Background:Patients with human epidermal growth factor receptor 2(HeR2)-positive early breast cancer with residual invasive disease after neoadjuvant systemic therapy have a high risk of recurrence and *** primary analysis of KATHeRINe,a phase 3,open-label trial,showed that the risk of invasive breast cancer or death was 50%lower with adjuvant trastuzumab emtansine(T-DM1)than with trastuzumab alone.

Pancreatic cancer diagnosis by free and exosomal miRNA

Patients with pancreatic adenocarcinoma(PaCa) have a dismal prognosis. This is in part due to late diagnosis prohibiting surgical intervention, which provides the only curative option as PaCa are mostly chemo- and radiation resistance. Hope is raised on a reliable noninvasive/minimally invasive diagnosis that is still missing. Recently two diagnostic options are discussed, serum MicroRNA(miRNA) and serum exosomes. Serum miRNA can be free or vesicle-, particularly, exosomesenclosed. This review will provide an overview on the current state of the diagnostic trials on free serum miRNA and proceed with an introduction of exosomes that use as a diagnostic tool in serum and other body fluids has not received sufficient attention, although serum exosome miRNA in combination with protein marker expression likely will increase the diagnostic and prognostic power. By their crosstalk with host cells, which includes binding-initiated signal transduction, as well as reprogramming target cells via the transfer of proteins, mRNA and miRNA exosomes are suggested to become a most powerful therapeutics. I will discuss which hurdles have still to be taken as well as the different modalities, which can be envisaged to make therapeutic use of exosomes. PaCa are known to most intensely crosstalk with the host as apparent by desmoplasia and frequent paraneoplastic syndromes. Thus, there is hope that the therapeutic application of exosomes brings about a major breakthrough.

Plastic pollution has become a global environmental issue,posing notable threats to ecosystems and human health. Plastic aging is a continuous dynamic process,during which plastics of various sizes are produced and exist,such as large plastics,microplastics,and nanoplastics. Regardless of the size,plastics continuously fragment and release plastic additives and other soluble products. Moreover,due to the same molecular composition,the aging mechanisms and characteristics of micro and nanoplastics are similar to those of macroplastics. However,the smaller the particle size,the greater the potential harm. To enhance understanding of the environmental effects of plastics,this review focused on the entire dynamic process from the initial form of plastics to their degradation and summarized the aging mechanisms involved,changes in properties,and the products released during this process. These products include macroplastics,micro and nanoplastics,plastic additives,and other soluble substances. The paper also summarized relevant studies and main conclusions on plastic aging experiments,further exploring factors that influence the leaching of plastic additives and the release of micro and nanoplastic particles. Finally,it provided a summary of the environmental hazards that were caused by plastic aging products. © 2025 Science Press. All rights reserved.

Precipitation comes in various phases, including rainfall, snowfall, sleet, and hail. Shifts of precipitation phases, as well as changes in precipitation amount, intensity, and frequency, have significant impacts on regional climate, hydrology,ecology, and the energy balance of the land-atmosphere system. Over the past century, certain progress has been achieved in aspects such as the observation, discrimination, transformation, and impact of precipitation phases. Mainly including: since the1980s, studies on the observation, formation mechanism, and prediction of precipitation phases have gradually received greater attention and reached a certain scale. The estimation of different precipitation phases using new detection theories and methods has become a research focus. Avariety of discrimination methods or schemes, such as the potential thickness threshold method of the air layer, the temperature threshold method of the characteristic layer, and the near-surface air temperature threshold method,have emerged one after another. Meanwhile, comparative studies on the discrimination accuracy and applicability assessment of multiple methods or schemes have also been carried out simultaneously. In recent years, the shift of precipitation from solid to liquid(SPSL) in the mid-to-high latitudes of the Northern Hemisphere has become more pronounced due to global warming and human activities. It leads to an increase in rain-on-snow(ROS) events and avalanche disasters, affecting the speed, intensity, and duration of spring snow-melting, accelerating sea ice and glacier melting, releasing carbon from permafrost, altering soil moisture, productivity, and phenological characteristics of ecosystems, and thereby affecting their structures, processes, qualities, and service functions. CNB-001 reduces paraplegia in rabbits following spinal cord ischemia

Spinal cord ischemia associated with trauma and surgical procedures including thoraco-abdominal aortic aneurysm repair and thoracic endovascular aortic repair results in devastating clinical deficits in patients. Because spinal cord ischemia is inadequately treated, we studied the effects of [4-((1 e)-2-(5-(4-hydroxy-3-methoxystyryl-)-1-phenyl-1 H-pyrazoyl-3-yl) vinyl)-2-methoxy-phenol)](CNB-001), a novel curcumin-based compound, in a rabbit SCI model. CNB-001 is known to inhibit human 5-lipoxygenase and 15-lipoxygenase and reduce the ischemia-induced inflammatory response. Moreover, CNB-001 can reduce the level of oxidative stress markers and potentiate brain-derived neurotrophic factor and brain-derived neurotrophic factor receptor signaling. The Tarlov scale and quantal analysis technique results revealed that CNB-001 administered as an intravenous dose(bolus) 30 minutes prior to spinal cord ischemia improved the behaviors of female New Zealand White rabbits. The improvements were similar to those produced by the uncompetitive N-methyl-D-aspartate receptor antagonist memantine. At 48 hours after aortic occlusion, there was a 42.7% increase(P erated ischemia duration(n = 14) for rabbits treated with CNB-001(n = 16), and a 72.3% increase for rabbits treated with the positive control memantine(P ed to vehicle-treated ischemic rabbits(n = 22). CNB-001 is a potential important novel treatment for spinal cord ischemia induced by aortic occlusion. All experiments were approved by the CSMC Institutional Animal Care and Use Committee(IACUC #4311) on November 1,2012.
