Association of Congenital Heart Defects (CHD) with Factors Related to Maternal Health and Pregnancy in newborns in Puerto Rico

Background:Given the pervasive issues of obesity and diabetes both in Puerto Rico and the broader United States,there is a compelling need to investigate the intricate interplay among body mass index(BMI),pregesta-tional,and gestational maternal diabetes,and their potential impact on the occurrence of congenital heart defects(CHD)during neonatal development.Methods:Using the comprehensive System of Vigilance and Surveillance of Congenital Defects in Puerto Rico,we conducted a focused analysis on neonates diagnosed with CHD between 2016 and 2020.Our assessment encompassed a range of variables,including maternal age,gestational age,BMI,pregestational diabetes,gestational diabetes,hypertension,history of abortion,and presence of preeclampsia.Results:A cohort of 673 patients was included in our study.The average maternal age was 26 years,within a range of 22 to 32 years.The mean gestational age measured 39 weeks,with a median span of 38 to 39 weeks.Of the 673 patients,274(41%)mothers gave birth to neonates diagnosed with CHD.Within this group,22 cases were linked to pre-gestational diabetes,while 202 were not;20 instances were associated with gestational diabetes,compared to 200 without;and 148 cases exhibited an overweight or obese BMI,whereas 126 displayed a normal BMI.Conclusion:We identiﬁed a statistically signiﬁcant correlation between pre-gestational diabetes mellitus and the occurrence of CHD.However,our analysis did not show a statistically signiﬁcant association between maternal BMI and the likelihood of CHD.These results may aid in developing effective strategies to prevent and manage CHD in neonates.

neurological disorders are a diverse group of conditions that affect the nervous system and include neurodegenerative diseases(Alzheimer’s disease,multiple sclerosis,Parkinson’s disease,Huntington’s disease),cerebrovascular conditions(stroke),and neurodevelopmental disorders(autism spectrum disorder).Although they affect millions of individuals around the world,only a limited number of effective treatment options are available today.Since most neurological disorders express mitochondria-related metabolic perturbations,metformin,a biguanide type II antidiabetic drug,has attracted a lot of attention to be repurposed to treat neurological disorders by correcting their perturbed energy metabolism.However,controversial research emerges regarding the beneficial/detrimental effects of metformin on these neurological disorders.Given that most neurological disorders have complex etiology in their pathophysiology and are influenced by various risk factors such as aging,lifestyle,genetics,and environment,it is important to identify perturbed molecular functions that can be targeted by metformin in these neurological disorders.These molecules can then be used as biomarkers to stratify subpopulations of patients who show distinct molecular/pathological properties and can respond to metformin treatment,ultimately developing targeted therapy.In this review,we will discuss mitochondria-related metabolic perturbations and impaired molecular pathways in these neurological disorders and how these can be used as biomarkers to guide metformin-responsive treatment for the targeted therapy to treat neurological disorders.

Comparison of extraction abilities of deep eutectic solvents and aqueous acid solutions for extraction of rare earths and transition metals

In recent years,deep eutectic solvents have attracted increasing interest as effective extractants for the separation of both organic substances and metals from various objects.Acid-based deep eutectic solvents are most often used as extractants for the extraction of metals.In this work,for the first time,the extraction efficiency of transition metals and rare earth elements(Y,Zr,nb,La,Ce,Pr,nd,Sm,Eu,Gd,Tb,Dy,Ho,Er,Tm,Yb,Lu,Hf and Ta)from mining tailings and na,K,Ca,Mg,Cu,Fe,Mn,Pb,Ba,ni and Sr from biodiesel samples using acidic deep eutectic solvents and saturated aqueous solutions of the same acids(malonic,malic,tartaric and citric)is compared.For this,deep eutectic solvents based on acids and their aqueous saturated solutions were prepared and studied.The prepared mixtures were analyzed by IR spectroscopy to confirm the formation of eutectic solvents.Properties such as the density and viscosity of the resulting mixtures were also investigated,as this can be of key importance for the efficiency of metal extraction.The extraction of rare earth metals from mining tailings took a long time(up to several days),while the extraction of metals from fuel took no more than 30 min.Atomic emission spectral methods were used as an analysis method.It is shown that the extraction efficiency with aqueous solutions is better than that with eutectic solvents,which casts doubt on the need for deep eutectic solvents use in this area.

HIFs in hypoxic regulation of the extracellular matrix:focus on little-known player HIF-3

The structural and associated molecules of the extracellular matrix(ECM)complex is an important component of the local milieu of cells,both for maintaining their functions and homeostasis.It is a dynamic structure that is finely tuned to changes in the microenvironment.One of these factors is hypoxia,which can arise in tissues due to physiological or pathological effects.As a result of the hypoxic effect,the properties of the ECM are significantly modified,stiffness increases,the balance between degradation and synthesis of structural proteins shifts,and the deposition of biologically active mediators’changes.Hypoxia-inducible factors(HIFs)contribute significantly to the modification of the properties of the ECM under hypoxia.Among the three HIF isoforms,HIF-1 is the most studied,with numerous identified target genes encoding proteins that participate in all matrisome compartments.Much less is known about HIF-2 and HIF-3.In the context of the effects of hypoxia on the matrisome,HIF-3 isoform is of particular interest.Unlike the first two isoforms,transiently expressed during the first hours of hypoxia,HIF-3 is activated after around 24 h of exposure to hypoxia and persists for a longer period.In addition,its transcription is more pronounced under hypoxia than are HIFs 1 and 2.HIF-3,rather than the first two isoforms,is possibly responsible for the changes that occur in matrisome during prolonged hypoxic conditions.This review attempts to summarize the available data on the involvement of HIF-3 in the matrisome modification under hypoxia.

Applying a Context-based Method to Build a Knowledge Graph for the Blue Amazon

Knowledge graphs are employed in several tasks,such as question answering and recommendation systems,due to their ability to represent relationships between concepts.Automatically constructing such a graphs,however,remains an unresolved challenge within knowledge representation.To tackle this challenge,we propose CtxKG,a method specifically aimed at extracting knowledge graphs in a context of limited resources in which the only input is a set of unstructured text documents.CtxKG is based on OpenIE(a relationship triple extraction method)and BERT(a language model)and contains four stages:the extraction of relationship triples directly from text;the identification of synonyms across triples;the merging of similar entities;and the building of bridges between knowledge graphs of different documents.Our method distinguishes itself from those in the current literature(i)through its use of the parse tree to avoid the overlapping entities produced by base implementations of OpenIE;and(ii)through its bridges,which create a connected network of graphs,overcoming a limitation similar methods have of one isolated graph per document.We compare our method to two others by generating graphs for movie articles from Wikipedia and contrasting them with benchmark graphs built from the OMDb movie database.Our results suggest that our method is able to improve multiple aspects of knowledge graph construction.They also highlight the critical role that triple identification and named-entity recognition have in improving the quality of automatically generated graphs,suggesting future paths for investigation.Finally,we apply CtxKG to build BlabKG,a knowledge graph for the Blue Amazon,and discuss possible improvements.

Clinical implications of single cell sequencing for bladder cancer

Bladder cancer(BC)is the 10th most common cancer worldwide,with about 0.5 million reported new cases and about 0.2 million deaths per year.In this scoping review,we summarize the current evidence regarding the clinical implications of single-cell sequencing for bladder cancer based on PRISMA guidelines.We searched PubMed,CEnTRAL,Embase,and supplemented with manual searches through the Scopus,and Web of Science for published studies until February 2023.We included original studies that used at least one single-cell technology to study bladder cancer.Forty-one publications were included in the review.Twenty-nine studies showed that this technology can identify cell subtypes in the tumor microenvironment that may predict prognosis or response to immune checkpoint inhibition therapy.Two studies were able to diagnose BC by identifying neoplastic cells through single-cell sequencing urine samples.The remaining studies were mainly a preclinical exploration of tumor microenvironment at single cell level.Single-cell sequencing technology can discriminate heterogeneity in bladder tumor cells and determine the key molecular properties that can lead to the discovery of novel perspectives on cancer management.This nascent tool can advance the early diagnosis,prognosis judgment,and targeted therapy of bladder cancer.

DnA Tandem Repeats as Iterable Objects to Count Cell Divisions: A Computational Model

Cell lineages of nematodes are completely known: the adult male of Caenorhabditis elegans contains 1031 somatic cells, the hermaphrodite 959, not one more, not one less;cell divisions are strictly deterministic (as in the great majority of invertebrates) but so far nothing is known about the mechanism used by cells to count precise numbers of divisions. In vertebrates, each species has its invariable deterministic numbers of somites, vertebrae, fingers, and teeth: counting the number of iterations is a widespread process in living beings;nonetheless, it remains an unanswered question and a great challenge in cell biology. This paper introduces a computational model to investigate the possible role of satellite DnA in counting cell divisions, showing how cells may operate under Boolean logic algebra. Satellite DnA, made up of repeated monomers and subject to high epigenetic methylation rates, is very similar to iterable sequences used in programming: just like in the “iteration protocol” of algorithms, the epigenetic machinery may run over linear tandem repeats (that hold cell-fate data), read and orderly mark one monomer per cell-cycle (cytosine methylation), keep track and transmit marks to descendant cells, sending information to cell-cycle regulators.

5-Bromo-2'-deoxyuridine labeling:historical perspectives,factors infiuencing the detection,toxicity,and its implications in the neurogenesis

The halopyrimidine 5-bromo-2′-deoxyuridine(BrdU)is an exogenous marker of DnA synthesis.Since the introduction of monoclonal antibodies against BrdU,an increasing number of methodologies have been used for the immunodetection of this synthesized bromine-tagged base analogue into replicating DnA.BrdU labeling is widely used for identifying neuron precursors and following their fate during the embryonic,perinatal,and adult neurogenesis in a variety of vertebrate species including birds,reptiles,and mammals.Due to BrdU toxicity,its incorporation into replicating DnA presents adverse consequences on the generation,survival,and settled patterns of cells.This may lead to false results and misinterpretation in the identification of proliferative neuroblasts.In this review,I will indicate the detrimental effects of this nucleoside during the development of the central nervous system,as well as the reliability of BrdU labeling to detect proliferating neuroblasts.Moreover,it will show factors influencing BrdU immunodetection and the contribution of this nucleoside to the study of prenatal,perinatal,and adult neurogenesis.Human adult neurogenesis will also be discussed.It is my hope that this review serves as a reference for those researchers who focused on detecting cells that are in the synthetic phase of the cell cycle.

Chebyshev polynomial-based Ritz method for thermal buckling and free vibration behaviors of metal foam beams

This study presents the Chebyshev polynomials-based Ritz method to examine the thermal buckling and free vibration characteristics of metal foam beams.The analyses include three models for porosity distribution and two scenarios for thermal distribution.The material properties are assessed under two conditions,i.e.,temperature dependence and temperature independence.The theoretical framework for the beams is based on the higher-order shear deformation theory,which incorporates shear deformations with higher-order polynomials.The governing equations are established from the Lagrange equations,and the beam displacement fields are approximated by the Chebyshev polynomials.numerical simulations are performed to evaluate the effects of thermal load,slenderness,boundary condition(BC),and porosity distribution on the buckling and vibration behaviors of metal foam beams.The findings highlight the significant influence of temperature-dependent(TD)material properties on metal foam beams'buckling and vibration responses.
