Colorectal cancer(CRC)is the second most common cancer in Europe and its incidence is steadily *** trend could be reversed through timely secondary prevention(screening).In the last twenty years,CRC screening programs across Europe have experienced considerable improvements(fecal occult blood testing;transition from opportunistic to population based program settings).The Czech Republic is a typical example of a country with a long history of nationwide CRC screening programs in the face of very high CRC incidence and mortality *** year,approximately 8000 people are diagnosed with CRC and some 4000 die from this *** years ago,the first pilot studies on CRC screening led to the introduction of the opportunistic Czech National Colorectal Cancer Screening Program in ***,this program was based on the guaiac fecal occult blood test(FOBT)offered by general practitioners,followed by colonoscopy in cases of FOBT *** program has continuously evolved,namely with the implementation of immunochemical FOBTs and screening colonoscopy,as well as the involvement of *** the establishment of the Czech CRC Screening Registry in 2006,2405850 FOBTs have been performed and 104565 preventive colonoscopies recorded within the screening *** overall program expanded to cover 25.0%of the target population by ***,stagnation in the annual number of performed FOBTs lately has led to switching to the option of a population-based program with personal invitation,which is currently being prepared.

In spite of continuous research efforts directed at early detection and treatment of pancreatic cancer, the outlook for patients affected by the disease remains dismal. With most cases still being diagnosed at advanced stages, no improvement in survival prognosis is achieved with current diagnostic imaging approaches. In the absence of a dominant precancerous condition, several risk factors have been identified including family history, chronic pancreatitis, smoking, diabetes mellitus, as well as certain genetic disorders such as hereditary pancreatitis, cystic fibrosis, familial atypical multiple mole melanoma, and Peutz-Jeghers and Lynch syn- dromes. Most pancreatic carcinomas, however, remain sporadic. Current progress in experimental molecular techniques has enabled detailed understanding of the molecular processes of pancreatic cancer development. According to the latest information, malignant pancre- atic transformation involves multiple oncogenes and tumor-suppressor genes that are involved in a variety of signaling pathways. The most characteristic aberrations （somatic point mutations and allelic losses） affect onco- genes and tumor-suppressor genes within RAS, AK-I- and Wnt signaling, and have a key role in transcription and proliferation, as well as systems that regulate the cell cycle （SMAD/DPC, CDKN2A/p16） and apoptosis （TP53）. Understanding of the underlying molecular mechanisms should promote development of new methodology for early diagnosis and facilitate improvement in current approaches for pancreatic cancer treatment.

Longitudinal molecular characterization of endoscopic specimens from colorectal lesions

AIM: To compare molecular profiles of proximal colon, distal colon and rectum in large adenomas, early and late carcinomas. To assess feasibility of testing directed at molecular markers from this study in routine clinical practice. METHODS: A prospective 3-year study has resulted in the acquisition of samples from 159 large adenomas and 138 carcinomas along with associated clinical parameters including localization, grade and histological type for adenomas and localization and stage for carcinomas. A complex molecular phenotyping has been performed using multiplex ligation-dependent probe amplification technique for the evaluation of Cp G-islandmethylator phenotype(CIMP), PCR fragment analysis for detection of microsatellite instability and denaturing capillary electrophoresis for sensitive detection of somatic mutations in KRAS, BRAF, TP53 and APC ***: Molecular types according to previously introduced Jass classification have been evaluated for large adenomas and early and late carcinomas. An increase in CIMP+ type, eventually accompanied with KRAS mutations, was notable between large adenomas and early carcinomas. As expected, the longitudinal observations revealed a correlation of the CIMP+/BRAF + type with proximal location. CONCLUSION: Prospective molecular classification of tissue specimens is feasible in routine endoscopy practice. Increased frequency of some molecular types corresponds to the developmental stages of colorectal tumors. As expected, a clear distinction is notable for tumors located in proximal colon supposedly arising from the serrated(methylation) pathway.

Non-linear Constitutive Model for the Oligocarbonate Polyurethane Material

The polyurethane, which was the subject of the constitutive research presented in the paper, was based on oligocarbonate diols Desmophen C2100 produced by Bayer？. The constitutive modelling was performed with a view to applying the material as the inlay of intervertebral disc prostheses. The polyurethane was assumed to be non-linearly viscohyperelastic, isotropic and incompressible. The constitutive equation was derived from the postulated strain energy function. The elastic and rheological constants were identified on the basis of experimental tests, i.e. relaxation tests and monotonic uniaxial tests at two different strain rates, i.e. λ= 0.1 min-1 and λ= 1.0 min-1. The stiffness tensor was derived and introduced to Abaqus？finite element（FE） software in order to numerically validate the constitutive model. The results of the constants identification and numerical implementation show that the derived constitutive equation is fully adequate to model stress-strain behavior of the polyurethane material.

Technological Properties of Yeast Produced Surface Active Compounds in Connection with Their Application in Clean-Up Technologies

Biologically produced surfactants （SACs） can mobilize and solubilize non-aqueous phase liquids （NAPL） adsorbed onto soil constituents. The interest in microbial surfactants has increased during recent years due to their lower toxicity, higher biodegradability, selectivity and specific activity under extreme conditions than synthetic SACs. Main output of the project represents preparation of this yeast biosurfactant intended for washing of matrices contaminated by NAPL. The influence of cultivation media composition on biosurfactant production was studied and basic properties （critical micelle concentration （CMC）, minimum surface tension） of isolated biosurfactants were compared with properties of synthetic surfactant with surface tension measurement. The interracial tension of the systems containing aqueous solutions of different concentrations and non-polar substances was measured with petroleum compounds （kerosene Jet A-l）, aromatic and aliphatic hydrocarbons （represented by toluene and hexane）. The solution of biosurfactant Yarrowia lipolytica （YAR） in the concentration range of 0-500 mg/L reduced interracial tension by 80% in all representative systems with model contaminants; biosurfactant Candida bombicola （CAN） was less efficient. Solubilization properties were proved with toluene and hexachlorocyclohexane （HCH） isomers alpha and gamma, and effective concentration of biosurfactants was determined as 100 mg/L for toluene and HCH. SACs produced by lipophilic yeast with non-toxic and non-pathogenic status （Yarrowia lipolytica, Candida sp., etc.） seem to be very promising. The results obtained will be used for the application of biosurfactants in the clean-up technologies as agents for the mobilization of non-polar contaminants as well as for stimulation of bioremediation processes.

Statistical variation of characteristics of random packed beds of Raschig rings:The influence of the sample size

Shape Optimization of the Turbomachine Channel by a Gradient Method - Accuracy Improvement

An algorithm of the gradient method of the channel shape optimization has been built on the basis of 3D equations of mass, momentum and energy conservation in the fluid flow. The gradient of the functional that is posed for minimization has been calculated by two methods, via sensitivities and - for comparison - by the finite difference approximation. The equations for sensitivities have been generated through a differentiate-then-discretize approach. The exemplary optimization of the blade shape of the centrifugal compressor wheel has been carried out for the inviscid gas flow governed by Euler equations with a non-uniform mass flow distribution as the inlet boundary condition. Mixing losses have been minimized downstream the outlet of the centrifugal wheel in this exemplary optimization. The results of the optimization problem accomplished by the two above-mentioned methods have been presented. In the case sparse grids have been used, the method with the gradient approximated by finite differences has been found to be more consistent. The discretization accuracy has turned out to be crucial for the consistency of the gradient method via sensitivities.

Temporal inequalities for sequential multi-time actions in quantum information processing

A new kind of temporal inequalities are discussed, which apply to algorithmic processes, involving a finite memory processing unit. They are an alternative to the Leggett-Grag ones, as well as to the modified ones by Brukner et al. If one considers comparison of quantum anti classical processes involving systems of finite memory （of the same capacity in both cases）, the inequalities give a clear message why we can expect quantum speed-up. In a classical process one always has clearly defined values of possible measurements, or in terms of the information processing language, if we have a sequential computations of some function depending on data arriving at each step on an algorithm, the function always has a clearly defined value. In the quantum case only the final value, after the end of the algorithm, is defined. All intermediate values, in agreement with Bohr＇s complementarity, cannot be ascribed a definite value.
