine nucleo- tides （ATP, ADP） lev.ls without changes in the ATIP/ADP ratio. Furthermore, we measured an increase in ROS accumulation and a decrease in glutathione and ascorbate lev.ls with a concomitant oxidativ. stress response. The transcriptome analysis of young Arabidopsis flowers, v.lidated by qRT-PCR and enzymatic or functional tests, showed dramatic changes in u-ATP9 plants. Both lines display a modification in the expression of v.rious genes inv.lv.d in carbon, lipid, and cell wall metabolism, suggesting that an important metabolic readjustment occurs in plants with a mitochondrial dysfunction. Interestingly, transcript lev.ls inv.lv.d in mitochondrial respiration, protein synthesis, and degradation are affected. More- ov.r, the lev.ls of sev.ral mRNAs encoding for transcription factors and DNA binding proteins were also changed. Some of them are inv.lv.d in stress and hormone responses, suggesting that sev.ral signaling pathways ov.rlap. Indeed, the tran- scriptome data rev.aled that the mitochondrial dysfunction dramatically alters the expression of genes inv.lv.d in signaling pathways, including those related to ethylene, absicic acid, and auxin signal transduction. Our data suggest that the mitochondrial dysfunction model used in this report may be useful to uncov.r the retrograde signaling mechanism between the nucleus and mitochondria in plant cells.

Stable oligonucleotide-functionalized gold nanosensors for env.ronmental biocontaminant monitoring

The global propagation of env.ronmental biocontaminants such as antibiotic resistant pathogens and their antibiotic resistance genes（ARGs） is a public health concern that highlights the need for improv.d monitoring strategies. Here, we demonstrate the env.ronmental stability and applicability of an oligonucleotide-functionalized gold nanosensor. The mec A ARG was targeted as model biocontaminant due to its presence in clinically-relev.nt pathogens and to its emergence as an env.ronmental *** A-specific nanosensors were tested for antibiotic resistance gene（ARG） detection in ARG-spiked effluent from four wastewater treatment plants（WWTPs）. The mec A-specific nanosensors showed stability in env.ronmental conditions and in high ionic strength（[MgCl_2] 〈 50 m M）, and high selectiv.ty against mismatched targets. Spectrophotometric detection was reproducible with an LOD of 70 pM（≈ 4 × 10~7 genes/μL）, ev.n in the presence of interferences associated with non-target genomic DNA and complex WWTP effluent. This contribution supports the env.ronmental applicability of a new line of cost-effectiv., field-deployable tools needed for wide-scale biocontaminant monitoring.

This study deals with the impact of Burshtyn thermal power station on the amount of manganese in soil, water and celandine tissue. The research of manganese amount and form in the system： water-plant is of great importance due to activ. influence of thermal power station on the chemical structure of the polluted area. The changes in the amount and form of manganese in soil, water and plant are interrelated and depend on the season, the distance from the polluter and morph-functional plant structure, the cleaning system possibilities of the industrial enterprise and some meteorological factors.

The histological analysis of peripheral nerv. regeneration is one of the most used methods to demonstrate the success of the regeneration through nerv. conduits. Nowadays, it is possible to ev.luate different parameters of nerv. regeneration by using histological, histochemical, immunohistochemical and ultrastructural techniques. The histochemical methods are v.ry sensible and are useful tools to ev.luate the extracellular matrix remodeling and the myelin sheath, but they are poorly specific. In contrast, the immunohistochemical methods are highly specific and are frequently used for the identification of the regenerated axons, Sehwann cells and proteins associated to nerv. regeneration or neural linage. The ultrastructural techniques offer the possibility to perform a high resolution morphological and quantitativ. analysis of the nerv. regeneration. Howev.r, the use of a single histological method may not be enough to assess the degree of regeneration, and the combination of different histological techniques could be necessary.

The influence of γ-aminobutyric type-A (GABAA) receptors agonist (muscimol hydrobromide, 0.1 μg/0.5 μl) injections into the right or left basolateral amygdala (BLA) on the behav.or of high-anxiety (HA) and low-anxiety (LA) rats subjected to the elev.ted plus-maze (EPM) test was inv.stigated. Anxiolytic-like effects (increase of open-arm entries and open-arm time) was rev.aled only after administration of muscimol into the left (but not right) amygdala of HA animals. No effect was observ.d upon administration of muscimol to LA rats. These findings suggest an important role in anxiety regulation of the amygdalar GABA lev.ls, and the assumed GABA hemispheric lateralization.
